This dissertation describes bioactives that have been chemically incorporated into novel, biodegradable polymers via covalent bonds for controlled, sustained, and tunable release properties. Bioactives are released from polymers via hydrolytic degradation. The polymers described herein utilize alternative synthetic methods and a wide array of bioactives, including antibiotics, antioxidants, antimicrobials, and anti-inflammatory drugs. One goal is to focus on naturally-occurring bioactives that are generally regarded as safe (GRAS) by the FDA. First, poly(anhydride-amides) comprised of ampicillin were synthesized and formulated as coatings. Polymer adhesion onto medical-grade stainless steel surfaces was assessed and in vitro release characterized. Cytocompatibility and antibacterial activity elucidated polymer safety and efficacy for potential in vivo use. These localized delivery systems could mediate the issues caused by implant surgery. Second, poly(anhydride-esters) comprised solely of naturally-occurring phenols and EDTA were synthesized and physicochemical properties determined. Bioactive release was ascertained, in addition to antioxidant activity and activity against Gram-positive and Gram-negative bacteria. These polymers can act as potential preservatives, increasing cosmetic and food product shelf life through antioxidant and antimicrobial pathways. Third, through environmentally sustainable (i.e, green) methods, polyesters with pendant anti-inflammatory groups and a sugar-based backbone were prepared with minimal solvent use, enzymatic catalysis, and biorenewable reactants and reagents. Three comonomers of varying hydrophobicity were tested to elucidate changes in polymer thermal properties and bioactive release rate. Fourth, poly(anhydride-esters) with a mannitol backbone and multiple bioactive groups per repeat unit were developed as the first linear, biodegradable polymers with high bioactive loading (~70%) using a polyol. In vitro ibuprofen release was quantified and an anti-inflammatory assay determined that bioactive retained activity upon polymer degradation. Alteration of polyol, bioactive class, and other facets leads to highly tunable polymer properties. Last, to combat bacterial spoilage and oxidation, poly(anhydride-esters) containing natural antimicrobials were designed for food applications. Bioactive released from polymer exhibited radical scavenging ability and antibacterial activity. Furthermore, polymers were blended with current food packaging materials (e.g., polyethylene) and molded into films for active food packaging that contains a higher percentage of biodegradable content.

Nature has solved this issue with the use of cationic antimicrobial peptides, which act as nonspecific antibiotics against invading species. Herein, we have tried to mimic this general mechanism in a biocompatible and biodegradable polymer micelle based on the polymerization of naturally occurring amino acids lysine and phenylalanine linked to a PEG tether. This amphiphilic structure allows for the spontaneous collapse into stable nanoparticles in solution, which contains a hydrophilic outer layer and a hydrophobic core. Our polymers have shown activity against clinically relevant strains including Methicillin Resistant S. epidermidis, B. subtilis, K. pneumoniae, and P. aeruginosa. To further the application of our biopolymers, we have used them as drug delivery vehicles as well. First, we have used an anionic analogue based on glutamic acid to encapsulate a super hydrophobic drug Tanshinone IIA, and use it against a hepatoma bearing mouse model. Second, we have used a cationic analogue to form a complex with miRNA-139 and use it against a hepatoma bearing mouse model as well. In both cases, our PEG poly(amino acids)s have shown promising efficacy in drastically reducing the tumor size compared to the control only. Taken together, our results show that our nanoparticles have the potential to be versatile biomaterials as antibacterials as well as drug delivery vehicles in vivo.

In recent years, the development of multifunctional nanomaterials has provided a new diagnosis and treatment platform to combat cancer. Polymer-inorganic nanomaterials with novel structures such as bowl-shaped/Janus/core-shell have drawn much attention owing to their diversity in composition or asymmetry in structure. More importantly, imparting unique optical, electrical, and magnetic properties to these nanocomposites can further extend their function repertoire. However, to fulfill this vision, fundamental understandings regarding strategies of precise synthesis, mechanisms of structure formation, in vivo synergistic effects in bioapplications, and biosafety of these materials are needed. Besides, nanomaterials with novel structures are well positioned for imaging-guided cancer theranostic. On one hand, nanomaterials themselves are suitable for imaging because of their intrinsic properties such as fluorescent or magnetic properties. On the other hand, nanomaterials can serve as functional platforms that integrate various therapeutic modalities including photothermal therapy, chemodynamic/ion-interference therapy, photodynamic therapy, and cuproptosis to efficiently kill cancer cells. This Research Topic aims at collecting works about synthesis, and biomedical applications of polymer/mesoporous inorganic nanomaterials, especially in the aspect of novel synthetic approaches for fabricating nanomaterials with unique structures. Additionally, we hope that in-depth research articles on this topic can provide insights into the mechanism of nanomaterials acting in cancer diagnosis and therapy. These include the mechanisms of customized drug load/release and synergistic effects in theranostics of these materials. Meanwhile, elucidations of key proteins’ roles in cancer development are also anticipated.
