Understanding the first-ever accurate Age Predictor…” Greying hair and wrinkles are external signs of ageing, but they are not very precise. Now research shows that a code written into the body's epigenome — the chemical tags that modify DNA — can accurately tell the age of human tissues and cells. This ‘clock’ innovated by Steve Horvath could provide insights into why certain tissues age faster than others, and why those tissues may be more cancer-prone. In the past few years, researchers have been homing in on regions of DNA that accrue lots of chemical tags called methyl groups as people age. Such methylation can selectively switch off genes. What was not yet known was that one can develop an age predictor that really works well across most tissues and cell types. It would be of interest to mention that women’s breast tissue accrues methylation in a way that makes it look an average of 2–3 years older than other healthy tissues from the same woman. In women with breast cancer, healthy tissue situated next to diseased tissue appeared to be an average of 12 years older than other tissues in the body. And Steve Horvath found that tissue from 20 cancer types looked an average of 36 years older than healthy tissue. An attempt has been made in this booklet to describe, how methylation levels change in human tissues from before birth to the age of 101, and shows that it is a near-perfect predictor of age for non-cancerous tissues. Since Ageing is a major health problem, and interestingly there are really no objective measures of ageing other than a verified birth date, studies like this one provide important new efforts to increase the rigor of human ageing studies.” It would be worth mentioning here that Horvath, who has developed software to help people use his clock to track the age of tissue samples, hopes that the research could lead to treatments that slow or reset the ticking of the biological clock. ……Dr.H.K.Saboowala.

Many epigenetic alterations can have an impact on aging processes such as stem cell aging, control of telomerase, modifications of telomeres, and epigenetic drift can impact the aging process as evident in the recent studies of aging monozygotic twins. Numerous age-related diseases are affected by epigenetic mechanisms. For example, recent studies have shown that DNA methylation is altered in Alzheimer’s disease and autoimmunity. Other prevalent diseases that have been associated with age-related epigenetic changes include cancer and diabetes. Paternal age and epigenetic changes appear to have an effect on schizophrenia and epigenetic silencing has been associated with several of the progeroid syndromes of premature aging. Moreover, the impact of dietary or drug intervention into epigenetic processes as they affect normal aging or age-related diseases is becoming increasingly feasible.

Book excerpt: Candidate gene based studies have identified a handful of aberrant CpG DNA methylation events in prostate cancer (Brooks et al. 1998; Yegnasubramanian et al. 2004). However, large scale DNA methylation profiles have not been examined for normal prostates or prostate tumors. Additionally, the mechanisms behind these DNA methylation alterations are unknown. In this thesis, I describe the results of my efforts to better understand these previously unexplored areas of biology. For the study presented in this thesis, I quantitatively profiled 95 primary prostate tumors and 86 healthy prostate tissue samples for their DNA methylation levels at 26,333 CpGs representing 14,104 gene promoters by using the Illumina HumanMethylation27 platform. When the profiles of the prostate tissue samples were compared, I observed a substantial number of tumor-specific DNA methylation alterations. This accumulated data allows researchers to assess the rate of aging processes in various organs and systems, and to individually monitor the effectiveness of therapies intended to slow aging. In an introductory chapter, the editor defines biomarkers of aging as molecular, cellular and physiological parameters that demonstrate reproducible changes - quantitative or qualitative - with age. The introduction recounts a study which aimed to create a universal model of biological age, whose most predictive parameters were albumin and alkaline phosphatase (indication liver function), glucose (metabolic syndrome), erythrocytes (respiratory function) and urea (renal function). The book goes on to describe DNA methylation, known as the "epigenetic clock," as currently the most comprehensive predictor of total mortality. It is also useful for predicting mortality from cancer and cardiovascular diseases, and for analyzing the effects of lifestyle factors including diet, exercise, and education. Individual contributions draw additional insight from research on genetics and epigenetic aging markers, and immunosenescence and inflammaging markers.
