Book excerpt: Angiogenesis is a highly complex phenomenon where new blood vessels are formed for the supply of oxygen and nutrients in different organs of the body. It plays a critical role in both physiological processes such as growth and development as well as pathological processes including cancer and different types of tumors. Angiogenesis is also essential for the regeneration and survival of cells in several disease conditions such as ischemic heart disease (myocardial infarction), atherosclerosis, brain injury (stroke) and diabetes. Since the mechanisms of angiogenesis are organ specific and differ among various diseases, it is proposed to devote one section of this book to the development of angiogenesis in some selected diseases such as cancer, ischemic heart disease, atherosclerosis, diabetes and stroke. It is pointed out that extensive research work in this regard has been carried out in the area of cancer and heart disease, whereas relatively less attention has been paid to studying angiogenesis in other disease conditions.

Individual chapters introduce the anatomy of the coronary blood vessels and cardiac development, while others consider current imaging modalities utilized for ischemic heart disease, including stress echo, nuclear diagnostic tests, non-invasive coronary artery imaging, and coronary angiography. Imaging chapters provide key clinical information on techniques and indications, and include examples of both normal and abnormal patterns. The principle thrust of the book concerns coronary atherosclerosis, the pathology of which is presented in conjunction with the results of anatomic, non-invasive imaging and angiographic studies. Related chapters cover atherogenesis, presenting new insights into the pathophysiology of the vulnerable plaque, the role of progenitor cells in vascular injury, inflammation and atherogenesis, and the genomics of vascular remodeling. Additional topics covered include angina pectoris, acute coronary syndromes, healed myocardial infarction and congestive heart failure, catheter-based and surgical revascularization, and surgical treatment of myocardial infarction and its sequelae. With contributions from a diverse group of internationally-known physicians with broad experience in the diagnosis and treatment of coronary heart disease, this book will be a valuable resource for practitioners in clinical cardiology, thoracic surgery, pathology, and cardiovascular molecular research, as well as for students in training.

Book excerpt: It is long known that many cells can shed extracellular vesicles, small membrane-enclosed cell fragments. Although the existence of extracellular vesicles has been recognized for many years, researchers are only beginning to understand their physiologic significance. Several recent studies have demonstrated that extracellular vesicles released from cells serve as a mode of cellular communication. They can carry diverse molecular payload (e.g. nucleic acids, bioactive lipids and proteins) to distal organs and recipient cells. Extracellular vesicles can be classified into three major groups: exosomes, microvesicles, and apoptotic bodies. All these types of extracellular vesicles can be found in a variety of biologic specimen and their numbers, distribution and composition may serve as biomarkers for various disorders, including cardiovascular disease. Although extracellular vesicle-mediated processes are currently best characterized in the fields of cancer biology and neurobiology, evidence is accumulating that extracellular vesicles play a key role in the pathophysiology of diabetes, thrombosis, inflammation and cardiovascular calcification. In this Research Topic, we invited review and methodological articles that advance our understanding of extracellular vesicle-related processes in vascular biology.

Available in PDF, EPUB and Kindle. Book excerpt: The microcirculation is highly responsive to, and a vital participant in, the inflammatory response. All segments of the microvasculature (arterioles, capillaries, and venules) exhibit characteristic phenotypic changes during inflammation that appear to be directed toward enhancing the delivery of inflammatory cells to the injured/infected tissue, isolating the region from healthy tissue and the systemic circulation, and setting the stage for tissue repair and regeneration. The best characterized responses of the microcirculation to inflammation include impaired vasomotor function, reduced capillary perfusion, adhesion of leukocytes and platelets, activation of the coagulation cascade, and enhanced thrombosis, increased vascular permeability, and an increase in the rate of proliferation of blood and lymphatic vessels. A variety of cells that normally circulate in blood (leukocytes, platelets) or reside within the vessel wall (endothelial cells, pericytes) or in the perivascular space (mast cells, macrophages) are activated in response to inflammation. The activation products and chemical mediators released from these cells act through different well-characterized signaling pathways to induce the phenotypic changes in microvessel function that accompany inflammation. Drugs that target a specific microvascular response to inflammation, such as leukocyte-endothelial cell adhesion or angiogenesis, have shown promise in both the preclinical and clinical studies of inflammatory disease. Future research efforts in this area will likely identify new avenues for therapeutic intervention in inflammation. New techniques and discoveries continuously give rise to observations that result in the de?nition of new research objectives. In the past, - search departments were clearly demarcated. Nowadays, technology that is shared by all lines of research stimulates convergence of research interests. This also - plies to cardiovascular research. Vascular occlusive disease is now core business for researchers employed by cardiology, vascular surgery, vascular medicine, - diology, cell biology, chemistry, physiology, and many other areas. Knowledge on actual research development is shared by researchers with different skills. It is sometimes dif?cult to acquire expertise when a researcher feels his experimental work could be improved by introducing a new research technique. In this book, the investigator will ?nd an overview of recent developments that are relevant for research in general but cardiovascular research in particular. Genomics, p- teomics, microarray, RNAi, stem cells, and progenitor cells are just some phrases that have become increasingly prevalent in literature in the last few years and that are recognized by many, but are fully understood by few. In this book, experts share the most appreciated new developments and techniques in cardiovascular research. We hope that this book will help the reader who is working in the ?eld of cardiovascular research to understand and critically appreciate current research, and that it will help improve the quality of experimental work.
