Book excerpt: The International Symposium on Prostaglandins and Related Compounds, first held in Vienna 1972, revisited the city after 24 years for the 10TH Symposium. For the many re searchers working in this multi-disciplinary field it was an opportunity to exchange their ex periences and share new data with colleagues from all around the world. This scientific exchange was largely encouraged by the unseasonably cold and rainy weather. For the first time, there was quite a large attendance from the former Communist countries. Eugene Garfield prepared a key note address delivered during the meeting (The Sci entist 1996, 12) reviewing the contribution of the Nobel Laureates U.S. von Euler, l.R. Vane, S.K. Bergstrom, and B.I. Samuelsson, discussing the relevance of the more than 40,000 pa pers in this area published since 1991. Overall, there is still a rapidly growing interest, and in particular a great variety of clinical applications of this family of compounds which were dis cussed in detail during the meeting. Beside the lectures there were 19 workshops covering nearly all the topics of key in terest. All the speakers were invited to prepare a manuscript which has resulted in the volume now in your hands. Special thanks to Dr. Patrick Wong and the new publisher of this series who helped to publish the proceedings in the usual quality and reasonable period of time. Looking forward to seeing all of you again in Florence in 1999, hopefully with much more sun.

Comprising 29 chapters divided into four major parts, the book describes the molecular natures of enzymes, transporters, and receptors for lipid mediators (Part I), the function of lipid mediators in Drosophila and Zebrafish (Part II), the relationships between lipid mediators and various diseases (Part III), and detailed procedures of extraction, preparation, and quantification of lipid mediators (Part IV). Research on lipid mediators initially started with analysis of the action of aspirin, and subsequent biochemical experiments identified many enzymes and receptors responsible for the biosynthesis and signal transduction of individual lipid mediators. Through the phenotypic analyses of transgenic and knockout mice, it has been shown that the dysregulation of some lipid mediators causes inflammatory, immune, or oncogenic disorders. Lipid mediators have attracted increased attention because their structures are conserved among different species, and their biosynthetic and signaling pathways have been deciphered at the molecular level. Many drugs that target lipid mediators are already being used in hospitals, and this book suggests further possibilities for development of a wide variety of such drugs. Very recently, highly sensitive mass spectrometry has begun to be used to identify novel lipid mediators that are present only in trace amounts in tissues but with robust biological activity. Bioactive lipids, or lipid mediators, transduce signaling as intracellular messenger like phosphoinoitides, and also regulate cell-cell communication through G protein-coupled receptors (GPCRs) that are potentially valuable drug targets in many diseases. Until now, about 40 GPCRs within ~300 rhodopsin-like (class A) GPCRs, are identified as lipid GPCRs. Advances of lipid research have enabled to develop novel small molecules targeting lipid GPCRs for several diseases. Most notably, fingolimod (FTY720), a sphingosine 1-phosphate (S1P) receptor modulator, became the first FDA-approved medicine as an orally bioavailable drug for treating relapsing forms of multiple sclerosis (MS). In addition to fingolimod, other drugs targeting lipid GPCRs had been developed such as latanoprost (prostaglandin F2a analogue, used for ocular hypertension and glaucoma), epoprostenol and treprostinil (prostaglandin I2 analogue, used for pulmonary arterial hypertension), montelukast and pranlukast (cysteinyl leukotriene receptor antagonist, used for asthma and allergies), etc. Novel drugs are also expected like lysophosphatidic acid (LPA) receptor antagonist for treatment of pulmonary fibrosis. Drug development targeting lipid signalling pathways are backdated to more than a century, when aspirin was synthesized and selling by Bayer, while the basic mechanism of aspirin's effects (block prostanoid synthesis by inhibiting cyclooxygenases) had not been discovered until 1970s. Nowadays, non-steroidal anti-inflammatory drugs (NSAIDs) like aspirin and ibuprophen are commonly used as antipyretic analgesics and available readily over-the-counter oral drugs. Both upstream and downstream enzymes, such as phospholipase A2s and prostaglandin E synthases, respectively, are also potential therapeutic targets for inflammatory diseases. Recent studies of lipid metabolism expand the lipid biology field from pro-inflammatory lipid mediators to anti-inflammatory epoxy fatty acids (epoxyeicosatrienoic acids), and also omega-3 fatty acid-derived pro-resolving lipid mediators (lipoxin, resolvin, and neuroprotectin). These bioactive lipids, their metabolic pathways and receptors are of great interest in developing next-generation anti-inflammatory and pro-resolving drugs for a wide variety of diseases including. This book summarizes not only historical overview of lipid signaling pathways but also provides summary of cutting-edge studies that may provide some hints of novel “druggable” lipid signaling targets.

