In particular, cellular biology is, besides population dynamics, a field where tech niques of mathematical modeling are widely used. This is reflected by the large number of journal articles and congress proceedings published every year on the dynamics of complex cellular processes. This applies, among others, to metabolic control analysis, where the number of articles on theoretical fundamentals and experimental applications has increased for about 15 years. Surprisingly, mono graphs and textbooks dealing with the modeling of metabolic systems are still exceptionally rare. We think that now time is ripe to fill this gap. This monograph covers various aspects of the mathematical description of enzymatic systems, such as stoichiometric analysis, enzyme kinetics, dynamical simulation, metabolic control analysis, and evolutionary optimization. We believe that, at present, these are the main approaches by which metabolic systems can be analyzed in mathematical terms. Although stoichiometric analysis and enzyme kinetics are classical fields tracing back to the beginning of our century, there are intriguing recent developments such as detection of elementary biochemical syn thesis routes and rate laws for the situation of metabolic channeling, which we have considered worth being included. Evolutionary optimization of metabolic systems is a rather new field with promising prospects. Its goal is to elucidate the structure and functions of these systems from an evolutionary viewpoint.

Book excerpt: Decades of research on the tumor suppressor p53 have revealed that it plays a significant role as a "guardian of the genome," protecting cells against genotoxic stress. In recent years, p53 research has begun to move into the clinic in attempts to understand how p53 is frequently inactivated in-and sometimes even promotes-human cancer. Written and edited by experts in the field, this collection from Cold Spring Harbor Perspectives in Medicine covers the rapid progress that has recently been made in basic and clinical research on p53. The contributors review new observations about its basic biology, providing updates on the functions of its isoforms and domains, the myriad stresses and signals that trigger its activation or repression, and its downstream effects on genome stability and the cell cycle that enforce tumor suppression in different cell and tissue types. They also discuss how p53 dysfunction contributes to cancer, exploring the various inherited and somatic mutations in the human TP53 gene, the impact of mutant p53 proteins on tumorigenesis, and the prognostic value and clinical outcomes of these mutations. Drugs that are being developed to respond to tumors harboring aberrant p53 are also described. This book is therefore essential reading for all cancer biologists, cell and molecular biologists, and pharmacologists concerned with the treatment of this disease.

Book excerpt: Cell Cycle Regulation describes the interaction of the nuclear genome, the cytoplasmic pools, the organelles, the cell surface, and the extracellular environment that govern the cell cycle regulation. Comprised of 12 chapters, this book includes cell cycle regulation around nuclear chromatin modulation and some aspects of chromatin modification and its effects on gene expression. The opening chapters describe the macromolecular structure of chromatin subunits and the types and kinds of postsynthetic modifications occurring on histones, such as acetylation, methylation, and phosphorylation. The subsequent chapter deals extensively on histone phosphorylation, especially histone H1, H1M, H2A, and H3, during the cell cycle. Another chapter describes a selective histone leakage from nuclei during isolation accounting for the role of histone acetylation and phosphorylation in gene expression. This book goes on examining the assembly of microtubules and structural analysis on the regulatory role of calcium into a pattern for mitosis regulation. Other chapters discuss the methods used to measure intracellular pH changes as a function of the cell cycle of Physarum and the quantitative and qualitative changes taking place during the various phases of the cell cycle. The use of mammalian cell fusion to study cell cycle regulation and the protein synthesis regulation during the cell cycle in Chlamydomonas reinhardi are then discussed. The final chapters focus on the regulation of expression of an inducible structural gene during the cell cycle of the green alga Chlorella. The chapters provide evidence for a model of positive and negative oscillatory control of inducible gene expression. An analysis of the expression of cytoplasmic genes as a function of the cell cycle using pedigrees of a large number of individual yeast cells is also included. This book will appeal to a wide variety of life scientists and to molecular, cellular, and developmental biologists.

