Book excerpt: The immunoglobulin gene complex is responsible for generating an extraordinarily wide range of antibodies, each possessing a unique antigen specificity. The Second Edition of Immunoglobulin Genes brings the reader up to date with the rapid progress in our understanding of this system. Firmly established as the definitive book on the topic, it provides a fully comprehensive account of the organization, function, rearrangement and expression of these and related genes. Since the publication of the first edition, the work has been extensively revised and updated to cover new research data on human immunoglobulin genes as well as mammalian and lower vertebrate systems. New chapters include "Somatic mutation and B cell Maturation", "Antigen receptors" and "B lymphocyte tolerance in the mouse."For the immunologist, there are detailed descriptions of the molecular mechanisms which generate this diverse range of antibodies and the tight regulation of the genes encoding them. Molecular biologists will find discussions of important phenomena including gene rearrangement, differential splicing and deletion in the context of immunoglobulin genes. The excitement of research on immunoglobulin genes is conveyed by the range and complexity of issues discussed here; the book contains invaluable information for clinicians and research scientists with an interest in the immune response.* Conveys the excitement of research on immunoglobulin genes by the range and complexity of issues* Contains invaluable information for clinicians and research scientists with an interest in the immune response* For the Immunologist* Detailed descriptions of the molecular mechanisms generating this diverse range of antibodies and the regulation of genes encoding them* For the Molecular Biologist* Discusses important phenomena including gene rearrangement, differential splicing, and deletion in the context of immunoglobulin genes.

Book excerpt: Offering a broad appeal to microbiologists, immunologists, and infectious disease specialists, this four volume encyclopedia covers all autoimmune, tropical, and infectious diseases. Emphasis will also be placed on genetics, physiology, metabolism, pathogenesis and applied microbiology. Under the leadership of some of the most world renowned names in the field, the encyclopedia will bring together an outstanding collection of contributions by top scientists in a variety of fields. Volumes 1-3: Diseases will be divided by the 11 main sections of the body, namely Integumentary, Skeletal, Respiratory, Digestive, Urinary, and Reproductive. For some of the autoimmune disease, more then one system will be involved but the delineation serves to broadly break down the diseases into systems. Volume 4 will cover the vaccines for said diseases and future prospects will be offered by leaders in industry and academia. Volume 4 will also be broken down into all the body systems, as in the other two volumes. For each vaccine, for each disease, and in each system the following will be included: • A list of the vaccines currently available along with a list of the companies that manufacture them • Molecular Immunology of the Vaccine • Type of Immunity involved in protection • Mode of Vaccination for each vaccine; repeated boosters and length of immunological memory • Commercial production of vaccines • Storage of vaccines • Standardization and Control of Vaccines • WHO programs and World-Wide Disease Eradication Programs based upon Vaccines.

Available in PDF, EPUB and Kindle. Book excerpt: The diversity of RNAs inside living cells is amazing. We have known of the more “classic” RNA species: mRNA, tRNA, rRNA, snRNA and snoRNA for some time now, but in a steady stream new types of molecules are being described as it is becoming clear that most of the genomic information of cells ends up in RNA. To deal with the enormous load of resulting RNA processing and degradation reactions, cells need adequate and efficient molecular machines. The RNA exosome is arising as a major facilitator to this effect. Structural and functional data gathered over the last decade have illustrated the biochemical importance of this multimeric complex and its many co-factors, revealing its enormous regulatory power. By gathering some of the most prominent researchers in the exosome field, it is the aim of this volume to introduce this fascinating protein complex as well as to give a timely and rich account of its many functions. The exosome was discovered more than a decade ago by Phil Mitchell and David Tollervey by its ability to trim the 3’end of yeast, S. cerevisiae, 5. 8S rRNA. In a historic account they laid out the events surrounding this identification and the subsequent birth of the research field. In the chapter by Kurt Januszyk and Christopher Lima the structural organization of eukaryotic exosomes and their evolutionary counterparts in bacteria and archaea are discussed in large part through presentation of structures.

Book excerpt: One of the holy grails in biology is the ability to predict functional characteristics from an organism's genetic sequence. Despite decades of research since the first sequencing of an organism in 1995, scientists still do not understand exactly how the information in genes is converted into an organism's phenotype, its physical characteristics. Functional genomics attempts to make use of the vast wealth of data from "-omics" screens and projects to describe gene and protein functions and interactions. A February 2020 workshop was held to determine research needs to advance the field of functional genomics over the next 10-20 years. Speakers and participants discussed goals, strategies, and technical needs to allow functional genomics to contribute to the advancement of basic knowledge and its applications that would benefit society. This publication summarizes the presentations and discussions from the workshop.

Book excerpt: Marten Hofker and Jan van Deursen have assembled a multidisciplinary collection of readily reproducible methods for working with mice, and particularlyfor generating mouse models that will enable us to better understand gene function. Described in step-by-step detail by highly experienced investigators, these proven techniques include new methods for conditional, induced knockout, and transgenic mice, as well as for working with mice in such important research areas as immunology, cancer, and atherosclerosis. Such alternative strategies as random mutagenesis and viral gene transduction for studying gene function in the mouse are also presented.

Available in PDF, EPUB and Kindle. Book excerpt: Stem cells have been gaining a lot of attention in recent years. Their unique potential to self-renew and differentiate has turned them into an attractive model for the study of basic biological questions such as cell division, replication, transcription, cell fate decisions, and more. With embryonic stem (ES) cells that can generate each cell type in the mammalian body and adult stem cells that are able to give rise to the cells within a given lineage, basic questions at different developmental stages can be addressed. Importantly, both adult and embryonic stem cells provide an excellent tool for cell therapy, making stem cell research ever more pertinent to regenerative medicine. As the title The Cell Biology of Stem Cells suggests, our book deals with multiple aspects of stem cell biology, ranging from their basic molecular characteristics to the in vivo stem cell trafficking of adult stem cells and the adult stem-cell niche, and ends with a visit to regeneration and cell fate reprogramming. In the first chapter, “Early embryonic cell fate decisions in the mouse”, Amy Ralson and Yojiro Yamanaka describe the mechanisms that support early developmental decisions in the mouse pre-implantation embryo and the current understanding of the source of the most immature stem cell types, which includes ES cells, trophoblast stem (TS) cells and extraembryonic endoderm stem (XEN) cells.

Book excerpt: In stem cell research there are several key methods that, once mastered, can be extremely powerful. These methods enable you to rigorously test hypotheses, compare results to "gold standards," and may even spur improvements to existing protocols. This book describes numerous methods to derive, manipulate, target, and prepare stem cells for clinical use. The methods described here help you derive and test human embryonic stem cells, analyze bone marrow stem cell function in vitro and in vivo, image a stem cell transplant, cryopreserve stem cells and differentiate stem cells using microscale tec.
