Available in PDF, EPUB and Kindle. Book excerpt: Chapter I: Design and synthesis of SLe sp{\rm x} Mimetics. The design and synthesis of several SLe sp{\rm x} mimetics are described. In mimetic A, an ethylene glycol linkage bridges the fucose and galactose moieties, and a carboxymethyl group replaces the sialic acid. In mimetic B, a scD-tartaric acid derivative replaces the trans-3,4-dihydroxyl groups of the glucosamine, and links the fucose and carboxypentyl groups. These compounds were tested as inhibitors of the binding of SLe sp{\rm x} to E-selectin. Mimetic C contains a macrocyclic structure which restricts the conformation by linking the fucose and galactose moieties. Mimetic D is a glycopeptide derived from the structure of B. Mimetics C and D are expected to show better binding activities than compound A and B, respectively. Chapter II: Enzymes in organic chemistry. (1) A strategy was developed for the enzymatic solid-phase synthesis of tetrasaccharide rm NeuAc\alpha2 \to 3Gal\beta1 \to 4GlcNAc\beta1 \to 3Gal, an inhibitor of the attachment of H. pylori to mucous cells. The conversion for each step was found to be > 98%. (2) Several oligosaccharides containing negatively charged groups, sulfates or carboxylates, were synthesized for the substrate specificity study of glycosyltransferases potentially responsible for the biosynthesis of the natural ligand(s) for L-selectin. (3) Preliminary research is reported on the application of galactose oxidase for the synthesis of a potential substrate for alpha -2,3-fucosyltransferase VII, as well as other oligosaccharides. An improved procedure was developed for the GOase catalyzed oxidation of weak substrates to nearly completion. (4) A kinetics study of the lipase resolution of alcohol 82, and other nucleophiles, was performed for comparison to the antibody 21H3 catalyzed transesterification. The product inhibition and solvent effects were also discussed. (5) The regioselectivity of the transesterification of disaccharides catalyzed by enzymes such as subtilisin was investigated. Conditions were optimized to obtain either a 6 sp\prime -monoacetylated compound or a 6,6 sp\prime -diacetylated compound as the major product.

Available in PDF, EPUB and Kindle. Book excerpt: Glycans play essential roles in diverse biological and etiological processes and their structural complexity endow various functions. The glycome is the entire set of glycans produced by an individual organism. As the glycan microarray emerged, a good amount of knowledge has been obtained in understanding the functions of glycans. However, limited accessibility of glycans is a major obstacle to the functional glycomics study. Although isolation from biology samples provided some structures, the low abundance of glycans obtained and the difficulty in complete structural assignment restricted the subsequent assay. To circumvent this limitation, many synthetic strategies, including chemical, enzymatic and chemo-enzymatic ones have been developed to make libraries of structurally defined complex glycans available. The glycans provided by these techniques combined with high-throughput glycoarray techniques have broadened and deepened our understanding about functional glycomics. The aim of this book is to provide a comprehensive review of the current state of the synthetic glycome and a brief introduction of the application of the synthetic glycome in glycoarray assay. Accordingly, synthetic strategies toward generating glycans with comprehensive structures as well as the glycoarrays to unveil the glycan functions are described in this book.

This new volume contains critical reviews covering the latest findings in both chemical and biological sciences, and demonstrates the interdisciplinary nature of modern carbohydrate research. This book addresses diverse applications that continue to be major challenges for carbohydrate chemists. The book starts with a review of Gérard Descotes contribution to the field as a pioneer of French modern carbohydrate chemistry. Green nanocatalytic oxidation of free sugars, photosensitive glycomacrocycles, the application of disaccharides in supramolecular chemistry, recent advances in the radiation chemistry of polysaccharides, and the cell wall pectic rhamnogalacturonan II, an enigma in plant glycobiology are just some of the diverse topics presented in Volume 45. This set of reports will certainly benefit any researcher who wishes to learn about the latest developments in the carbohydrate field.

Book excerpt: Besides causing direct damage associated with blood feeding and in some cases through the excretion of toxins with their saliva, the main relevance of ticks lies in the wide variety of pathogens that they can transmit, including viruses, bacteria, protozoa and helminths. Owing to socioeconomic and environmental changes, tick distribution is changing with incursions of ticks and tick-borne diseases occurring in different regions of the world when the widespread deployment of chemical acaricides and repellents has led to the selection of resistance in multiple populations of ticks. New approaches that are environmentally sustainable and that provide broad protection against current and future tick-borne pathogen (TBP) are thus urgently needed. Such development, however, requires improved understanding of factors resulting in vector competence and tick-host-pathogen interactions. This Research Topic provides an overview of known molecular tick-host-pathogen interactions for a number of TBPs and highlights how this knowledge can contribute to novel control and prevention strategies for tick-borne diseases.