Book excerpt: Aspects of Cell Regulation deals with the various aspects of cellular control mechanisms. The book begins with a discussion of the role of hormones in the control of cell differentiation and biochemical control over the life of an organism. This is followed by separate chapters on an intracellular control problem, namely, the proper reproduction of cellular organelles and their apportionment between daughter cells at cell mitosis; cell mutants with different temporal order for macromolecular synthesis; and the control of a plant enzyme that converts triglycerides into carbohydrate during the early stages of postgerminative growth within seed cotyledon. Subsequent chapters deal with the possible role of mitochondria in changing the control mechanism of cells to that associated with carcinogenesis; the possibility of transforming genes being detectable in the genome after the transformation to neoplastic cell; the correlation between viruses and tumor cells; and whether viruses can originate from cells rather than coming before cells.

Available in PDF, EPUB and Kindle. Book excerpt: The "Progress in Cell Cycle Research" series is dedicated to serve as a collection of reviews on various aspects of the cell division cycle, with special emphasis on less studied aspects. We hope this series will continue to be helpful to students, graduates and researchers interested in the cell cycle area and related fields. We hope that reading of these chapters will constitute a "point of entry" into specific aspects of this vast and fast moving field of research. As PCCR4 is being printed several other books on the cell cycle have appeared (ref. 1-3) which should complement our series. This fourth volume of PCCR starts with a review on RAS pathways and how they impinge on the cell cycle (chapter 1). In chapter 2, an overview is presented on the links between cell anchorage -cytoskeleton and cell cycle progression. A model of the Gl control in mammalian cells is provided in chapter 3. The role of histone acetylation and cell cycle contriol is described in chapter 4. Then follow a few reviews dedicated to specific cell cycle regulators: the 14-3-3 protein (chapter 5), the cdc7/Dbf4 protein kinase (chapter 6), the two products of the pI6/CDKN2A locus and their link with Rb and p53 (chapter 7), the Ph085 cyclin-dependent kinases in yeast (chapter 9), the cdc25 phophatase (chapter 10), RCCI and ran (chapter 13). The intriguing phosphorylation dependent prolyl-isomerization process and its function in cell cycle regulation are reviewed in chapter 8.

As this meeting attracted many investigators involved in cyclic nucleotides and calcium role in intracellular regulation, it was thought that this opportunity could be used to organize, prior to the Congress, an in-depth introductory course on the subject. This was carried out as a NATO Advanced Study Institute. The participants included Ph. D. students and M. D. s engaged in a research training, but also fully trained and well known researchers who wanted a refresher course on the whole subject. During the course, most of the participants and lecturers asked to be provided with a text summarizing the basic lectures of the course. This book was therefore conceived as a basic textbook on the regulation and action of intracellular signal molecules, concentrating mainly on cyclic nucleotides and calcium. It was deliberately kept at a basic level. We would therefore be happy if it could be used as an introduction for interested M. D. s or Ph. D. s working in other fields or entering this field and as a general refresher for researchers interested in the subject. For this reason, very general schemes have been asked of the authors, along with reading lists of available reviews rather than extensive bibliographies. The editors should like to thank the NATO Scientific Affairs Committee for having supported the course and Mrs. Gh. Wilmes who prepared the manuscripts. S. SWILLENS J. E.

Book excerpt: "Cell signaling, which is also often referred to as signal transduction or, in more specialized cases, transmembrane signaling, is the process by which cells communicate with their environment and respond temporally to external cues that they sense there. All cells have the capacity to achieve this to some degree, albeit with a wide variation in purpose, mechanism, and response. At the same time, there is a remarkable degree of similarity over quite a range of species, particularly in the eukaryotic kingdom, and comparative physiology has been a useful tool in the development of this field. The central importance of this general phenomenon (sensing of external stimuli by cells) has been appreciated for a long time, but it has truly become a dominant part of cell and molecular biology research in the past three decades, in part because a description of the dynamic responses of cells to external stimuli is, in essence, a description of the life process itself. This approach lies at the core of the developing fields of proteomics and metabolomics, and its importance to human and animal health is already plainly evident"--Provided by publisher.
