The aim of this book is to provide an overview of research advances relating to the principle and applications of DLS in various fields. The book is divided into two parts Part 1 discusses the uses of DLS in material science and engineering applications and Part 2 focuses on applications of DLS in biological sciences. Chapter 1 aims to provide an overview of the working principle, mathematical models and different types of DLS analysis methods. In addition, recent trends in DLS studies and applications in various fields are also discussed. Chapter 2 discusses the uses of DLS for nanomaterial characterization in terms of the size, size distribution and zeta potential of particles. Light scattering by small particles such as, for instance, droplets in the terrestrial clouds is a well understood area of physical optics. On the other hand, exact theoretical calculations of light scattering p- terns for most of nonspherical and irregularly shaped particles can be performed only for the restricted values of the size parameter, which is proportional to the ratio of the characteristic size of the particle to the wavelength?. For the large nonspherical particles, approximations are used (e. g. , ray optics). The exact th- retical techniques such as the T-matrix method cannot be used for extremely large particles, such as those in ice clouds, because then the size parameter in the v- iblex=2?a/???,wherea is the characteristic size (radius for spheres), and the associated numerical codes become unstable and produce wrong answers. Yet another problem is due to the fact that particles in many turbid media (e. g. , dust clouds) cannot be characterized by a single shape. Often, refractive indices also vary. Because of problems with theoretical calculations, experimental (i. e. , la- ratory) investigations are important for the characterization and understanding of the optical properties of such types of particles. Book excerpt: Dynamic light scattering (DLS) is a method to size submicron particles by measuring their thermal motion (diffusion) in suspensions and emulsions. However, the validity of the Stokes-Einstein equation that relates the diffusion coefficient and the particle size is limited to spherical particles and very low concentrations. Within this thesis, DLS is used for the characterization of suspensions of pyrogenic silica which consists of fractal-like aggregates composed of sintered spherical primary particles. These structural features clearly complicate the understanding of DLS experiments and have been a severe obstacle to employing DLS as routine standard tool for the characterization of pyrogenic silica. The main objective of this thesis is therefore to evaluate the application of DLS in product development and quality assurance of pyrogenic silica industry, what essentially means to identify those structural properties of fractal aggregates which are measurable with DLS and to quantify the method's sensitivity to changes in these properties. The investigations presented here are split up into four parts, simulations that establish a relation between structural and hydrodynamic properties, experiments validating the simulation results, the characterization of concentrated suspensions and the application-oriented analysis of DLS data for specific industrially relevant measurement tasks.

Book excerpt: We present a new 3D cross-correlation instrument that not only allows for static and dynamic scattering experiments with turbid samples but measures at four angles simultaneously. It thus extends the application of cross-correlation light scattering to time-resolved studies where we can, for example, efficiently investigate the temporal evolution of aggregating or phase separating turbid dispersions. The combination of multiangle 3D and on-line transmission measurements is an essential prerequisite for such studies. This not only provides time-resolved information about the overall size and shape of the particles through measurements of the mean apparent radius of gyration and hydrodynamic radius, but also on the weight-average apparent molar mass via the absolute forward scattering intensity. We present an efficient alignment strategy based on the novel design of the instrument and then the application range of the instrument using well-defined model latex suspensions. The effectiveness of the cross-correlation multiangle technique to monitor aggregation processes in turbid suspensions is finally shown for the acidification of skim milk during the yoghurt-making process. Due to the self-assembled nature of the casein micelles an understanding of the sol- el process induced by the acidification is only feasible if time-resolved light-scattering experiments on an absolute scale are possible under industrially relevant conditions, where the casein solutions are highly turbid.

Book excerpt: Abstract: A novel approach to a light scattering experiment that employs a lounge image capture assay has been developed for both static and dynamic measurement on a wide variety of samples. In this work, the 'Development and Application of an Area Recorded Generalized Optical Scattering Technique', abbreviate[d] as ARGOS, an approach is based on the placement of a translucent screen where the scattered light, now converted to a diffuse light, is imaged as a function of time. The placement and size of the screen determines the range of scattering wave-vectors, while the time between images determines the fastest dynamic resolvable. The overall experimental stability determines the slowest dynamic resolvable and so can be made arbitrarily long. The scattering intensity is measured by a camera whose dynamic range (number of bits per pixel), resolution, and speed determines the sensitivity and accuracy of intensity of the scattered light. The system allows for an extremely well-defined scattering experiment geometry, where images may be manipulated to extract structure (i.e. average I(q)over t) and dynamics (average I(q, t) over q) for a wide range of samples (liquids and solids). A number of innovation were developed. Instead of a beam stop to block the high intensity thru-beam, a thru-beam attenuator was developed such that the exact center of the scattering geometry is determined for each image acquired. A variety of image processing algorithms were also developed to correct for dead pixels, camera response, and intensity normalization. Of particular use was the development of the root-mean-square difference (RSD) image to probe dynamic. Here, the first image is taken as the time-zero reference image and subtracted from all succeeding images where averaged over a specified q and plotted as a function of time revealing the build up image changes due to dynamics. The technique was validated for both static and dynamic measurements with a set of dilute suspended latex spheres in water (sizes ranging from 0.01 to 2 [mu]m). The static light scattering result in terms of particle radius and dimension closely matched the expected values. The dynamic light scattering results could only several the end of the dynamics of these suspended latex spheres due to Brownian motion because of the speed limitation of the camera. However, sedimentation dynamics that are much slower, were early observed. The ARGOS technique was then applied to two biological systems, the evolution of a dilute suspension of E. coli bacteria through the life-cycle and the evolution of the same suspended bacteria but infected with the MS2 bacteria-phage virus. High temporal resolution data were obtained of the bacteria life-cycle which were correlated with size, structure, and dynamics that revealed potentially new insights on behavior not well understood in the literature. The MS2 infected bacteria revealed the effect of the bacteria-phage on every character of the E. coli life-cycle. The successful development and implementation of ARGOS here has also revealed avenues to greatly improve the performance.
