While studies have revealed precisely timed responses to vocalizations in the auditory cortex (ACx) of many species, few have directly examined the contributions of spike timing to the discrimination of natural sounds by cortical neurons. Songbirds are an attractive choice for examining the processing of complex sounds because they communicate in complex acoustic settings, using vocal calls that bear striking similarities to human speech. They also possess a combination of well-studied vocal communication behavior and well defined auditory neural circuitry. Neurons in field L (the avian analog of ACx) exhibit strong responses to birdsong that could play an important role in song discrimination. In this work, we investigate neural discrimination of conspecific songs in zebra finch field L. We describe a classification framework for quantifying song discrimination based on the responses of single neurons. The technique is first applied to responses of model neurons constructed from experimentally derived spectro-temporal receptive fields of field L. We demonstrate that model neurons can accurately discriminate between songs if the responses are read at relatively fine timescales ([approximate] 10ms). We extend our analysis to electrophysiological recordings from field L neurons. We quantify the performance accuracy of single neurons and examine the timescales relevant for discrimination. A comparison with avian behavioral performance reveals that neural performance matches behavioral accuracy for the best neurons using a spike-timing based code. Finally we examine neural identification of birdsong (targets) in the presence of competing sounds (maskers). We identify two distinct forms of neural interference: addition of spurious spikes during the silent gaps between the target syllables and suppression of informative spikes during the syllables. These effects systematically decrease discrimination performance as the target intensity decreases relative to that of the masker. The behavioral performance of songbirds in a comparable task also degrades in a similar manner. Our results reveal interference effects that could explain the perceptual difficulties that occur when identifying target sounds from a mixture of competing sounds, which is often refered to as the cocktail party problem.

Book excerpt: Abstract: Cortical auditory processing is critical for speech perception, yet we understand little about how the neural representations available at the cortical level allow for sound recognition. In particular, the neural mechanisms for recognition of sounds via spiking patterns are currently unknown, and the characterization of auditory cortical neuron response properties using stimulus-response models remains incomplete. Previous work has shown that spike trains can be used to discriminate between sounds using analytical methods, but how biological circuits might perform such computations remains unclear. Therefore we devised a model for discrimination (inspired by a spike distance metric) that combines a network of integrate-and-fire model neurons with a decision network. We applied this model to the birdsong system, a powerful animal model for the study of audition. We then examined how stimuli could be recognized by sensory systems from spiking patterns by transforming spiking patterns into spatial representations. Previous "spike pattern recognition" models used artificially pre-processed and noiseless signals as input, despite the fact that neural spike trains show variability. Therefore, we used cortical-level neural recordings as input to a novel spike pattern recognition system designed to deal with the intrinsic variability and diverse response properties of cortical spike trains. We show that the model can learn to recognize neural responses to auditory stimuli using spike-timing dependent plasticity, and can play back learned spike patterns in reverse, not unlike the reverse spike train playback observed in hippocampus. A standard model for central auditory neurons is the spectro-temporal receptive field (STRF), which quantifies the spectral and temporal stimulus parameters that modulate neural activity. While the STRF has been used to predict neural responses, a causal connection between the STRF structure and neural responses is lacking. We used an adaptive stimulation paradigm to address this problem. Specifically, for each neuron, we filtered out stimulus frequencies predicted to be unimportant by the STRF (estimated using normalized reverse correlation), and measured responses to these stimuli. For some neurons, this filtering had little effect on neural firing, but adding a noise masker in the unimportant frequency regions adversely affected their responses, suggesting that frequency regions outside the STRF can play a role in determining neural responses.

The question is, whether learning has the power to trigger functional and structural changes in the brain. Several levels of thinking are involved in an interdisciplinary way. Thus, on a psychological level, 3 major topics enter this work: learning, memory and preconscious or pre-attentive perception and processing of information. Pre-attentive perception means that the subjects' attention and awareness is not mirrored in the neuronal response at a great deal. Learning is involved in this study as an improving discrimination of fine frequency and word duration differences; the latter was examined in a group of native and non-native speakers. Memory is referred to as sensory memory, a short-time memory trace that is established through the repetition of the same “standard” stimulus. In the auditory modality this has been termed “echoic memory”. A long, repetitive training engraves deep “traces” into the memory. The lifelong training of one’s native language results in a very fast and highly automated long-term memory access. On a neurophysiological level the main topics are plasticity and the reorganization of the underlying representational brain areas. Plastic changes on a molecular, synaptic and neuronal level and reorganization of cortical “maps” have been demonstrated abundantly in animal studies. On a physical level the measured magnetic fields and the calculation of the source parameters of their underlying neural generators are discussed in the light of the neurophysiological and psychological phenomena. Therefore, the aim of this dissertation thesis was, to transfer the insights of animal plasticity research onto the human brain and to draw a connection line between discrimination learning and the underlying neurophysiological changes. In a second step, these effects of discrimination learning are tested on speech perception.

