Improvement of spatiotemporal resolution of fMRI to enhance neuronal activity specificity and pinpoint the microvasculature activation is the ultimate goal. fMRI functional activity/connectivity and graph theory-based connectome analyses with a multiplex fast imaging technique are recently developed new techniques to provide both a whole-picture and regional specialization of brain resource utilization in a systematic way. The integration of neuroscience and mathematical as well as physical and physiological metrics also promote the innovation and applications of multi-modal imaging equipment development and multi-parametric utilizations in clinical environments, including disease diagnosis and treatment.The aim of this book is intended to provide a whole picture of new fMRI imaging methodological developments from principles to applications, to both beginners and experts in biomedical imaging and healthcare. Several fundamental principles and metrics including BOLD, microvasculature detection and fMRI/EEG coupling will be presented together with some new and up-to-date analytical methods. The conventional statistical methods and relatively new methods of wavelets, dynamic linear correlation coefficients, independent component analysis, EEG source imaging and multi-modal integration will be covered. Book excerpt: Volume 1: The current understanding of the organization of the human visual system is discussed, with emphasis on the travelling wave and population receptive field (pRF) modeling methods of measuring visual field maps (VFMs) in humans. Functional magnetic resonance imaging (fMRI) measurement of pRFs of neurons in the visual cortices of humans revealed VFMs organized into roughly circular `clover leaf' clusters. These VFMs were compared between normal, aging, and diseased brains to understand how the brain changes over time and help develop tools to differentiate normal aging and dementia. Finally, visual working memory, the ability to hold things in mind for brief periods of time, and its likely relationship to VFMs is discussed, with implications for future research. Volume 2: Plasticity, the brain's ability to change itself, is discussed in the context of normal and abnormal development in individuals with rod monochromatism, achiasmia, and a missing right hemisphere, with an emphasis on human fMRI measurement methodology. The rod scotoma lacks rod photoreceptors and thus visual input under low-light conditions, yet subjects report perception in the scotoma to varying degrees across stimuli, which we linked to cortical fMRI measurements in VFMs. Plasticity was measured for visuo-motor transformations by left-right reversing the visual field for normal subjects for extended periods and using fMRI to track changes in their VFMs in posterior parietal cortex. Short-term adaptation to left-right visual field reversal was compared between two polymorphisms of brain-derived neurotrophic factor (BDNF), revealing that subjects with resulting higher plasticity took longer to adapt than their counterparts. Volume 3: The current understanding of the organization of the human auditory system with respect to the tonotopic (frequency) mapping of the cochlea is discussed, with emphasis on the challenges of localizing auditory maps with only one sound dimension. A modified version of the travelling wave method was used to investigate auditory cortex with fMRI, providing the first evidence in humans for two-dimensional auditory field maps (AFMs) consisting of orthogonal tonotopic (frequency) and periodotopic (temporal) gradients. Finally, cross-sensory activation in human primary visual and auditory cortex was measured using the modified travelling wave and pRF modeling fMRI methods.

Visual field mapping using fMRI has been used clinically to assess patients with cortical pathology and to plan surgical treatment impacting the visual system. The accuracy of fMRI-based visual field mapping methods needs to be better understood for clinical use. This accuracy can be important for presurgical mapping of brain function near a tumor resection site since inaccurate rendition of the underlying neural function could lead to inappropriate resection of viable brain tissue. The most widely used method for visual field mapping is temporal phase mapping. This dissertation investigates the accuracy of temporal phase mapping, specifically focused on the detection of polar angle visual field locations in primary visual cortex. Early studies show that polar angle positions are not uniformly distributed as suggested by animal studies. These non-uniformities are seen as relatively under-represented areas in the visual field maps used to display the fMRI data. This dissertation shows that temporal phase mapping is susceptible to hemodynamic distortions that lead to missassignment of visual field locations. Further analysis of the non-uniformity in the frequency distribution of voxels representing different angular position within the visual field shows an under-representation of locations near the vertical meridia in V1. These results led to the development of a new retinotopic mapping technique, code-based mapping. The main reason for developing a new retinotopic mapping technique was to reduce the under-representations of vertical meridia posed by using temporal phase mapping when assigning a stimulus location to a voxel. This dissertation shows that code-based mapping is a viable method for mapping visual field locations and produces a uniform distribution of voxels representing different angular positions within the visual field. Furthermore, the code-based mapping method is less susceptible to the hemodynamic biases than temporal phase mapping. With respect to clinical utility of fMRI mapping techniques, the code-based mapping shows a greater potential to accurately map a patient's visual field in the presence of a tumor or other malformations that can induce large noise effects in the fMRI voxel responses.

Book excerpt: Conventional fMRI image analysis is performed by carrying out a massive number of parallel regression analyses. fMRI signal is known for its low signal-noise-ratio, and its complexity, such as reflected by spatial and temporal autocorrelation. In order to ensure accurate localization of brain activity, stimulus administration in an fMRI session is often lengthy and repetitive. In real time fMRI, signal processing is carried out while the signal is being observed. This method allows for the dynamic adjustment of stimuli through sequential experimental designs. We have developed a voxel-wise sequential probability ratio test (voxel-wise SPRT) approach for dynamically localizing activation associated with stimuli, as well as decision rules for the stopping of experimentation. Stopping is dynamically determined when sufficient statistical evidence is collected to assess the activation status of voxels across regions of interest. Simulation studies show that the number of scan units can be reduced substantially compared to standard fMRI experimental designs that are fixed and predetermined, while still achieving comparably high levels of classification accuracy. An analysis based on actual brain imaging confirms the promise of this approach.An interesting application of dynamic adjustment of fMRI stimuli is in the area of Alzheimer's disease (AD). It is clear that there is a fair amount of heterogeneity in the cognitive course of the disease. This has led to the development of theories related to the notion of cognitive reserve, which posits that neural capacity, efficiency, and plasticity play a role in this heterogeneity. It has been further hypothesized that cognitive reserve levels at pre-symptomatic stage of AD will manifest specific neural activation patterns under carefully designed fMRI experimentation that systematically varies difficulty levels of a targeted task. A sequential testing approach is proposed for efficiently and accurately identifying and classifying such patterns. Methods for characterizing cognitive reserve that are studied here are comprised of two approaches. The first is sequential estimation through monitoring confidence interval lengths over a range of experimental conditions to assess efficiency and capacity. The other is sequential selection of difficulty levels, to detect neural compensation, which is a reflection of plasticity. Both approaches show high efficiencies and high detection accuracies in our fMRI simulation studies. These two approaches open up new possibilities for studying and characterizing cognitive reserve, which will in turn lead to a better understanding of processes in AD.

Activation was detected in the primary motor cortex, the supplementary motor area, the premotor cortex, the somatic-sensory cortex, the posterior parietal cortex, the primary visual cortex, and the middle temporal area. A method for measuring the time interval between onset of the average blood oxygenation level dependent response in the primary visual and motor cortices is proffered. On average, the measured time interval was shown to correlate with the kinematically measured visuomotor reaction time. The same method was used to measure delays in primary visual cortex of each hemisphere in an experiment where there were known delays between half-field visual stimuli. The measured time interval is shown to correlate with stimulus lag over a range of delays from 0-1000 ms.
