The aphid-like insect feeds on Vitis vinfera L. roots producing swellings that decay and lead to vine decline. The only long-term economic solution for control of grape phylloxera (GP) is to plant vines grafted onto a resistant rootstock, but over 6000 acres of Oregon's wine grapes are planted with root-susceptible V. vinfera cultivars. Ten GP infestations have been verified throughout the state since August 1990. The objective of this research was to characterize GP development and within-site distribution in Western Oregon. Grape phylloxera were collected from roots dug at two depths (15-30 cm and 30-45 cm) in three infested vineyards in the Willamette Valley during the summers of 1992 and 1993. A sieve centrifugation method was used to extract GP for counting. The number of eggs, nymphs, n/adults (late-instar), and adults per 30 grams of roots were recorded twice a month. Mean population levels were calculated for each site, sampling date and depth. Population development of GP varied between sites and between dates sampled. Population size appeared to be directly related to soil temperature. The site with the highest overall GP populations had the warmest soil temperatures but the least above-ground symptomatic vine decline ratings. Nymphal population peaks, identified at one site, may correlate with generation time or changes in root and soil environments. Similar numbers of GP were found at both depths sampled, but higher populations were collected from vine roots sampled on the downhill side of an infestation and from vines immediately adjacent to satellite infestations. Sticky-trunk-wraps, placed on the base of vine trunks from June to November at the three sites, collected emerging crawlers (nymphs) and winged-alates in July/August of 1993. Development of GP populations collected from each of the three sites was studied in the laboratory at five constant temperatures (7, 10, 13, 16, and 21 °C). No significant differences in development were observed between the three populations. Grape phylloxera generally did not molt past the nymphal stage at 7, 10, 13, and 16°C. The first adults appeared before day 35 at 21°C and this was followed by a significant increase in first generation eggs. In contrast to the laboratory results, eggs were first recovered from field extractions in April/May when mean soil temperatures were 10.5 to 12.0°C in 1993. Eggs were not produced from laboratory populations below 16°C. These results indicate that either the field conditions allowed GP to develop at lower temperatures or the laboratory assay did not provide sufficient time to full development. The low temperature threshold calculated for zero development from the laboratory assay was 5°C. A degree day accumulation, which better reflects GP field development, was estimated at 437.5 using maximum field soil temperatures and a vine development threshold of 10°C. Further use of degree day models may help to predict generation time of GP.

In recent years, grape acreage has increased dramatically in new regions, including the United States of America, Chile, Asia (China and India), and Turkey. A major limiting factor to the sustained production of premium grapes and wines is infections by viruses. The advent of powerful molecular and metagenomics technologies, such as molecular cloning and next generation sequencing, allowed the discovery of new viruses from grapes. To date, grapevine is susceptible to 64 viruses that belong to highly diverse taxonomic groups. The most damaging diseases include: (1) infectious degeneration; (2) leafroll disease complex; and (3) rugose wood complex. Recently, two new disease syndromes have been recognized: Syrah decline and red blotch. Losses due to fanleaf degeneration are estimated at $1 billion annually in France alone. Other diseases including leafroll, rugose wood, Syrah de cline and red blotch can result in total crop loss several years post-infection. This situation is further exacerbated by mixed infections with multiple viruses and other biotic as well as adverse abiotic environmental conditions, such as drought and winter damage, causing even greater destruction. The book builds upon the last handbook (written over twenty years ago) on the part of diagnostics and extensively expands its scope by inclusion of molecular biology aspects of select viruses that are widespread and economically most important. This includes most current information on the biology, transmission, genome replication, transcription, subcellular localization, as well as virus-host interactions. It also touches on several novel areas of scientific inquiry. It also contains suggested directions for future research in the field of grapevine virology.

Book excerpt: Grapevine phylloxera (Daktulosphaira vitifoliae Fitch) is a destructive, root feeding insect pest that is difficult to detect and is a major biosecurity pest which causes economic damage to ungrafted European grapevines Vitis vinifera L. Early detection of this pest would enable rapid implementation of quarantine procedures and rootstock replanting and is vital to ensure the sustainability and profitability of the viticulture industry both in Australia and internationally. This dissertation explores an untargeted metabolomics approach to early detection of grape phylloxera by studying biochemical changes in the metabolome of leaf material of V. vinifera in response to infestation. LC-MS and NMR analytical platforms were applied to leaf samples acquired under both glasshouse and field conditions. Metabolic profiling using LC-MS was conducted on V. vinifera cv. Shiraz independently infested with two endemic high-virulence genetic strains of grape phylloxera, G1 and G4, and uninfested grapevines grown under glasshouse conditions. Statistical discrimination between treatments (infested vs. uninfested) was observed from 4 weeks post-infestation. Several unidentified metabolites were detected which were differentially regulated in infested grapevines. No discrimination was observed between G1 and G4-infested grapevines, tentatively suggesting that a grape phylloxera-induced stress response in grapevines may be independent of grape phylloxera genetic strain. These results also indicated a measurable metabolic response to phylloxera infestation in relation to the relative abundance of active life stages of grape phylloxera on the roots at the time of sampling. A second glasshouse trial compared the metabolic profile of G1-infested V. vinifera cv. Chardonnay with nutrient stressed, water stressed and unstressed grapevines over a 14-week period using LC-MS. Statistical discrimination between grape phylloxera-infested grapevines, uninfested controls and nutrient-stressed grapevines was achieved at 14 weeks post-infestation, with partial separation of phylloxera-infested from water stressed grapevines. Nineteen statistically significant metabolites were identified. Results principally indicated induction of both phenylpropanoid and shikimate pathways and the production of carotenoid oxidation products in response to grape phylloxera infestation. A field study of G1-infested mature V. vinifera cv. Chardonnay in a commercial vineyard over a single growing season was conducted and analysed using NMR and LC-MS techniques. Statistical analysis of LC-MS data showed separation of phylloxera-infested from uninfested grapevines at both the peak aboveground first instar abundance and two months prior. Analysis confirmed induction of the phenylpropanoid pathway in response to grape phylloxera infestation. Statistical discrimination of treatments was observed in NMR data of aqueous extracts of leaf material, with differential regulation of carbohydrates, amino acids and phenylpropanoid products. Overall, quercetin was identified as the main biomarker of grape phylloxera infestation as quercetincontaining flavonoid metabolites were observed up-regulated by LC-MS and NMR in infested grapevines under both glasshouse and field conditions. Further validation is recommended in order to develop a robust early detection system for grapevine phylloxera using biochemical markers.
