Book excerpt: Hybridization poses the greatest threat to the recovery of the critically endangered red wolf (Canis rufus ) in eastern North Carolina. Mitigating the impacts of hybridization requires understanding the scale at which this process occurs across the landscape. Such descriptions of hybridization also require an evaluation of the methods used to assess hybridization and mechanisms regulating this process. The goal of this dissertation was to discern landscape-level patterns of hybridization between red wolves and coyotes ( C. latrans ) and elucidate mechanisms governing interbreeding between these species. To describe the patterns of hybridization between these species, we used non-invasive genetic sampling to characterize the genetic profile of canids across the landscape. In 2008, we collected scat across a 22 000 km 2 region surrounding the Red Wolf Experimental Population Area (RWEPA) and used mitochondrial and nuclear microsatellite DNA data to identify individual canids and determine their ancestry (Chapter 1). From this survey we identified no pure red wolves and found little evidence of red wolf introgression into the coyote population. A similar follow-up survey conducted in 2010 confirmed that red wolves were restricted the RWEPA (Chapter 2). Coyotes were found to be widely distributed across the entire landscape; hybrids, however, were rare compared to the abundance of the parental species. For both surveys we utilized Bayesian clustering methods to estimate genetic ancestry for individuals identified through molecular genotyping. We tested the ability of these methods to detect genetic introgression under different scenarios using data from the red wolf pedigree (Chapter 3). We also examined characteristics of red wolves involved in known hybridization events within the RWEPA to illuminate potential mechanisms governing this process. Our review found that most of the red wolves that hybridized with coyotes did so following the disruption of a stable breeding pair of red wolves, often due to human-caused mortality (Chapter 4). Most hybrid litters occurred in the furthest west portion of the RWEPA and were produced by young, first-time breeding red wolves with mixed ancestry. Our findings provide valuable information regarding the patterns, process and detection of hybridization that can guide future management of this system.

Longitudinal reproductive events were investigated to determine whether inbreeding, parental age, and breeding experience were factors in reproductive performance and fitness. A behavioral preference study using olfactory presentations of conspecific and congeneric social odors also was conducted to determine those factors that are important in the selection of mates. Over 30 years of managed breeding, the level of inbreeding in the captive population has increased, and litter size has declined. Inbreeding levels were lower in sires and dams that reproduced than in those that did not reproduce, but there was no difference in the level of inbreeding of actual and predicted litters. Litter size was negatively affected by offspring and paternal levels of inbreeding, but the effect of inbreeding on offspring survival was restricted to a positive influence. Younger wolves were more likely to reproduce, and were more likely to produce larger litters, than were older individuals. The age of the dam, but not the sire, had a significant negative effect on pup survival. Sires and dams that had prior experience in the production of offspring were more likely to reproduce again than were individuals without prior reproductive success, but prior sexual experience alone was not a factor in the production of offspring. Parental breeding experience had a significant negative effect on pup survival, but no apparent relationships with size or sex ratio of the litter. In general, females responded to the presentation of social odors of conspecific males more quickly than did males, and males responded more quickly to social odors of conspecific females than did females. There were no differences in the proportion of time males and females spent proximal to conspecific social odors. Males and females typically responded more quickly to and spent more tim.

Before human disruption of the environment, rufus, lups, and latrans maintained their distinct status, and available specimens show no evidence that the red wolf intergraded with either coyote or gray wolf. A series of 71 skulls shows a complete variation from typical rufus to typical latrans. Apparently modification and decline of the red wolf population led to the breakdown of isolation and the formation of a hybrid swarm that suggests possible interbreeding with the coyote. Specimens indicate that as red wolves were extirpated in eastern Texas and elsewhere, a hybrid swarm moved eastward to occupy the niche that was created. By 1969, the swarm spread throughout most of eastern Texas and moved into Louisiana. Pure red wolves continued to survive only in a limited area along the Gulf Coast from Brazoria Counties, Texas, and in Cameron Parish.
