Book excerpt: Dans le domaine de l'environnement, il est très important de disposer d'outils capables de détecter la présence de polluants ou de substances toxiques au sein des milieux aqueux, tels que les écosystèmes aquatiques, l'eau des stations d'épuration, les effluents industriels. Les techniques classiques d’analyse pour la détection d’une espèce (bio) chimique sont généralement complexes, coûteux, volumineux et souvent difficiles à mettre en œuvre. De plus, les phases de préparation des échantillons et d’exploitation des résultats augmentent souvent très fortement la durée totale d’analyse. Depuis une trentaine d’années, ils font face à l’avènement des capteurs (bio)chimiques. Malgré une très forte demande et de nombreuses recherches effectuées, très peu de réalisations commerciales ont vu le jour, surtout dans le domaine de l’environnement. La difficulté majeure qui freine le développement industriel de ces capteurs est liée aux problèmes de sélectivité et de stabilité de l’élément sensible fixé sur la surface du transducteur au cours de l’étape de fonctionnalisation. Pour remédier à ces inconvénients, notre recherche est orientée vers la création de nouvelles matrices d’immobilisation de l’élément sensible et l’élaboration de nouveaux matériaux d’électrodes. Ce travail de thèse porte sur la réalisation et la caractérisation de capteurs chimiques et biochimiques basés sur l’utilisation de trois types de transduction. Le premier transducteur est conductimétrique avec des électrodes interdigitées : Le premier est un biocapteur bienzymatique à protéases immobilisées pour la détection de protéines en tant que marqueur de la matière organique ; Le deuxième est un biocapteur élaboré pour la détermination des nitrates en utilisant l’enzyme nitrate réductase (RN) de l’aspergillus niger immobilisé dans un film de PVC, en présence de Nafion® et de Méthyle viologen en tant que médiateur ; Le troisième est un capteur chimique pour détecter l’ammonium en immobilisant sur des électrodes interdigitées des membranes en PVC contenant la nonactine comme un élément de reconnaissance de l’ammonium ; Le deuxième type de transducteur est voltamétrique avec des électrodes en carbone vitreux pour la détermination de traces de mercure et de nickel. Le troisième type est un transducteur à fibre optique (chimiluminescence) pour la détection du cobalt immobilisé dans un film de chitosan ; le principe de ce capteur repose sur l’effet catalytique des ions métalliques comme le cobalt sur la réaction de chimiluminescence du luminol en présence de peroxyde d’hydrogène. Ces (bio)capteurs ont été validés sur des échantillons d’eau naturelle. Ils ont prouvé qu’ils constituent une technique d’analyse alternative performante, présentant des limites de détection très sensibles, de l’ordre du ng/l au μg/l, comparables à celles trouvées dans la littérature. Ils ont montré une rapidité de réponse de 5 à 7 minutes, une bonne sélectivité avec une bonne reproductibilité pour une longue durée de vie de un à trois mois. Enfin, ces techniques à faible coût, fonctionnant à température ambiante, sont prêtes à être utilisées sur le terrain. Ils permettent ainsi de réaliser des analyses de contrôle et de surveillance de milieux complexes sans traitement préalable des échantillons

Ils ont constitué les alternatives les plus séduisantes pour les méthodes analytiques classiques dans des domaines aussi variés que l'agro-alimentaire, la médecine et la biologie clinique, ou le contrôle de la qualité de l'environnement. Dans ce travail de recherche, nous nous sommes intéressés, dans un premier volet, aux développements des immunocapteurs et capteurs électrochimiques à base des polymères à empreintes moléculaires (MIPs) pour le contrôle de la qualité des miels. La première partie de ce volet concerne le développement des immunocapteurs sur des structures dites Bio-MEMS basées sur des microélectrodes en or. L'élaboration de ces immunocapteurs a été dédiée à la détection des résidus d'antibiotiques à savoir la sulfapyridine (SPy) et la tétracycline (TC). Une nouvelle structure des nanoparticules magnétiques (MNPs) revêtues du copolymère poly (acide pyrrole-co-pyrrole-2-carboxylique) a été exploitée dans ces travaux pour leur réseau d'immobilisation tridimensionnel ainsi que pour leur stabilité pendant de longues périodes. La détection de la SPy et de la TC a été réalisée par différentes approches compétitives en utilisant des anticorps polyclonaux. Dans la deuxième partie de ce volet, nous avons fabriqué des capteurs à base des MIPs pour la détection de la sulfaguanidine, la doxycycline et le chloramphénicol dans le miel. Ces dispositifs ont été développés sur la surface des électrodes sérigraphiées en or en employant une matrice polymérique du polyacrylamide en présence des molécules empreintes. Les performances de ces capteurs et biocapteurs (limite de détection, sélectivité, reproductibilité, application dans les milieux réels) ont ensuite été évaluées. Dans un deuxième volet, nous nous sommes parvenus à appliquer un dispositif de la langue électronique voltammétrique (Langue-EV) pour des analyses agroalimentaires et biomédicales. Dans un premier temps, nous avons discriminé les miels issus de quatorze régions de la France et du Maroc par le dispositif de la Langue-EV. Nous nous sommes aussi parvenus à démontrer la fiabilité de ce dispositif à prédire les résultats des différents paramètres physico-chimiques d'après les réponses des méthodes analytiques utilisées. Dans un deuxième temps, nous sommes passés à l'application du dispositif de la Langue-EV en analyse biomédicale dans le but de discriminer les urines des patientes souffrants des infections urinaires avec celles des femmes saines.
