Book excerpt: Ce travail a été effectué au Département Physique et Métrologie des Oscillateurs (L.P.M.O.), du laboratoire FEMTO-ST, UMR 6174 du CNRS, en collaboration avec l'U.F.R. des Sciences Médicales et Pharmaceutiques de l'Université de Franche-Comté. Ces travaux de thèse ont été encadrés par le professeur Lysiane RICHERT et le docteur Wilfrid BOIREAU (Chargé de Recherches au C.N.R.S.). Des colonnes chromatographiques en silicium mésoporeux ont été conçues et fabriquées. Après avoir appris à utiliser des techniques de salle blanche (RIE, Pulvérisation cathodique, photolithographie...), j'ai procédé aux diverses étapes de fabrication à partir d'un substrat en silicium dopé au phosphore. Leur caractérisation a été effectuée à l'aide de microscopes électroniques à balayage et optiques. Une séparation chromatographique de deux molécules a été réussie en utilisant le pouvoir séparateur de la silice. La deuxième partie de mes travaux de thèse s'inscrit dans le cadre d'une étude de Biologie Cellulaire et de recherches en protéomique. En collaboration avec l'U.F.R. des Sciences Médicales et Pharmaceutiques, les effets d'un traitement à la Nafénopine (un proliférateur de péroxisomes) sur les protéines microsomiales des cellules hépatiques de rats ont été étudié. Après la lyse des cellules hépatiques des sujets et la récupération des protéines microsomiales, l'échantillon est injecté dans un appareil de chromatographie liquide bidimensionnelle conçu pour la protéomique, le ProteomeLab PF2D. Cet appareil sépare un mélange de protéines suivant deux types de chromatographie liquide; suivant leur point isoélectrique par chromatofocusing et suivant leur hydrophobicité par RP-HPLC. Après étude différentielle des cartes protéiques obtenues, un bio-indicateur a été découvert et identifié par Western Blotting. Il s'agit du Cytochrome P450 4Al.
