Book excerpt: L'administration de préparations pharmaceutiques par voie parentérale s'est développée au XIXème siècle, suite aux découvertes de Pasteur qui ont conduit à la stérilisation des préparations injectables. Parmi ces préparations, les lyophilisats injectables sont des poudres stériles, réparties en doses unitaires, lesquelles après reconstitution sous forme liquide, sont destinées à être injectées ou perfusées. La lyophilisation, procédé utilisé pour la fabrication de cette forme galénique, est une opération de séchage qui vise à limiter au maximum les altérations de substances ou principes actifs fragiles en solution. Le procédé de lyophilisation est complexe et nécessite une maîtrise de tous les paramètres physiques et environnementaux afin de garantir la qualité du produit fini. Les contrôles effectués sur le produit en fin de fabrication ne suffisent pas à garantir l'obtention de la qualité requise. Dans ce but, la validation du procédé de fabrication permet d'apporter les preuves documentées que ce procédé permet d'obtenir, de façon reproductible, un produit répondant aux spécifications et aux critères de qualité définis par les BPF. Ce mémoire a pour objet de présenter la qualification d'une installation de lyophilisation et la démarche de validation du procédé de fabrication d'un lyophilisat injectable au moyens de protocoles issues de situations réelles et présentés sous formes de modes opératoires.
