The three structures include the unliganded 1F7 Fab' fragment, the Fab' complex with a transition state analog, and the Fab' complex with the reaction product (prephenate). The structural data suggest that the antibody stabilizes the same conformationally restricted, polarized transition state that occurs in the uncatalyzed reaction. The overall shape and charge complementarity between the combining site and the transition state analog appears to dictate preferential binding of the correct substrate enantiomer [(-)-chorismate] in a conformation appropriate for reaction. Comparison with the structures of bacterial chorismate mutase enzymes indicates an overall similarity between the catalytic mechanisms employed by the antibody and the enzymes. The smaller number of interactions available for constraining the conformation of the flexible substrate, and the lack of multiple electrostatic interactions that might stabilize charge separation in the dipolar transition state, are likely to account for the observed 104-fold lower activity of the antibody relative to the natural enzymes that catalyze this reaction. The structural features of the antibody correspond closely to the chemical nature of the transition state analog, which was intended primarily to elicit a template-like binding site that would favor the geometry of the transition state.
