The sequence is 'open' when read in an appropriate frame, which is to say that there is a long run of amino acid codons before the appearance of a terminator codon. How can we find out if this 'unidentified reading frame' (URF) really codes for a genuine protein, and how can we identify it if it exists? There are two general strategies, both of which can also be applied to the characterization of any 'open reading frame' (ORF), whether or not it has been 'identified'. The first and simplest approach involves computer searching and analysis; the second employs antibodies raised against synthetic peptides patterned on the sequence of the expected gene product. Both methods have been used with great success by many investigators. Each has, nonetheless, its pitfalls and frustrations. This primer is meant to guide the researcher past those obstacles as much as possible. Such an approach is unfortunately hampered by the following constraints : the costs as weil as the technical difficulties of analyzing every individual chemical which may be present in the sampies, and the difficulty of assessing the hazards and risks of environmental contaminations from a set of chemical data. During the last decades the scientific and regulatory community has gradually realized that biological methodologies have to be taken into consideration for an ecologically meaningful assessment of the toxicological hazards of contaminants. Effect evaluations obtained with biological techniques indeed integrate the impact of all the contaminants to which living biota are exposed. Bioassays with selected test species representative for the biological commumtles of the environments under consideration, are now applied more or less regularly to determine toxic and genotoxic effects. Taking into account the species specific and chemical specific character of toxicity to biota, the necessity of a «battery of tests» approach with species of different trophic levels is currently also generally accepted and implemented. It is dear that a balanced partnership between chemical, biological, toxicological and microbiological analyses is always the best strategy for generating the broadest information base on environmental hazards.

Although they are not considered a formal cate gory by some taxonomists and some biologists consider the name inappropriate (inferring that they are the first unicellular animals, although some photosynthe size), it is still convenient to consider this group of organisms as an informal collection under the heading of protozoa. Their cosmopolitan distribution, sig nificant ecological role in mineral recycling and enhancement of carbon flow through lower trophic levels of food webs, and remarkable cellular adaptations to enhance survival in diverse environments make them significant organisms for biological investigation. In some cases, biologists are introduced to this group in first level courses or in invertebrate zoology, but never develop a full appreciation for the diverse and biologically sophisticated characteristics of these organisms. This book is intended as a survey of broad concepts in protozoan biology with an emphasis on comparative data. The focus is on the zoological aspects of the group. Topics more closely related to plantlike characteristics, as presented in books on phycol ogy, are not considered in detail here. A sound background in modern biology and an introduction to cellular biology will be helpful in understanding Chapters 15 and 16, which include a substantial amount of information on biochemistry.
