Book excerpt: Spotted wilt disease, caused by the thrips-vectored Tomato spotted wilt virus (TSWV), is an important disease affecting peanuts in the southeastern United States. Peanut Rx is a risk index that allows growers to determine and avoid combinations of production practices that are conducive severe spotted wilt epidemics. However, the intensity of spotted wilt disease varies among fields having the same risk category depending on location and year as documented in this study. To account for the variability in spotted wilt intensity, a 'thrips risk factor' was created based on location- and year-specific predictions of peak thrips dispersal from the 'Thrips Infestation Predictor Tool for cotton'. A 'modified Peanut Rx' was developed through the integration of the 'thrips risk factor' into the risk index which provided a better risk assessment than Peanut Rx. With it, peanut growers can directly account for the thrips vector in assessing their risk of spotted wilt in order to determine the suitable production practices that would help them avoid severe epidemics and yield losses.

Book excerpt: Tomato Spotted Wilt virus (TSWV), a thrips-vectored virus, has recently become one of the most devastating pathogens of peanut (Arachis hypogaea) in North Carolina. North Carolina peanut growers saw a dramatic increase in infestations of TSWV in 2000. Certain cultural techniques in Georgia have shown to lessen the amount of virus, and these were evaluated to determine their effect on TSWV incidence. The production systems are discrete between the runner-type peanuts grown in GA and the virginia-type peanuts in NC, thereby requiring that these practices be evaluated in the NC. Treatments included plant populations, varieties, twin and single row plantings, reduced tillage, planting dates, and in-furrow insecticides. During the growing seasons of 2001 and 2002, treatments compared were plant populations of 7, 13, and 17 plants per meter of row; varieties Gregory, NC V-11, and Perry; twin and single rows; conventional tillage and strip tillage; early and late planting dates; and phorate and aldicarb. Research plots were scouted for visual symptoms of TSWV monthly in 2001, and weekly in 2002. In 2001, symptomatic plants were tested for presence of the virus using the ImmunoStrip test system (Agdia, Elkhart, IN). High plant populations had less virus than lower plant populations, Gregory was infected with less virus than either NC V-11 or Perry, twin rows had a lower amount of virus compared to single rows, preliminarily strip tillage has had less virus than conventional tillage, and peanut treated with in-furrow phorate had less incidence of virus than those treated in-furrow with aldicarb.

Book excerpt: Tomato spotted wilt virus (TSWV) and late leaf spot (LLS), caused by Cercosporidium personatum, are important diseases affecting peanuts (Arachis hypogaea L.). One of the most promising ways to combat these diseases is with resistant cultivars created through the development of recombinant inbred lines allowing for the discovery of resistance genes. These mapping populations can be very large making phenotyping time consuming and difficult; the objectives of this research aim to increase the efficiency of phenotyping methods. Field experiments were conducted to compare genotypes from multiple mapping populations for resistance to the two diseases as well as evaluate the use of unmanned aerial systems (UASs) and their ability to increase efficiency when phenotyping TSWV and LLS. Juvenile plants were also evaluated for resistance to LLS. Results indicated that some populations showed more resistance to disease than other and UASs proved to be helpful in the phenotyping of mapping populations. Parental lines evaluated were also susceptible to infection by C. personatum at a very early age.

Book excerpt: Exploring the genetic diversity and evolutionary history of plant viruses is critical to understanding their ecology and epidemiology. Tomato spotted wilt virus (TSWV) is an ambisense RNA virus that infects over 1000 species of plants and has been shown to reassort its genome. To further investigate this virus, maximum-likelihood and population genetics-based methods were used to investigate the population structure, genetic diversity, and sources of genetic variation in field isolates of TSWV from peanut in North Carolina and Virginia. Selected regions of the nucleocapsid, movement, and RNA-dependent RNA polymerase genes were amplified and sequenced to identify haplotypes and infer genetic relationships between isolates of TSWV with heuristic methods. The haplotype structure of each locus consisted of one or two predominant haplotypes and more than 100 haplotypes represented by a single isolate. No specific haplotypes or clades were associated with geographic area, peanut cultivar or year. The population was panmictic at the regional level and high levels of genetic diversity were observed among isolates. There was evidence for negative selection acting upon each locus and maximum-likelihood analyses indicated that exponential growth was occurring in the population. The results of compatibility analyses and the persistence of specific gene sequences in isolates collected over three field seasons suggest that recombination was occurring in the population. Phylogenetic analysis supports that each locus has an independent evolutionary history.
