Book excerpt: Ambient mass spectrometry (MS) is a new and growing sub-field in MS which has opened new research avenues, particularly for applications relating to the analysis of solid samples. Results on the implementation and application of ambient MS techniques including: desorption electrospray ionization (DESI) and direct analysis in real time (DART) indicated that these techniques could serve as complementary tools for the rapid qualitative screening of pharmaceuticals, allowing up to two orders of magnitude improvement in throughput compared to traditional methods such as liquid chromatography MS. The selectivity of DESI could be enhanced by performing the experiment in the reactive mode. In this mode, complexation reactions between reagents added to the spray solvent and analytes on the sample surface resulted in analyte stabilization, inhibiting fragmentation. They also resulted in a concomitant enhancement in the analyte surface activity, facilitating their evaporation from secondary droplets culminating in an improvement in sensitivity. Also for drug tablets analysis, the analyte signal dependency on DESI geometrical set-up variables could be mitigated following the careful and controlled addition of an isotopically labeled internal standard (IS) to the sample or by spraying samples with a pair of reagents with different affinities for the analyte. Either of these approaches resulted in an analyte-to-IS signal ratio (in the former) or an analyte complex ratio (in the later), which was largely independent of DESI experimental variables allowing quantitative analysis using this technique. DESI MS was also observed to be a very powerful tool for determining the 2-D distribution of various pharmaceutically important compounds on tablet and tissue surfaces. The ability to map the distribution of molecules of interest by DESI MS has very great implications in drug tablet quality control and in determining the role of chemical signals presented on tissue surfaces. DESI was observed to be limited to ionizing molecules of medium to high polarities without much limitation in terms of mass range, whereas DART was better suited for the analysis of molecules within a broader range of polarities, but within a more limited mass range (up to 800 Da approximately). These limitations were circumvented by implementing a novel multimode ambient ion source, desorption electrospray/metastable-induced ionization (DEMI), which combines various aspects of DESI and DART. Initial experiments with the DEMI ion source demonstrated its ability to enable the simultaneous analysis of molecules within a broader range of polarities and masses than DESI and DART alone.

Book excerpt: Drug chemistry sections of forensic crime laboratories are tasked with analyzing a variety of unknown materials, a subset of which are derived from plants. In comparison to the analysis of small-molecule drugs, these plant matrices are incredibly complex and present unique challenges. When traditional chromatographic approaches are used to interrogate these materials in forensic investigations, large amounts of time, resources, and funds are directed towards the extensive sample preparation steps required. Exacerbating the issue is that the matrix characteristics often result in instrument downtime to accomplish the maintenance required to address ingredients adhering to columns or clogging syringes, which are common problems when testing Cannabis-derived samples. Furthermore, the increased legalization and decriminalization of marijuana in the U.S. has contributed to the rise in popularity of Cannabis products, which will require new testing methods to analyze the myriad of sample types available as the industry continues to evolve. Therefore, the unique capabilities of direct analysis in real time – high-resolution mass spectrometry (DART-HRMS) were investigated for the analysis of plant materials, including Cannabis, as an approach to circumventing many of the sample preparation and other difficulties often encountered. A DART-HRMS triage approach, which requires no sample pretreatment steps, has been developed for the detection of Cannabis-related molecules in a wide variety of sample types, including edibles, beverages, concentrates, topicals, vaporizers, marijuana/hemp flower, and pre-rolls. Additional applications of DART-HRMS for the analysis of Cannabis materials include the following: (1) differentiation of cannabinoids; (2) quantification of cannabinoids in edibles extracts; and (3) distinguishing between hemp and marijuana varieties of C. sativa through statistical analysis of mass spectral data. DART-HRMS was also proven effective for the analysis of plant-based legal-highs, specifically those identified as “plants of concern” by the United Nations Office on Drugs and Crime. Ambient ionization mass spectrometry (AIMS) approaches were developed for the rapid detection and validated quantification of psychoactive small-molecule constituents in complex plant matrices. The methods described would greatly improve forensic science and criminal justice practices in the U.S. for the analysis of complex plant materials, both Cannabis-derived and plant-based legal-highs.

While colorimetric testing can be employed for drug evidence screening, the influx of novel psychoactive substances has significantly decreased the effectiveness of this investigation strategy. One approach to meeting the needs of today's practitioner is coupling flexible, ambient ionization techniques with portable mass spectrometric (MS) instrumentation. Ambient ionization techniques have a rich history of being amendable to the sample at hand. Since their inception in 2005, numerous techniques saturate the field. The main drive for this development is to allow the analysis of wider sample classes with doing as minimal sample preparation required. Herein, the efforts to develop and perform a selection of simplified, rapidly-interchangeable, ambient ionization sources will be discussed, including paper spray ionization (PSI) and paper cone spray ionization (PCSI).First reported by Cha and Kim, PCSI has been shown adept at bulk analyte analysis. Here, a modified PCSI source that allows for real-time extraction and filtration of forensic samples known to hinder screening or compromise hygiene on MS systems is investigated. Also, studies will be included that evaluates an inter-source comparison of ionization methods that have already shown broad utility within the forensic community, PSI and direct analysis in real time (DART), in order to discern analytical performance and usability in forensic evidence processing.While portable MS systems have been shown capable of assisting investigative decision-making, this single technology does not fulfill the two-tiered identification guidelines suggested by the Scientific Working Group for the Analysis of Seized Drugs (SWGDRUG) for generating prosecutorial data. By integrating surface-enhanced Raman spectroscopy (SERS) with a portable MS system, a rapid, yet robust, method for drug evidence identification is discussed, while looking at the development and characterization of this instrumental strategy.

Chapter 2 provides discussion on the robustness of DESI-MS for breast cancer tissue section evaluation and receptor status determination in a multi-center study, providing evidence of its potential use to compliment routine histopathological assessment. Book excerpt: Forensic scientists are currently actively evaluating new instrumental techniques to determine their potential application for solving some of the problems that are unique to their field. Liquid chromatography, flameless atomic absorption, electron scanning microscopy, and thermal analysis offer fertile areas for research and development. Mass spectroscopy is by no means a new instrumental technique; its application to analytical organic chemistry has been well documented since the early 1960's [1-4]. Unfortunately, its utilization in the forensic sciences has been minimal up to the present. A major limitation has been the cost and complexity of the instrument. However, with increased fundings becoming available to many laboratories and with the improved engineering and reliability of the latest generation of mass spectrometers, this technique is becoming feasible for many forensic laboratories. Its application to forensic drug identification has been demonstrated in previous publications [5-8].
