Book excerpt: Flax (Linum usitatissimum L.) is one of the richest plant sources of omega-3 fatty acids praised for their health benefits. In this study, the extent of the genetic variability for genes encoding stearoyl-ACP desaturase (SAD), fatty acid desaturase 2 (FAD2) and 3 (FAD3) was determined by sequencing the six paralogous genes from 120 flax accessions representing a broad range of germplasm including some EMS mutant lines. A total of 6 alleles for sad1 and sad2, 21 for fad2a, 5 for fad2b, 15 for fad3a and 18 for fad3b were identified. Deduced amino acid sequences of the alleles predicted 4, 2, 3, 4, 6, and 7 isoforms, respectively. Allele frequencies varied greatly across genes. Fad3a, with 110 SNPs and 19 indels, and fad3b, with 50 SNPs and 5 indels, showed the highest levels of genetic variation. While most of the SNPs and all the indels were silent mutations, both genes carried non-sense SNP mutations resulting in premature stop codons, a feature not observed in sad and fad2 genes. Some alleles and isoforms discovered in induced mutant lines were absent in the natural germplasm. Correlation of these genotypic data with fatty acid composition data of 120 flax accessions phenotyped in six field experiments revealed statistically significant correlations of some of the SAD and FAD isoforms on fatty acid composition, oil content and iodine value. The novel allelic variants and isoforms identified for the six desaturases will be a resource for the development of oilseed flax with unique and useful fatty acid profiles.

Available in PDF, EPUB and Kindle. Book excerpt: Abstract Flax (linseed, Linum usitatissimum L) is cultivated either for its seeds or its stem (bast fiber). With the goal of contributing to development of novel flax germplasm, a forward genetics screen of EMS mutagenized flax (var. CDC Bethune) was conducted to identify seeds with abnormal pigmentation or mucilage. Seeds of 16,764 M4 families were examined visually, with and without staining by Toluidine Blue. A total of 86 putative seed color mutants were identified, with the most common phenotype being yellow seeds. There were 1,975 identified putative mucilage mutants, with a range of phenotypes, including Blue Seed, No Mucilage Released, and Abnormal Mucilage Released. A subset of the seed colors and mucilage phenotypes were tested for heritability. The seed colors were generally heritable, but the mucilage phenotypes appeared to show variable expressivity that is probably highly dependent on the environment. Histological analysis revealed how seed coat anatomy changes affected seed color, but did not explain the mucilage phenotypes tested. The lines have been provided to flax breeders other collaborators for further analysis and potential use in breeding programs. GASA (Gibberellic Acid Stimulated in Arabidopsis) genes encode a family of small proteins of unknown biochemical function, whose expression has been correlated with various processes in plants including hormone response, defense, and development. GASA genes have also been reported in our lab to be expressed in developing flax stems. To better characterize the GASA-like gene family in flax, I identified 20 predicted GASA-like genes in the flax whole genome assembly. Using microarray and qRT-PCR analysis, seven of these genes showed differential transcript expression during stem development. I cloned and verified the sequences of these genes. Based on these studies, I was able to define groups of flax GASA-like genes whose expression was well-correlated with stem segments in either the elongation or cell wall deposition stage. These results provide further evidence that specific GASA genes are involved in different processes in flax stem and/or fiber development.

Among the bast fibers are utee, kenaf, ramie, flax, Hibiscus spp. and leaf fibers such as Agave sp. The present gives a detailed account of important bast fibers of world with respect to utility, botany, development of fibers, anatomy, methods of extraction, quality.
