Book excerpt: Knowledge of the mechanism of action of drugs at cellular, subcellular, or molecular levels is of vital importance not only in giving the basis of inter pretation of the systemic action of drugs but also in improving existing drugs; in designing new forms of drugs; and in giving the basis of therapeutic applications. Classical pharmacology, concerning the action of drugs at integrated levels, does not necessarily give sufficient information as to the mechanism of action of drugs. A variety of sophisticated concepts utilizing the methods of physics, chemistry, biophysics, biochemistry, and physiology must be synthesized to understand the mechanism of action. Only since the last decade, however, have these techniques been fully applied to pharma cological investigations. It is of utmost importance to realize that a new dimension of pharmacological research has indeed emerged as a result of such a multidisciplinary approach; this approach is encompassed in general and cellular pharmacology. Such recent studies of drug actions have led to a number of important findings. Certain chemicals and drugs were found to possess highly specific actions on cellular functions, so that they are widely being used as powerful tools for the study of a variety of physiological and pharmacological prob lems. Our knowledge of the cellular mechanisms of drug action has provided the basis for interpreting the systemic effects of the drugs and insight into the molecular mechanism involved.

Book excerpt: Understanding interaction between drug molecules and targets is a major pharmacological skill. Molecular and Cellular Pharmacology approaches have greatly increased the knowledge of the structure and function of drug targets, such as receptors, enzymes and transport molecules, revealing diversity far greater than had been realised. This textbook provides a comprehensive and accessible introduction to the rapidly expanding field of cellular and molecular pharmacology. The book deals with most commonly used experiments to understand the relationship of proteins and their involvement in a pathological state at molecular and cellular level. This book is off immense use for research scholars dealing with protein estimation and identification.

Book excerpt: This report from the Committee on Military Nutrition Research reviews the history of caffeine usage, the metabolism of caffeine, and its physiological effects. The effects of caffeine on physical performance, cognitive function and alertness, and alleviation of sleep deprivation impairments are discussed in light of recent scientific literature. The impact of caffeine consumption on various aspects of health, including cardiovascular disease, reproduction, bone mineral density, and fluid homeostasis are reviewed. The behavioral effects of caffeine are also discussed, including the effect of caffeine on reaction to stress, withdrawal effects, and detrimental effects of high intakes. The amounts of caffeine found to enhance vigilance and reaction time consistently are reviewed and recommendations are made with respect to amounts of caffeine appropriate for maintaining alertness of military personnel during field operations. Recommendations are also provided on the need for appropriate labeling of caffeine-containing supplements, and education of military personnel on the use of these supplements. A brief review of some alternatives to caffeine is also provided.

Available in PDF, EPUB and Kindle. Book excerpt: Receptors and Cellular Pharmacology is a collection of the proceedings of the Sixth International Congress of Pharmacology held in Helsinki, Finland, on July 20-25, 1975 and organized by the Finnish Pharmacological Society. The papers explore developments relating to receptors and cellular pharmacology and address a wide range of topics such as the hydrophobic properties of the neuromuscular blocking agents and the use of toxins as tools in receptor studies. Comprised of 21 chapters, this volume begins with an introduction to histamine receptors, their pharmacological taxonomy and classification, their presence in the gastric mucosa, and their applications in medicine. The discussion then turns to pharmacological investigations on cells in culture; the acetylcholine receptor of intact and cultured chicken retina cells; actions of neurotoxins on the acetylcholine receptor; and the isolation and characterization of membrane receptors. Subsequent chapters deal with cyclic nucleotides as mediators of drug action, with particular reference to the mechanism of the relaxation-promoting effects of epinephrine on the mammalian heart. This book should be of interest to pharmacologists, internists, psychiatrists, neurologists, and anesthesiologists.

Book excerpt: This book, with its 16 chapters, documents the present state of knowledge of the adenosine A receptor. It covers a wide range of information, including data from 3 studies of theoretical, molecular and cellular pharmacology, signal transduction, integrative physiology, new drug discoveries and clinical applications. It fills an important gap in the literature since no alternative source of such information is currently available. Although the A receptor is increasingly being recognized for 3 its increasing number of biological roles throughout the body and many A receptor 3 ligands have proven useful in elucidating peripheral and central pathologies, many issues remain unresolved. Moreover, research activity in this field continues to grow exponentially, resulting in a constant flow of new information. The chapters in this book cover both basic science and the relevant applications and provide an authoritative account of the current status of the field. They have enabled my goal as editor to make “A Adenosine Receptors from Cell Biology to Pharmacology and 3 Therapeutics” an up to date, scientifically excellent, reference source, attractive to basic and clinical scientists alike, a reality. Detailed understanding of the physico-chemical aspects and molecular biology of the A receptor provides a solid basis for its future development as a target for 3 adenosine-based pharmacotherapies (Chapters 2 and 3).
