Often, nonlinear programming methods can be tailored to take advantage of the least squares, or more generally, the maximum likelihood, objective function. In previous studies we have developed large-scale nonlinear programming methods that are based on tailored quasi-Newton updating strategies and matrix decomposition of the process model. The resulting algorithms converge in a reduced space of the parameters while simultaneously converging the process model. Moreover, tailoring the method to least squares functions led to significant improvements in the performance of the algorithm. These approaches can be very efficient for both explicit and implit [sic] models (i.e., problems with small and large degrees of freedom, respectively). In the latter case, degrees of freedom are proportional to a potentially large number of data sets. Applications of this case include errors-in- all-variables estimation, data reconciliation and identification of process parameters. To deal with this structure, we apply a decomposition approach that performs a quadratic programming factorization for each data set. Because these are small components of large problems, an efficient and reliable algorithm results. These methods have generally been implemented on standard von Neumann architectures and few studies exist that exploit the parallelism of nonlinear programming algorithms. It is therefore interesting to note that for implicit model parameter estimation and related process applications, this approach can be quite amenable to parallel computation, because the major cost occurs in matrix decompositions for each data set. Here we describe an implementation of this approach on the Alliant FX/8 parallel computer at the Advanced Computing Research Facility at Argonne National Laboratory. Special attention is paid to the architecture of this machine and its effect on the performance of the algorithm. This approach is demonstrated on five small, undetermined regression problems as well as a larger process example that demonstrates simultaneous data reconciliation and parameter estimation."

Book excerpt: In the analysis of complex phenomena arising in biology, medicine, physics, economics or the social sciences, it is not uncommon to employ mathematical models at different scales. Microscopic models tend to be better suited to capture the fine-scale details of a complex system and are typically stochastic in nature, while macroscopic models, sometimes arising as mean-field approximations to microscopic models, are typically used to describe the overall, population-level dynamics of the system. Typically, both coarse- and fine-scale models depend on parameters whose values are unknown or poorly known, hence need to be estimated. The estimation of the parameters of larger scale models tends to be more straightforward and approachable with standard optimization-based tools. In several important applications, the microscopic model parameters are of greater interest and importance, as they may be interpreted as characterizing the generative process underlying the large-scale model. Due to the stochastic nature of these types of models, the unknown parameters are difficult, if not impossible, to estimate directly. Ideally, we would like to estimate the parameters of the coarse-scale model and relate them to those of the fine-scale model. This connection, however, may be all but trivial to establish, due to the fact that models at different scales may depend on parameters that do not exhibit a one-to-one correspondence. In this work, we propose a Bayesian approach for inferring on the values of the microscopic model parameters, based on estimates of the parameters of the associated macroscopic model. Our methodology is based on connecting model parameters across scales via the posterior probability density, and the subsequent approximation of this density using random samples. We illustrate the viability of our technique with several computed examples, ranging from simple to fairly complex, with a focus on applications in the life sciences.
