Book excerpt: Photoacoustic tomography (PAT), as well as thermoacoustic tomography (TAT), utilize electromagnetic radiation in its visible, near infrared, microwave, and radiofrequency forms, respectively, to induce acoustic waves in biological tissues for imaging purposes. Combining the advantages of both the high image contrast that results from electromagnetic absorption and the high resolution of ultrasound imaging, these new imaging modalities could be the next successful imaging techniques in biomedical applications. Basic research on PAT and TAT, and the relevant physics, is presented in Chapter I. In Chapter II, we investigate the imaging mechanisms of TAT in terms of signal generation, propagation and detection. We present a theoretical analysis as well as simulations of such imaging characteristics as contrast and resolution, accompanied by experimental results from phantom and tissue samples. In Chapter III, we discuss the further application of TAT to the imaging of biological tissues. The microwave absorption difference in normal and cancerous breast tissues, as well as its influence on thermoacoustic wave generation and the resulting transducer response, is investigated over a wide range of electromagnetic frequencies and depths of tumor locations. In Chapter IV, we describe the mechanism of PAT and the algorithm used for image reconstruction. Because of the broad bandwidth of the laser-induced ultrasonic waves and the limited bandwidth of the single transducer, multiple ultrasonic transducers, each with a different central frequency, are employed for simultaneous detection. Chapter V further demonstrates PAT's ability to image vascular structures in biological tissue based on blood's strong light absorption capability. The photoacoustic images of rat brain tumors in this study clearly reveal the angiogenesis that is associated with tumors. In ChapterVI, we report on further developing PAT to image deeply embedded optical heterogeneity in biological tissues. The improved imaging ability is attributed to better penetration by NIR light, the use of the optical contrast agent ICG (indocyanine green) and a new detection scheme of a circular scanning configuration. Deep penetrating PAT, which is based on a tissue's intrinsic contrast using laser light of 532um [mu] green light and1.06 um [mu] near infrared light, is also presented.

It combines the good contrast of electromagnetic and good resolution of ultrasound imaging. The mathematical model of TAT is the observability problem for the wave equation: one observes the data on a hyper-surface and reconstructs the initial perturbation. In this dissertation, we consider several mathematical problems of TAT. The first problem is the inversion formulas. We provide a family of closed form inversion formulas to reconstruct the initial perturbation from the observed data. The second problem is the range description. We present the range description of the spherical mean Radon transform, which is an important transform in TAT. The next problem is the stability analysis for TAT. We prove that the reconstruction of the initial perturbation from observed data is not Hölder stable if some observability condition is violated. The last problem is the speed determination. The question is whether the observed data uniquely determines the ultrasound speed and initial perturbation. We provide some initial results on this issue. They include the unique determination of the unknown constant speed, a weak local uniqueness, a characterization of the non-uniqueness, and a characterization of the kernel of the linearized operator.

TAT is an emerging medical imaging technique that combines the merits of high contrast due to electromagnetic or laser stimulation and high resolution offered by thermal acoustic imaging. Image reconstruction via proper signal processing techniques is one of the key problems in TAT. The current image reconstruction methods used for TAT, such as the widely used Delay-and-Sum (DAS) approach, are data-independent and suffer from low resolution, high sidelobe levels, and poor interference rejection capabilities. Also, the amplitude and phase distortion problems of thermoacoustic signals due to propagation in the non-homogeneous media are not properly handled by the existing methods. The data-adaptive ART can have much better resolution and much better interference rejection capabilities than their data-independent counterparts. By allowing certain uncertainties in the parameters, ART can be used to mitigate the amplitude and phase distortion problems encountered in TAT. Using both simulated and experimentally measured data, we demonstrate the excellent performance of ART: high resolution, low sidelobe level, and much better interference rejection capability.

The underlying physics involved in this technique naturally divides its theoretical analysis into two different problems corresponding to each type of waves. One of them is the inverse thermoacoustic tomography problem which studies the propagation of ultrasound across biological tissues and aims to obtain internal information related with the initial source of the acoustic waves, from data acquired on transducers placed outside the body of interest. A natural difficulty in the analysis of ultrasound propagation is the attenuation effect caused by different tissues, in particular in the case of biological bodies. Its study is of course of great interest for real applications of this technology and it has received considerable attention in the last few years. In this thesis, I review some previous results for the thermoacoustic problem in the absence of acoustic dissipation, and in the second part, I analyze the inverse problem of thermoacoustic tomography in the presence of two types of attenuation models, local and nonlocal in time, and provide reconstruction formulas for both cases under some assumptions on the geometry of the problem. New results on uniqueness and stability of the inverse problem in the latter case are also established.

Microwave-induced thermoacoustic tomography was first used to verify the feasibility of noninvasively detecting the coagulated damage based on different dielectric properties between normal tissue and lesion treated with high intensity focused ultrasound. Good image contrasts were obtained for the lesions. A comparison of the size of the lesion measured with microwave-induced thermoacoustic tomography and the size measured by a gross pathologic photograph was presented to verify the effectiveness the proposed method. Clinical data for breast tumors were also collected to verify the feasibility of using microwave-induced thermoacoustic tomography in breast cancer imaging. Next, the effects of acoustic heterogeneities on microwave-induced thermoacoustic tomography in weakly refractive medium were investigated. A correction method based on ultrasonic transmission tomography was proposed to correct for the image distortion. Numerical simulations and phantom experiments verify the effectiveness of this correction method. The compensation is important for obtaining higher resolution images of small tumors in acoustically heterogeneous tissues. Finally, the effects of the highly refractive skull on transcranial brain imaging were studied. A numerical method, which considered wave reflection and refraction at the skull surfaces, was proposed to compensate for the image degradation. The results obtained with the proposed model were compared with the results without considering the skull-induced distortion to evaluate the skull-induced effects on the image reconstruction. It was demonstrated by numerical simulations and phantom experiments that the image quality could be improved by incorporating the skull shape and acoustic properties into image reconstruction. This compensation method is important when the thickness of skull cannot be neglected in transcranial brain imaging.