In contrast to other senses, human auditory cortex receives input that is highly pre-processed in a number of sub-cortical structures; this suggests that even primary auditory cortex already performs quite complex analyses. At the same time, much of the functional role of the various sub-areas in human auditory cortex is still relatively unknown, and a more sophisticated understanding is only now emerging through the use of contemporary electrophysiological and neuroimaging techniques. The integration of results across the various techniques signify a new era in our knowledge of how human auditory cortex forms basis for auditory experience. This volume on human auditory cortex will have two major parts. In Part A, the principal methodologies currently used to investigate human auditory cortex will be discussed. Each chapter will first outline how the methodology is used in auditory neuroscience, highlighting the challenges of obtaining data from human auditory cortex; second, each methods chapter will provide two or (at most) three brief examples of how it has been used to generate a major result about auditory processing. In Part B, the central questions for auditory processing in human auditory cortex are covered. Each chapter can draw on all the methods introduced in Part A but will focus on a major computational challenge the system has to solve. To address these questions, single- and multiple-unit activity was recorded in the primary auditory cortex (A1) and the middle-lateral belt cortex(ML) in two behavioral contexts: 1) while macaque monkeys perform an AM detection task (behaving condition) and 2) while they were passively presented with the same set of stimuli under no task demand (passive condition). Receiver Operating Characteristic (ROC) and Choice Probability (CP) analyses were used to examine a possible relationship between firing rate and/or temporal spike firing pattern and the perceptual performance in the AM detection task. The results showed a significant trial-by-trial correlation between the firing rate of A1 and ML neurons and animals' choice in the AM discrimination task. This is one of the first studies to demonstrate a direct link between auditory single cortical neurons' activity and animals' choice in an auditory perceptual task. AM discriminability based on firing rate of A1 and ML neurons also improved due to engagement in the AM task. In contrast, phase-locking of A1 and ML neurons was found more weakly coupled to animals' choice and behavioral engagement in the task, suggesting the preferential role of rate coding by A1 and ML neurons in the AM discrimination task.

Book excerpt: Perceptual objects are the elementary units used by the brain to construct an inner world representation of the environment from multiple physical sources, like light or sound waves. While the physical signals are first encoded by receptors in peripheral organs into neuroelectric signals, the emergence of perceptual object require extensive processing in the central nervous system which is not yet fully characterized. Interestingly, recent advances in deep learning shows that implementing series of nonlinear and linear operations is a very efficient way to create models that categorize visual and auditory perceptual objects similarly to humans. In contrast, most of the current knowledge about the auditory system concentrates on linear transformations. In order to establish a clear example of the contribution of auditory system nonlinearities to perception, we studied the encoding of sounds with an increasing intensity (up ramps) and a decreasing intensity (down ramps) in the mouse auditory cortex. Two behavioral tasks showed evidence that these two sounds are perceived with unequal salience despite carrying the same physical energy and spectral content, a phenomenon incompatible with linear processing. Recording the activity of large cortical populations for up- and down-ramping sounds, we found that cortex encodes them into distinct sets of non-linear features, and that asymmetric feature selection explained the perceptual asymmetry. To complement these results, we also showed that, in reinforcement learning models, the amount of neural activity triggered by a stimulus (e.g. a sound) impacts learning speed and strategy. Interestingly very similar effects were observed in sound discrimination behavior and could be explain by the amount of cortical activity triggered by the discriminated sounds. This altogether establishes that auditory system nonlinearities have an impact on perception and behavior. To more extensively identify the nonlinearities that influence sounds encoding, we then recorded the activity of around 60,000 neurons sampling the entire horizontal extent of auditory cortex. Beyond the fine scale tonotopic organization uncovered with this dataset, we identified and quantified 7 nonlinearities. We found interestingly that different nonlinearities can interact with each other in a non-trivial manner. The knowledge of these interactions carry good promises to refine auditory processing model. Finally, we wondered if the nonlinear processes are also important for multisensory integration. We measured how visual inputs and sounds combine in the visual and auditory cortex using calcium imaging in mice. We found no modulation of supragranular auditory cortex in response to visual stimuli, as observed in previous others studies. We observed that auditory cortex inputs to visual cortex affect visual responses concomitant to a sound. Interestingly, we found that auditory cortex projections to visual cortex preferentially channel activity from neurons encoding a particular non-linear feature: the loud onset of sudden sounds. As a result, visual cortex activity for an image combined with a loud sound is higher than for the image alone or combine with a quiet sound. Moreover, this boosting effect is highly nonlinear. This result suggests that loud sound onsets are behaviorally relevant in the visual system, possibly to indicate the presence of a new perceptual objects in the visual field, which could represent potential threats. As a conclusion, our results show that nonlinearities are ubiquitous in sound processing by the brain and also play a role in the integration of auditory information with visual information. In addition, it is not only crucial to account for these nonlinearities to understand how perceptual representations are formed but also to predict how these representations impact behavior.
