Book excerpt: This thesis presents research on novel X-ray imaging methods that improve the study of specimens with small density differences, revealing their inner structure and density distribution. Exploiting the phase shift of X-rays in a material can significantly increase the image contrast compared to conventional absorption imaging. This thesis provides a practical guide to X-ray phase-contrast imaging with a strong focus on X-ray speckle-based imaging, the most recently developed phase-sensitive method. X-ray speckle-based imaging only requires a piece of abrasive paper in addition to the standard X-ray imaging setup. Its simplicity and robustness combined with the compatibility with laboratory X-ray sources, make it an ideal candidate for wide user uptake in a range of fields. An in-depth overview of the state of the art of X-ray speckle-based imaging and its latest developments is given in this thesis. It, furthermore, explores a broad range of applications, from X-ray optics characterisation, to biomedical imaging for 3D virtual histology and geological studies of volcanic rocks, demonstrating is promising potential. Moreover, the speckle-based technique is placed in the context of other phase-sensitive X-ray imaging methods to assist in the choice of a suitable method, hence serving as a guide and reference work for future users.

Book excerpt: The evolution of x-ray equipment has been stagnant because of its demanding design requirements, but clinical need for an advanced imaging technique has been continually growing. To contribute to hardware advances in the x-ray imaging devices, we designed and implemented innovative instruments by applying both mechanical and electrical principles. Herein, we explore two essential novel instruments for x-ray imaging: (1) a new electric motor for an MR-compatible x-ray tube and (2) a testing system for a high-energy x-ray detector. Because each imaging modality has different strengths and limitations, we are able to obtain more accurate medical images by combining at least two modalities. A hybrid x-ray/MR imaging system, leveraging the complementary advantages of both x-ray and MR imaging, is ideal for image guidance of catheters, guide wires, and prosthetic devices in real time. However, the performance of conventional x-ray tube motors is degraded when exposed to an external magnetic field, thus we developed and evaluated a new x-ray tube motor that can operate in the presence of the strong magnetic fields of an MRI machine. We further explored simulation models to improve the performance and dynamic characteristics of the new motor. High-energy x-ray imaging used in concert with low-energy x-ray imaging creates clearer and more accurate images. It is necessary to have a thicker scintillating phosphor layer to improve quantum efficiency for high-energy x-rays, but the spreading becomes more severe in that case. To resolve this issue, we can use a thick scintillating layer segmented by other thin layers (a.k.a. pixelated detector). Even though the pixelated detector prototype based on Sawant's design demonstrates relatively high detective quantum efficiency (DQE), both measured DQE and spatial resolution differ from corresponding expectations from theory and simulations. In order for the Monte Carlo method to accurately simulate the imaging performances, the characteristics of the basic pixelated unit must be understood. Therefore, we developed a new instrument to measure both light reflection and transmission distribution within the basic detector unit as a function of material, surface, and angle of incidence. Based on manufacturing procedures and the light distributions of the candidate samples, we also made valuable suggestions for a better, more profitable detector. We expect that our instruments will lead the next generation of diagnostic and therapeutic equipment to produce accurate diagnoses and efficient treatments for various disease states.

Book excerpt: Interest in phase sensitive x-ray imaging techniques has grown rapidly in recent years. Numerous techniques have been demonstrated to measure the phase shift of x-rays as they pass through matter. One such method, grating-based interferometry, has shown the most potential for clinical use. This dissertation continues our group's efforts at characterizing the Talbot-Lau interferometer, with the overall goal of determining the clinical utility of the system. This work first covers the basics of x-ray phase contrast imaging with a grating interferometer. Phase contrast imaging with this method has the advantage over conventional absorption imaging in that it allows for the simultaneous measurement of multiple contrast mechanisms. This multi-contrast imaging modality has the potential to improve the diagnostic capabilities of traditional imaging systems. It has unique noise properties that allow it to move to higher spatial resolution at a lower dose penalty, as well as improved contrast for certain material combinations. However, questions still remain about where such a system would fit in the clinical realm. This dissertation delves into new research demonstrating the effects of beam hardening, improving the phase retrieval technique, determining the best method for grating fabrication, and exploring the impact of the small-angle scattering, or dark field, contrast mechanism. Both traditional beam hardening artifacts and new ones are shown, and their implication for clinical systems is discussed. The improved phase retrieval techniques aim at reducing the temporal requirements for the current acquisition method. While most of them succeed at reducing the time requirements, they also all come with an associated cost to the final image. Along with improved signal extraction, improved grating fabrication is discussed. The design and construction of a new grating fabrication system is detailed, demonstrating its capability of both producing absorption gratings with greater metal deposition depths and using cheaper, alternative absorbers. Finally, the impact of the dark-field contrast mechanism is discussed, highlighting the interplay between high-quality, low-noise phase and dark-field images.
