The ability to produce miniature sensors that are immune to electrical interference, that do not require a reference electrode, and that can be operated remotely are attractive advantages of this technology. Fiber optics provide a mechanism for continuous optical monitoring of process, biological, or hazardous environments that previously were difficult or impossible to access. Fluorescence spectroscopy is particularly suited for fiber optic sensing because of its sensitivity, selectivity, and versatility. In recent years, a number of sensing configurations have been described that take advantage of either the intrinsic fluorescence of the analyte or incorporate a fluorescent reagent at or near the sensor tip. Analyte concentrations can be determined from the enhancement, quenching, changing lifetime, or colorimetric modulation of the induced reagent fluorescence. These individual sensors integrate technology of fiber optics, chemistry, optics, and spectroscopy into their designs. The operating principles and optical configurations of typical fiber optic fluorescent sensors are described.

Semiconductor photochemistry plays an important role in these and is discussed with reference to both of these areas. Free radicals and reactive oxygen species are of major importance in many chemical, biological and medical applications of photochemistry, and are discussed in depth. The following chapters discuss the relevance of using light in medicine, both with various types of phototherapy and in medical diagnostics. The development of optical sensors and probes is closely related to diagnostics, but is also relevant to many other applications, and is discussed separately. Important aspects of applied photochemistry in electronics and imaging, through processes such as photolithography, are discussed and it is shown how this is allowing the increasing miniaturisation of semiconductor devices for a wide variety of electronics applications and the development of nanometer scale devices. The final two chapters provide the basic ideas necessary to set up a photochemical laboratory and to characterise excited states. This book is aimed at those in science, engineering and medicine who are interested in applying photochemistry in a broad spectrum of areas. Each chapter has the basic theories and methods for its particular applications and directs the reader to the current, important literature in the field, making Applied Photochemistry suitable for both the novice and the experienced photochemist.

The stated aim of the workshop was to highlight the significance of fluorescence work for the understanding of cell and tissue physiology, physiopathology and pharmacology, particulary in terms of the analytical use of fluorescent probes in oncology. In the organization of the workshop a multidisciplinary approach was selected. The purpose of the Advanced Research Workshop (ARW) was to bring together researchers in the various disciplines of tissue optics, imaging, microspectrofluorometry and state of the art probes, in order to explore the full benefits that can be derived in biomedicine through the convergence of these approaches. When applied to in vivo and in situ studies, fluorescence and related optical methods enable us to explore within tissues, cells and organelles photon effects previously understood only in solution photochemistry. Processes which can be studied at the molecular level by photophysics, photochemistry and physical chemistry can be evaluated in living tissue by fluorescence spectroscopy and imaging at the intracellular level in terms of structure and function. Thus, fluorescence adds a new dimension to cell biology and physiology. This approach is now supported by a full and versatile, rapidly growing armamentarium of new selective probes for organelles, enzymes, cations, cytoskeleton and metabolic control.

In particular, advances have been made in visible radiation transmission efficiency with concurrent reductions in fiber size, weight, and cost. Researchers at the Lawrence Livermore National Laboratory (LLNL) coupled these advances in fiber optic technology with analytical fluorescence analysis to establish a new technology - remote fiber fluorimetry (RFF). Laser-based RFF offers the potential to measure and monitor from one central and remote laboratory, on-line, and in near real time, trace (ppM) to substantial (g/L) concentrations of selected chemical species in typical process, product, and waste streams. The fluorimeter consists of a fluorescence or Raman spectrometer; unique coupling optics that separates input excitation (laser) radiation from return (fluorescence) radiation; a fiber optic cable; and an optrode - a terminal that interfaces the fiber to the measurement point, which is designed to respond quantitatively to a particular chemical species. At LLNL, research is underway into optrodes that measure pressure, temperature, and pH and those that detect and quantify various actinides, sulfates, inorganic chloride, hydrogen sulfide, aldehydes, and alcohols.

The improvements that can be effected in sensitivity, analy tical selectivity, and ability to handle "difficult" samples by laser fluorometry have only begun to be exploited. Likewise, time-resolved fluorometry has received widespread use in fundamental studies (a sizable number of which deal with biological systems), but has as of yet received relatively little analytical utilization. The use of electronic array detectors offers the promise of obtaining luminescence spectra more rapidly, and perhaps ultimately with greater sensitivity, than is possible by the use of scanning instruments equipped with conventional detectors. The increasing capabilities of microcomputers and the increasing sophistication of "smart" spectroscopic instrumentation signify that much more efficient acquisition and use can now be achieved of the information contained in the "excitation-emission matrix" inherent in the luminescence phenomenon.
