Book excerpt: Cette étude est consacrée à l'enrobage en discontinu des particules solides, poreuses ou non-poreuses, en lit fluidisé par l'injection d'une solution aqueuse contenant 200 kg/m3 de chlorure de sodium. Nous avons mis au point un dispositif expérimental qui a permis d'étudier l'influence de la taille et la distribution de la taille, et de la porosité des particules sur l'efficacité et la stabilité de l'opération ainsi que sur les mécanismes de croissance. Par ailleurs, nous avons aussi étudié l'influence des conditions opératoires sur la morphologie du dépôt. Les résultats ont montré que pour les particules de tailles supérieures à 160 um, la croissance des particules est gouvernée par le recouvrement en surface. En revanche, la croissance des particules de tailles inférieures à 160 um se fait par le mécanisme d'agglomération. Cependant, l'enrobage de fines particules peut être réalisé en les mélangeant avec une quantité adéquate de grosses particules de même nature. En ce qui concerne la morphologie du dépôt, les résultats expérimentaux ont montré qu'une diminution de la taille des gouttes (augmentation du débit d'air d'atomisation) ou de la tension superficielle (ajout d'un surfactant) peuvent conduire à un dépôt uniforme et compact à la surface de particules solides. Nous avons aussi développé des modèles de connaissances basés sur le concept du bilan de population, qui représentent trés bien l'ensemble des résultats expérimentaux obtenus au cours de cette étude.

Les systèmes d’enrobage à pulvérisation par le bas et à pulvérisation tangentielle conduisent à des enrobages de meilleure qualité que les systèmes à pulvérisation par le haut. Les récents progrès techniques des équipements et l’avancée des connaissances en formulation galénique rendent possibles de nombreuses applications galéniques innovantes de l’enrobage en lit fluidisé. Le dépôt de l’agent d’enrobage sous forme liquide peut être remplacé par l’utilisation de poudres sèches ou par un procédé de fusion lipidique et ce pour tous les systèmes de pulvérisation. Le microenrobage lipidique est préférentiellement réalisé en lit fluidisé avec pulvérisation par le haut alors qu’une pulvérisation par le bas ou tangentielle est plutôt employée pour l’enrobage de pellets par des poudres sèches. Le lit fluidisé avec pulvérisation par le bas offre la possibilité de pouvoir préparer des formes galéniques innovantes et/ou pour des voies d’administration particulières comme : des poudres sèches à inhaler (DPI) ; des pellets enrobés pour libération colique ; des poudres issues des biotechnologies pour administration épidermique à produire en lots de faible taille ; des chewing gums à libération ciblée et prolongée du principe actif et des gélules végétales enrobées gastrorésistantes prêtes à l’emploi. Enfin, le désir d’enrober des particules toujours plus petites a conduit au développement d’un nouveau type d’équipement : le lit fluidisé rotatif. : Galénique ; Enrobage ; Pelliculage ; Lit Fluidisé ; Film ; Poudre ; Libération prolongée ; Gastrorésistance.
