Abstract: Abstract of thesis entitled Complexity Reduction of Mechanical Assemblies for Layered Manufacturing Submitted by CHOW HIN YAN PETER for the degree of Master of Philosophy at The University of Hong Kong in October 2006 There have been rapid strides in the development of layered manufacturing (LM) in the field of computer aided design/computer aided manufacturing (CAD/CAM) last two decades. LM is becoming an increasingly important process, which can be applied in the creation of prototypes only to the fabrication of functional parts in the final product. Due to the differences between traditional manufacturing methods and LM, traditional designs may not be optimal if they are sent for fabrication by LM. Such designs need to be evaluated and modified to ensure that the best results are obtained when the products are fabricated through LM. This study aims to formulate an automated algorithm to redesign the parts or assemblies for LM. Various aspects in which parts and assemblies could be usefully redesigned are considered, and several suitable redesign approaches are suggested. A redesign algorithm is proposed for the redesign of the parts. Based on the construction of the attributed adjacency graph (AAG), parts are investigated thoroughly and redundancies identified. Joining faces and functional faces are used to locate redundant protrusions in various cases. Appropriate approaches are proposed to eliminate these protrusions so as to reduce the geometric complexity of the parts and produce simpler structures. The advantages and disadvantages of the proposed methodology are discussed. This body of work was inspired by a specific challenge involving ordered deposition of single DNAs on surfaces for massively parallel single molecule DNA sequencing via fluorescence microscopy. A potential 10-fold gain in data density is possible if single molecules can be forced into a regular array rather than randomly deposited. The dimensions of such an array are difficult to achieve with conventional lithography techniques. On one end, molecules must be separated by sufficient distance so their optical signatures do not overlap. This distance is on the order of hundreds of nanometers. On the other end, the attachment points for the molecules must have molecular dimensions. Bridging these two length scales is a formidable task. The ability to place nanometer scale objects with nanometer precision can been achieved through atomic force microscopy, scanning tunneling microscopy, optical tweezers, and ebeam lithography. All of these techniques, however, are serial in nature and hence do not serve the intended gain in data density. Another approach toward directed patterning of single molecules is through self-assembly. In this work, self-assembly of block copolymers is explored as a means to addressing the molecular and optical resolution length scales simultaneously. First, the challenge of molecular patterning for single molecule fluorescence microscopy is explored theoretically and the limits of this approach are defined. Block copolymers are introduced as a possible solution to generating the correct surface patterns for improved data density, and experimental results are compared to theoretical predictions. Second, the surface chemistry of these arrays is characterized, and I will show they can be selectively functionalized in preparation for directed assembly of DNAs. Third, these arrays are integrated into single molecule fluorescence imaging experiments to determine their potential for improved data density. What emerges from this work is not only a viable platform for increased single molecule fluorescence data density, but also a deeper understanding of the requirements for directed self-assembly of single molecules.

A variety of [2]catenanes have been assembled through different templating strategies but high order [n]catenanes (n>=4) with multiple interlocked rings are very rare. For the assembly of these high order catenanes, special reaction conditions or templates are required while the yields are modest. In order to access to high order catenanes, a strategy that combines ligand preorganization by ii + Cu -coordination and the use of cucurbit[6]uril catalyzed azide-alkyne cycloaddition (CBAAC) as the ring-closing reaction was adopted. It is believed that the stability of Cu -complexed precursor, in conjunction with the highly efficient ring-closing reaction, will result in the facile assembly of desired high order [n]catenanes. In this thesis, [3]-, [4]-, [5]- and [6]catenanes have been assembled in good yields (>80%) via this strategy and characterized. Chapter 1 gives a brief introduction of catenane. Previously reported assembly strategies of high order [n]catenanes (n>=4), commonly used templates and ring-closing reactions for catenane synthesis are discussed. Chapter 2 describes the design, synthesis, and characterization of the building blocks 1-5, and their self-assembly to [3]-, [4]-, [5]- and [6]catenanes. Careful control of the assembly condition (e.g. concentration), together with preorganization before ring-closing, promoted the assembly of [3]-, [4]-, [5]- and [6]catenanes in good yields (>80%). The assembled catenanes are characterized by NMR and MS experiments. The interlocked nature of assembled structures is demonstrated by NMR studies and further supported by a series of MS experiments. Chapter 3 discusses the synthesis of building block 6 and 7, and their assembly to a [2]catenane using copper-catalyzed alkyne azide cycloaddition (CuAAC). The assembled catenane was preliminarily characterized by MS experiments.
