Book excerpt: Allergic asthma is a serious socioeconomic and health issue that is characterized by chronic inflammation of the lungs and affects as many 300 million people worldwide of all ages. Inhaled environmental factors, such as allergens, can disrupt the airway epithelium, which is the physical barrier and the first line of defense against external irritants. Consequently, exposure to allergens can cause episodes of airway inflammation, increased mucus production and tissue remodeling, that leads to the obstruction and narrowing of the airways, which may present as difficulty breathing, wheezing, and persistent coughing. Unfortunately, there is no cure for allergic asthma, but allergen avoidance and prescription of medication can help manage symptoms. However, current treatment options fail in almost 10% of patients, making it necessary to study uncharacterized molecular causes to better understand this condition. Evidence suggests that mitochondria play a vital role in lung health and disease. Mitochondria are dynamic organelles that can change in size, shape, and distribution depending on cellular demands and are transported through the cell on microtubules. Miro1, a calcium-binding membrane-anchored GTPase that is strongly associated with mitochondrial transport, is the primary adaptor protein required for the subcellular positioning of this organelle. While mitochondria play an important role in lung physiology, the role of localized mitochondrial function in response to allergen-induced inflammation remained unknown. The main goal of this study was to investigate the effects Miro1-mediated mitochondrial trafficking in allergen-induced inflammation and provide a comprehensive phenotypic readout. To achieve this, our laboratory developed a novel tissue-inducible mouse model that allowed for the deletion of Miro1 from club cells, an abundant epithelial cell subtype in mice, and utilized this system with an established model of chronic allergen-induced inflammation. Our results show that epithelial deletion of Miro1 leads to a heightened inflammatory response, enhanced mucus metaplasia, pronounced tissue remodeling and smooth muscle levels that leads to increased airway hyperresponsiveness following chronic exposure to a complex allergen. Results from this study indicate a possible role for Miro1 in the development and progression of allergic asthma, providing insights into the role of Miro1-mediated mitochondrial positioning in allergic asthma severity. Future studies should be conducted to gain a better understanding of the mechanisms of Miro1 and mitochondrial positioning in asthma.

They may facilitate the adhesion and colonization of bacteria, influence the host immune response, assist spreading of the bacterium by e.g. evading recognition by immune cells, or allow bacteria to dwell within protected niches inside the eukaryotic cell. Exotoxins can be single polypeptides or heteromeric protein complexes that act on different parts of the cells. At the cell surface, they may insert into the membrane to cause damage; bind to receptors to initiate their uptake; or facilitate the interaction with other cell types. For example, bacterial superantigens specifically bind to major histocompatibility complex (MHC) II molecules on the surface of antigen presenting cells and the T cell receptor, while cytolysins cause pore formation. For intracellular activity, exotoxins need to be translocated across the eukaryotic membrane. Gram-negative bacteria can directly inject effector proteins in a receptor-independent manner by use of specialized needle apparatus such as bacterial type II, III, or type IV secretion systems. Other methods of translocation include the phagocytic uptake of bacteria followed by toxin secretion, or receptor-mediated endocytosis which allows the targeting of distinct cell types. Receptor-based uptake is initiated by the binding of heteromeric toxin complexes to the cell surface and completed by the translocation of the effector protein(s) across the endosomal membrane. In the cytosol, toxins interact with specific eukaryotic target proteins to cause post-translational modifications that often result in the manipulation of cellular signalling cascades and inflammatory responses. It has become evident that the actions of some bacterial toxins may exceed their originally assumed cytotoxic function. For example, pore-forming toxins do not only cause cytolysis, but may also induce autophagy, pyroptosis, or activation of the MAPK pathways, resulting in adjustment of the host immune response to infection and modification of inflammatory responses both locally and systemically. Other recently elucidated examples of the immunomodulatory function of cell death-inducing exotoxins include TcdB of Clostridium difficile which activates the inflammasome through modification of cellular Rho GTPases, or the Staphyloccocus d-toxin which activates mast cells. The goal of this research topic was to gather current knowledge on the interaction of bacterial exotoxins and effector proteins with the host immune system. The following 16 research and review articles in this special issue describe mechanisms of immune modification and evasion and provide an overview over the complexity of bacterial toxin interaction with different cells of the immune system.

Exposure to these substances at high concentrations may cause toxic effects to the respiratory and central nervous system; leading possible to death. T protect crew members on disabled submarines, scientists at the U.S. Navy Health Research Center's Toxicology Detachment have proposed two exposure levels, called submarine escape action level (SEAL) 1 and SEAL 2, for each substance. SEAL 1 is the maximum concentration of a gas in a disabled submarine below which healthy submariners can be exposed for up to 10 days without encountering irreversible health effects while SEAL 2 the maximum concentration of a gas in below which healthy submariners can be exposed for up to 24 hours without experiencing irreversible health effects. SEAL 1 and SEAL 2 will not impair the functions of the respiratory system and central nervous system to the extent of impairing the ability of crew members in a disabled submarine to escape, be rescued, or perform specific tasks. Hoping to better protect the safety of submariners, the chief of the Bureau of Medicine and Surgery requested that the National Research Council (NRC) review the available toxicologic and epidemiologic data on eight gases that are likely to be produced in a disabled submarine and to evaluate independently the scientific validity of the Navy's proposed SEALs for those gases. The NRC assigned the task to the Committee on Toxicology's (COT's) Subcommittee on Submarine Escape Action Levels. The specific task of the subcommittee was to review the toxicologic, epidemiologic, and related data on ammonia, carbon monoxide, chlorine, hydrogen chloride, hydrogen cyanide, hydrogen sulfide, nitrogen dioxide, and sulfur dioxide in order to validate the Navy's proposed SEALs. The subcommittee also considered the implications of exposures at hyperbaric conditions and potential interactions between the eight gases. Review of Submarine Escape Action Levels for Selected Chemicals presents the subcommittee's findings after evaluation human data from experimental, occupational, and epidemiologic studies; data from accident reports; and experimental-animal data. The evaluations focused primarily on high-concentration inhalation exposure studies. The subcommittee's recommended SEALs are based solely on scientific data relevant to health effects.
