Cette opération a pour but de préserver l'intégrité de tout organisme vivant. Le décryptage moléculaire des différentes étapes requises pour expliquer les systèmes complexes d'interactions (transduction du signal) nécessite de disposer de méthodes biophysiques puissantes, susceptibles de fournir des informations pertinentes pour la construction de modèles physiologiques ou pathologiques intégrés. Ainsi, trois méthodes biophysiques complémentaires (calorimétrie de titration isotherme, résonance magnétique nucléaire et fluorescence) ont été analysées en terme de stratégies d'investigation et de limites de validité. Par la suite, deux sujets liés à des modulations de pathogénicité bactérienne et virale ont été abordés. Dans une première phase, le phénomène de résistance de souches bactériennes aux molécules antibiotiques a été abordé selon deux angles différents. Tout d'abord, plusieurs expériences RMN ont été effectuées afin d'améliorer le niveau de connaissance actuel concernant les détails moléculaires de la transduction du signal requise pour la destruction des antibiotiques de type beta-lactames. Plus particulièrement la levée de répression du gène codant pour la beta-lactamase a été étudiée. Dans une seconde approche, de nouvelles cibles protéiques ont été recherchées et la structure en solution d'une nouvelle protéine a été résolue. Les résultats obtenus et les modèles déduits sont exposés dans ce manuscrit. Pour terminer, une étude combinant génie génétique, RMN, cristallographie et fluorescence in vivo a ouvert une brèche, à l'échelle atomique, vers la compréhension intégrée du phénomène de transmissibilité inter-espèce du virus influenza (notamment grâce à l'obtention de la première structure haute-résolution du ITagment C-terminal de la sous-unité PB2 de la polymérase virale d'irifluenza).

Jacques Lafontaine was successively assistant Professor at Paris Diderot University and Professor at the University of Montpellier, where he is presently emeritus. His main research interests are Riemannian and pseudo-Riemannian geometry, including some aspects of mathematical relativity. Besides his personal research articles, he was involved in several textbooks and research monographs.
