Book excerpt: Metabolic enzymes have been hypothesized to compartmentalize in cells to facilitate in metabolic control. Indeed, there are several lines of evidence from across species which demonstrate the compartmentalization of single or multiple metabolic enzymes in cells. In human cells, enzymes in de novo purine biosynthesis or glycolysis and gluconeogenesis have the propensity to compartmentalize in the cytoplasm. The formation of these multi-enzyme compartments has been hypothesized to stabilize metabolic intermediates, facilitate in substrate channeling, and alter metabolic flux. Therefore, we propose that the compartmentalization of metabolic enzymes provides a means for the cell to regulate metabolism.

Book excerpt: THIS BOOK HAS BEEN WRITTEN BECAUSE WE FEEL THAT THERE IS A NEED FOR AN up-to-date compact book on cell organelles that transmits the excitement and challenge of modern subcellular biology. We hope that the book will be interesting and useful to students of the biological sciences and medicine, and to those in the teaching professions who do not have ready access to research papers. Since space is at a premium, we have denied ourselves the luxury of a philosophical discussion of the problems of defining organelles. Rather we have chosen to include all those intracellular structures which have limiting membranes and definable compartments. The separate chapters consider nuclei, plastids, mitochondria, microbodies, endoplasmic and sarcoplasmic reticulum; Golgi bodies, lysosomes and various secretory vesicles, including chromaffin granules and synaptic vesicles. Nucleoli, ribosomes, and centrioles are included in the chapter on nuclei. New and exciting informatioIi about all these structures has emerged in recent years-for example; the nUcleosome, interrupted genes, signal sequences on proteins destined. for the bioenergetic organelles, mapping and sequencing of organelle genes, and consolidation of chemiosmosis as a unifying principle in energy transduction. We have outlined as many of these developments as possible and pointed out some areas of controversy. The literature on subcellular biology is so extensive that it would have been easier to have written a separate book on each organelle.

In three distinct parts, latest results from basic research on stable enzymes are explained and brought into context with possible industrial applications. Downstream processing technology as well as biocatalytic and biotechnological production processes from global players display the enormous potential of biocatalysts. Application of "extreme" reaction conditions (i.e. unconventional, such as high temperature, pressure, and pH value) - biocatalysts are normally used within a well defined process window - leads to novel synthetic effects. Both novel enzyme systems and the synthetic routes in which they can be applied are made accessible to the reader. In addition, the complementary innovative process technology under unconventional conditions is highlighted by latest examples from biotech industry.

Book excerpt: Mitochondria are sometimes called the powerhouses of eukaryotic cells, because mitochondria are the site of ATP synthesis in the cell. ATP is the universal energy currency, it provides the power that runs all other life processes. Humans need oxygen to survive because of ATP synthesis in mitochondria. The sugars from our diet are converted to carbon dioxide in mitochondria in a process that requires oxygen. Just like a fire needs oxygen to burn, our mitochondria need oxygen to make ATP. From textbooks and popular literature one can easily get the impression that all mitochondria require oxygen. But that is not the case. There are many groups of organismsm known that make ATP in mitochondria without the help of oxygen. They have preserved biochemical relicts from the early evolution of eukaryotic cells, which took place during times in Earth history when there was hardly any oxygen avaiable, certainly not enough to breathe. How the anaerobic forms of mitochondria work, in which organisms they occur, and how the eukaryotic anaerobes that possess them fit into the larger picture of rising atmospheric oxygen during Earth history are the topic of this book.

Book excerpt: This book provides the first comprehensive coverage of the quickly evolving research field of membrane contact sites (MCS). A total of 16 chapters explain their organization and role and unveil the significance of MCS for various diseases. MCS, the intracellular structures where organellar membranes come in close contact with one another, mediate the exchange of proteins, lipids, and ions. Via these functions, MCS are critical for the survival and the growth of the cell. Owing to that central role in the functioning of cells, MCS dysfunctions lead to important defects of human physiology, influence viral and bacterial infection, and cause disease such as inflammation, type II diabetes, neurodegenerative disorders, and cancer. To approach such a multifaceted topic, this volume assembles a series of chapters dealing with the full array of research about MCS and their respective roles for diseases. Most chapters also introduce the history and the state of the art of MCS research, which will initiate discussion points for the respective types of MCS for years to come. This work will appeal to all cell biologists as well as researchers on diseases that are impacted by MCS dysfunction. Additionally, it will stimulate graduate students and postdocs who will energize, drive, and develop the research field in the near future.

Available in PDF, EPUB and Kindle. Book excerpt: Organized Multienzyme Systems: Catalytic Properties describes the kinetic and catalytic properties of organized enzyme systems. This book is composed of nine chapters that specifically cover both immobilized and naturally occurring systems. The first two chapters examine the nature and function of enzyme organization in the mitochondrion, as well as the structural/functional coupling of the components in energy-transducing membrane systems. These topics are followed by discussions on ""dynamic compartmentation"" in soluble multienzyme systems; the allosteric enzyme systems; and allosterism in reversibly adsorptive enzyme systems. Other chapters explore model studies with specific immobilized multienzyme sequences, as regards the analysis of microenvironmental effects, and the mathematical exposition on the kinetic analysis of multienzyme systems in homogeneous solution. The last chapters present some theoretical and experimental studies on the behavior of immobilized systems. These chapters also provide a speculative integrative view of the kind of functional coherence that may be operative in organized states in vivo. This book is of great value to cell biologists, biochemists, and enzyme scientists and researchers.

Book excerpt: Metabolic and Cellular Engineering (MCE) is more than an exciting scientific enterprise. It has become the cornerstone for coping with the challenges ahead of mankind. Continuous developments, new concepts, and technological innovations will enable us to deal with emerging challenges, and solve problems once thought impossible ten years ago. Challenges in MCE are broad- from unraveling fundamental aspects of cellular function to meeting unsatiated energy and food demands that are rising in parallel with population growth. In charting the progress of MCE during the last decade, we could not help but feel in awe of the enormous strides of progress made from the nascent Metabolic Engineering to the Systems Bioengineering of today. The burgeoning availability of genomic sequences from diverse species has been spectacular. It has become the engine that drives the genetic means for the modification of existing organisms and the generation of synthetic, man-made ones. From the initial attempts at purposeful genetic modification of a cell for the production of valuable compounds, we have now moved on to changing microbes genetically or metabolically. The arsenal of experimental and theoretical tools available for Metabolic and Cellular Engineering has expanded enormously, driven by the re-emergence of Physiology as Systems Biology. The revival of the concept of networks fueled by new developments has become central to Systems Biology. Networks represent an integrative vision of how processes of disparate nature relate to each other, and as such is becoming a key analytical and conceptual tool for MCE. This book reflects and addresses all these ongoing changes while providing the essential conceptual and analytical tools needed to understand and work in the MCE research field.

Book excerpt: Microcompartmentation refers to nonhomogeneous distribution of solutes in compartments of cells or associated structures without intervening membranal barriers. Such variations in concentration of ions, metabolites, intracellular messengers, and nutrients can introduce significant heterogeneity in subcellular function and regulation. The current focus on biological examples of microcompartmentation provides ample evidence for its importance and the role of physical structure in determining local chemical environments. The examples present in the different chapters include microcompartmentation of Ca2+,H+,ATP. ADP, O2, glycolytic intermediates, fatty acids, amino acids, and nucleic acid precursors. In reviewing these systems, the authors provide a useful resource for experimental approaches to study microcompartmentation and provide the basis for future studies of its role in regulation of cell functions.