The program consisted of 83 oral and 29 poster presentations, 74 of which are included in these proceedings. The major sponsors of the conference were the Armed Forces Radiobiology Research Institute, located in Bethesda, Maryland, the Radiation Oncology Research and Development Center of the Gershenson Radiation Oncology Center, Harper Hospital in Detroit, Michigan, and Schering AG of West Germany. Eighteen other organizations provided additional support. The conference was unique in its attempt to link the eicosanoid and lipid researchers in the radiobiology and cancer disciplines. The diverse roles that eicosanoids and other bioactive lipids play in these biological phenomena including the participation of lipid oxidation in conversion of procarcinogens, positive and negative modulation of tumor growth, immunomodulation, tissue injury, and yet protection and enhancement of cancer therapy, necessitated scientific interaction to sort out and understand these complex and sometimes contradictory observations. The success of this effort is reflected not only through these proceedings, but also through the decision to continue the conference series with a second meeting to be held in Berlin between September 17-21, 1991.

The headquarters of the meeting was the Radisson Kauai Beach Resort. This international meeting was attending by approximately 80 individuals from 23 nations and 4 continents. Seventy-five papers were presented either as platform presentations or poster presentations. Taurine, first isolated from ox bile in 1827 by Tiedemann and Gmelin and named in 1838 by Demarcay, became of significant scientific interest in 1968 when the first extensive review article was published by Jacobsen and Smith. Interest in taurine grew exponentially after 1975 when the first taurine symposium was organized by Ryan Huxtable in Tucson, Arizona. Since that date, taurine symposia have been held approximately every two years held in various cities and resort areas around the world. Taurine investigators have had the privilege of attending these scientific meetings on three continents - Asia, Europe, and North America. Since the initial meeting in 1975, a central question addressed during many of the symposia has been: "What is physiological, pharmacological, nutritional, and pathological role of taurine?". Although taurine has been established as an important osmolyte, it appears to affect many other biological processes. However, the exact mechanism(s) by "which taurine acts" has not yet been definitively answered. In Kauai, the patticipants discussed many topics and asked many questions regarding the role and actions of taurine.

Book excerpt: It is with great pleasure that I write this Foreword to the Proceedings of the International Conference on Behçet’s Disease which was held in Berlin in June 2002. This was the first International Conference held under the auspices of the International Society for Behçet’s Disease which was founded in 2000 in Seoul. First, I congratulate our colleagues in Berlin, led by Professor Christos Zouboulis of the Department of Dermatology at the Free University of Berlin, for having organised a most successful conference and for having compiled these proceedings so rapidly. It will be realised immediately on scanning the contents of this book that the conference was truly international with 210 participants from 26 countries, as Professor Zouboulis has noted in his preface. These included basic scientists, epidemiologists, pathologists, clinicians and, importantly, representatives from patient organisations. The latter held their own conference alongside the scientific-medical conference to mutual benefit. The combined session of patients and doctors (abstracts on pp 601 – 626) gave the opportunity for an exchange of information and fruitful discussion. The wide ranging scope of the communications is evident from the index and it was most encouraging to see their origin – from all parts of the world, from senior and junior colleagues and, from many different disciplines. Many communications may be regarded as preliminary reports of research in progress and we look forward to seeing the definitive publications in appropriate journals in due course.

Over 250 delegates from 28 countries attended the Symposium. The Symposium logo (Picture 4, next page) presents a bacteriophage attached to the surface of the bacterium. One can easily imagine that most of the aspects covered in this Symposium are included in the logo: the bacteriophage genome encodes for structural proteins, adhesins and effector proteins that interact with the host cell in most intricate ways to carry out their mission. Life of the bacteriophage depends on the tightly regulated interplay between the phage and the host proteins. This all is also true between Yersinia and the different hosts and environments it encounters during its life cycle. This Symposium Proceedings volume is based on the oral and poster presentations given during the Symposium. The volume has been divided into six parts covering topics such as genomics, surface structures, bacteriophages, molecular and cellular pathogenesis, molecular epidemiology and diagnostics, gene regulation, clinical aspects and vaccines. These topics reflect righteously the present trends in the bacteriology research.
