Sodium is detected in the ablated vapor by using two tunable laser probes to induce resonantly enhanced multiphoton ionization via the intermediate 3P and 4D levels. At present, the technique provides only relative quantitative results. For absolute quantitative results, characterization of complex laser-solid interaction phenomena in a buffer gas is required. The minimum detectable sodium density in the solid is estimated to be about 10 to the 11th power atoms/cu cm. Generalization of this technique to other impurities is limited only by laser availability. (Author).

Book excerpt: Ever since the first report of laser action, it has been recognized that laser ablation (evaporation/volatilization) may provide a useful sampling mechanism for chemical analysis. In particular, laser ablation is rapidly gaining popularity as a method of sample introduction for mass spectrometry. While most laser ablation/mass spectrometry has been performed with fixed frequency lasers operating at relatively high intensities/fluences (≥108 W/cm2, ≥1 J/cm2), there has been some recent interest in the use of tunable lasers to enhance the ionization yield of selected components in an analytical sample. This process has been termed resonant laser ablation (RLA), and typically relies on irradiation of a sample in a mass spectrometer with modest intensity laser pulses tuned to a one- or two-photon resonant transition in the analyte of interest. Potential advantages of RLA include: (1) simplification of the mass spectrum, by enhancement of signal from the analyte of interest; (2) improvement of the absolute detection limits by improving the ionization efficiency, and (3) improvement in relative sensitivity. The sensitivity enhancement results from reduction of spurious signal, and accompanying noise, in the detection channel. This spurious signal may be due to bleed through from adjacent mass channels, or from isobaric interferences. RLA tends to produce higher mass resolution because of minimal spatial spread in the ion source and small space charge effects. In this manuscript we present a survey of RLA attributes and applications.

Book excerpt: This dissertation manuscript concentrates on methods to improve the utility of laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) as a rapid and accurate technique for quantitative analysis, forensic matching, and source attribution of solid samples. A solution calibration method for LA-ICP-MS is described that does not require matrix-matched solid standards or prior knowledge of sample composition. Desolvated particles from a microconcentric nebulizer and particles ablated from a localized position on a solid sample are mixed and introduced simultaneously to the ICP-MS. The transport of sample particulates from the ablation cell is measured online with piezoelectric microbalance to provide signal normalization. The relative accuracy of the quantitative analysis of NIST 612 glass and NIST 1264a steel is 3% and 12%, respectively, except for the elements Pb and Bi in steel. LA-ICP-MS is also used with principal components analysis (PCA) to simplify routine comparisons of forensic materials without time-consuming and destructive sample dissolution. The relative concentrations of trace elements in a solid sample are examined to provide a "fingerprint" composition that can be used for definitive identification and source matching of the material. Residue samples are matched to bulk materials using PCA. Variation of laser focus and PCA are also used to diagnose the presence and severity of elemental fractionation, which remains the most significant limitation for accurate quantitative analyses by LA-ICP-MS. High-speed photographs and video of an ICP are used to examine the validity of solution calibration for LA-ICP-MS and to study the trajectories, lifetimes, and emission behavior of particles traversing the plasma under a variety of conditions. No red emission clouds from YO are visible in the ICP when a 20-[mu]L min−1 nebulizer is used to spray a 2000 ppm aqueous yttrium solution, suggesting that the wet droplets from this nebulizer desolvate almost completely before entering the ICP. These desolvated particles appear to atomize and ionize like the small dry particulates from laser ablation. However, some large ablated particulates are observed to fly through the plasma intact, possibly contributing to elemental fractionation. Nevertheless, accurate calibration results using solution standards can be obtained for elements not affected by fractionation.

Book excerpt: Using laser ablation inductively coupled plasma mass spectrometry (LA-ICP-MS) this research analyzed human cadaver hair for gadolinium and mice hair samples for cadmium. This project combined fundamental bench science (analytical chemistry and animal studies), and insight into exposures to an emerging contaminant. This study demonstrated the feasibility of LA-ICP-MS to measure time resolved exposure profiles in hair samples, potentially opening up a transformative new exposure analysis method for investigators conducting both bench toxicology and human epidemiology studies. Consecutive autopsy cases that presented at the University of Washington's medical department, who had received a gadolinium-based contrasting agent (GBCA) to enhance magnetic resonance imaging procedures constituted the human population component of this research. Multiple hairs from numerous subjects were analyzed using LA-ICP-MS along with calibration gels (bovine-skin based) and a standard reference material glass disc with trace metals. Overall analytical method accuracy and precision was acceptable. There was moderate correlation between gadolinium dose and area under the curve as well as gadolinium peak position and GBCA injection day. Hair concentrations were also compared to the other tissue (brain, bone, and skin) concentrations using linear regression. This analysis showed good correlation between hair gadolinium concentrations and brain and skin gadolinium concentrations. These results suggest that hair may serve as a safe and effective biomonitoring tissue for patients who receive GBCA injections. A mouse animal model was used to evaluate LA-ICP-MS as a biomonitoring tool in assessing cadmium exposure. Hair samples from mice that were part of a mechanistic toxicology study at the University of Washington were opportunistically obtained for inclusion in this analytical methods-focused element of the research. The mice were dosed with cadmium-spiked water at three levels (0, 0.6, and 3 mg/L) for 14 weeks before they were sacrificed. Hairs from seven mice were analyzed using LA-ICP-MS. This research showed that there were increasing concentrations of cadmium in hair shafts with increasing doses of cadmium in drinking water among mice.
