Estos tumores presentan un importante impacto en la calidad de vida de los pacientes afectados y un mal pronóstico, sobre todo en los tumores de grado alto. La monitorización no invasiva de los tumores cerebrales es muy importante para el diagnóstico y seguimiento de la enfermedad y se hace sobre todo por técnicas de Imagen por Resonancia Magnética (IRM), la cual aporta información morfológica. La Espectroscopía de Resonancia Magnética (ERM), tanto en su modalidad voxel único como imagen espectroscópica (IERM) son técnicas complementarias de gran utilidad, por aportar información del entorno metabólico, que no puede conseguirse mediante la IRM. Las dificultades éticas para desarrollar mejoras en los protocolos de IRM/ERM, así como para la prueba de nuevos agentes de contraste en pacientes humanos han llevado al desarrollo de los modelos murinos de cáncer cerebral. Los modelos más utilizados son los creados por inyección estereotáxica, aunque también se han desarrollado modelos transgénicos (GEM, genetically engineered mice) que desarrollan tumores cerebrales espontáneamente y reproducen las características infiltrativas de tumores humanos. En esta tesis, se llevó a cabo en el capítulo 2 un amplio estudio de caracterización de diferentes colonias GEM. Se comprobó la baja penetrancia de tumores en estos modelos (16,7% en la colonia que mejores resultados produjo), lo que plantea dificultades para realizar estudios con grupos homogéneos. Este hecho, sumado a la escasez de modelos preclínicos de tumores cerebrales de bajo grado actualmente disponibles, motivaron a su vez el estudio que se llevó a cabo en el capítulo 3 de ésta tesis. Dicho capítulo trata del establecimiento de una línea transplantable de tumor cerebral murino de bajo grado a partir de un tumor desarrollado por un animal de la colonia GEM. El objetivo de éste capítulo consistía en generar un tumor inducible mediante inyección estereotáctica intracraneal de gliosferas generadas en cultivo a partir de la disgregación del tumor glial de origen. Se llevó a cabo una puesta a punto con tumores murinos de tipo GL261 y en un siguiente paso, se continúo con el establecimiento y caracterización de la línea transplantable a partir de un oligodendroglioma anaplásico desarrollado por un especimen de la colonia GEM. Esta línea transplantable ha sido ampliamente caracterizada de manera no invasiva mediante IRM/ERM y también mediante estudios de imagen espectroscópica con perturbación del patrón espectral (PE-IERM).Se obtuvieron resultados prometedores, con un 60% de penetrancia del fenotipo tumoral, y manteniendo en la mayoria (1 de 12 analizados por histopatologia) de los casos la misma patología del tumor de origen. Paralelamente, en el último capítulo se realizaron estudios con nuevos agentes de contraste (AC) que actualmente se están desarrollando para IRM con potencial interés para tumores cerebrales y que permitirán superponer información funcional a la información anatómica. Para ello, se ha desarrollado un método de análisis postmortem de estos AC. Dicho método permite no sólo reducir el número de animales utilizados en la experimentación, sino también reducir en 800 veces la cantidad de agente necesaria para estudios preliminares en IRM preclínica. Nuestros resultados preliminares apuntan a que este método es más preciso que los análisis de relajatividad in vitro, debido a una mejor simulación del entorno in vivo y de la posible interacción del AC con tejidos, moléculas y receptores presentes en el entorno de utilización. Los resultados obtenidos con el mejor agente analizado (GNP(E), con un incremento de contraste relativo, IRC, 330% en comparación con contralateral) se han corroborado en un análisis de IRM in vivo en el modelo preclínico de tumor glial GL261 (IRC 122% en comparación con contralateral), siendo a su vez sensiblemente más eficiente que el AC comercial de tipo comparable actualmente disponible y en uso en clínica.
