Moths and butterflies (Lepidoptera) are the dominant group of silk producers and have some of the greatest diversity of silk uses. Still, very little is known about silk production, especially in terms of understanding how a genotype influences its phenotype. This thesis presents what is known about Lepidoptera silk, and the challenges associated with this research. I show that there are significant knowledge gaps which are unequally distributed across taxonomic scales, but that recent advances in sequencing technology provide ample opportunity to make significant advancements in silk research. Using the Luna moth (Actias luna) as a native study species, this research provides the foundation for characterizing silk production in non-model lepidopterans and in non-pupal silks. Here, I produce a high-quality reference genome assembly and annotation using high fidelity (HiFi) long read sequencing to fully capture challenging areas of the genome associated with silk production. This research will allow for future elucidation of the underlying mechanisms that change how silk is produced throughout larval development in this species, and across the moth and butterfly tree of life.
