Many of these toxicants are exogenous compounds that possess the ability to mimic steroid hormones or inhibit normal endocrine functions, which are known as endocrine disrupting compounds (EDCs). The Law laboratory previously obtained mRNA expression data indicating that exposure to aquatic effluents from a combined news/kraft pulp and paper mill changed the expression of genes associated with EDC exposure in the livers of fathead minnows (Pimephales promelas). Societal desire to reduce the use of animals in toxicity testing has encouraged the development and use of in vitro systems, including vertebrate cell models. In this study, the gene expression of the rainbow trout (Oncorhynchus mykiss) liver cell line RTL-W1 and the fathead minnow liver cell line FHM-L was examined following effluent exposure. In the RTL-W1 and FHM-L cell lines, 24 h exposure to effluents was sufficient for assessing changes in gene expression, with maximum changes observed between 4 and 6 h after exposure.

One aspect of fish immunity, that of endogenously produced modulating factors, has recently received a great deal of attention. Understanding the functions and roles of endogenous factors that regulate fish immunity is paramount to expanding the fields of fish immunology and vaccinology. It is know that several lymphoid cell derived factors are detectable in in vitro cell culture systems and exhibit immune modulating effects similar to well studied proteins in mammals. However in comparison, few genes or gene products involved in the modulation of the trout immune responses have been isolated, cloned and characterized. The studies described herein were designed to isolate specific genes from rainbow trout (0ncorhynchus mykiss) and characterize their involvement in the modulation and regulation of the trout immune system. Two distinct genes were isolated cloned and sequenced. The first, non-specific cytotoxic cell enhancement factor (NCEF) gene is closely related to a human gene termed "natural killer enhancement factor" (NKEF) which is important in the modulation of human natural killer cell activity. The second gene is closely related to a group of recently characterized mammalian genes involved in the signal transduction of cytokines termed "STATs". The role of these genes and their respective protein products will be examined and discussed. The antigenic structures of the fish proteins (NCEF and STAT5) were examined by western blot and immunohistochemistry. Monoclonal antibodies derived against the respective human proteins were found to cross react with the corresponding trout proteins, demonstrating antigenic relatedness. The monoclonal regents were also used to analyze the expression of these proteins in fish cells of lymphoid and non lymphoid origin. In vitro cell culture analysis was used to determine the effects and roles of NCEF and STAT5 gene products in the trout immune system. The cytolytic and apoptotic killing activities of spleen, head kidney and peripheral blood leukocytes were found to be enhanced by NCEF. Mitogenic stimulation of peripheral blood lymphoid cells resulted in the trout STAT5 protein binding to a know sequences contained with in the promoters of genes transcriptionally activated in response to cytokine exposure.

Book excerpt: Flesh qualities and muscle characteristics were investigated in the diploid (2n) and triploid (3n) rainbow trouts (Oncorhynchus mykiss) under normal production (Experiment trout station Relliehausen, University of Göttingen) and commercial production conditions in 2003-2005 year classes. The diploid and triploid fishes were divided into 5 stages by age and body weight. For all flesh qualities the 170 g until portion size (400 g) rainbow trouts the polyploidy induction did not have a great influence on all performances. Only the final body weight and carcass percentage of the third group (75 weeks old) which the triploid fishes were significantly higher than the diploid fishes (390g vs. 430g and 73 % vs. 74 %, respectively). The others qualities, the diploid fishes had better than the triploid fishes such as: a redness value of fillets, water holding capacity and water content in muscle. In the maturing rainbow trouts (95 weeks and 6-7 years old), the parameters were significantly higher in the triploid fishes than in the diploid fishes. The final body weight of the biggest group in the 2n and 3n fishes were 4,200g and 5,900g respectively. Both 95 weeks and 6-7 years old fishes, the redness values were higher in the triploid fishes than in the diploid fishes (12.4 vs. 6.02 and 18.0 vs. 13.8). Also we found better results of WHC, cooking yield and tenderness in the triploid compared to the diploid fillets. As for the pH and EC values from 5 min to 5 hr. after slaughter, there were no differences observed between 2n and 3n. The results did not indicate PSE conditions of the meat. With respect for muscle characteristics significant differences of ploidy fishes were found. In general, white and intermediate muscle fibers areas were bigger in triploid than in diploid fishes. These results were found both in the dorsal and pelvic fins samples. As for the red muscle fibers no clear effect of polyploidy were observed.
