However, the synthesis and characterization of alloy nanoparticles are still posing significant challenges, leading to an increasing demand for in situ characterization techniques. Small-angle X-ray scattering (SAXS) is a powerful method for structural analysis of nanoparticles. As the SAXS signal is essentially the Fourier transform of the electron distribution, it provides structural information for the entire ensemble of nanoparticles. The development of SAXS has been facilitated by significant advances in synchrotron X-ray sources and data processing methods, leading to the development of the 3D-SAXS method, which enables the reconstruction of the 3D structures of particles from SAXS profiles.Although SAXS has the potential to be a powerful tool for investigating the internal structures of alloy nanoparticles, its application is hindered by the challenges posed by polydispersity, which can cause smearing effects that complicate the geometry recovery process. This dissertation presents a novel approach to overcome the problem of polydispersity in SAXS data analysis, thus demonstrating the utility of SAXS in investigating the internal electron density distributions of alloy nanoparticles. In Chapter 2, the SharPy algorithm is introduced as a size-refocusing program that reduces the smearing effect caused by polydispersity in SAXS data. SharPy is based on a penalized iterative regression approach to fit the pair distance distribution function (PDDF) with an estimated size distribution. It can provide detailed information about the shape of nanoparticles from the smeared SAXS signal under various scenarios and conditions. Chapter 3 investigates the simulated SAXS profiles of AuAg core-shell nanoparticles with varying size distribution, core-shell ratio, and degrees of alloying. It demonstrates the capability of SAXS in observing the electron density distribution of AuAg core-shell structures. These findings provide insights into the potential of SAXS as a reliable method for investigating the internal structures of alloy nanoparticles. Chapter 4 focuses on synthesizing and characterizing AuAg nanoparticles. Their SAXS profiles and PDDF analysis demonstrate that SAXS can distinguish between homogeneous and core-shell nanoparticle structures. In this chapter, the SharPy algorithm is first-time applied to real experimental data, demonstrating its ability to reveal the core-shell structure from a polydisperse nanoparticle system. Chapter 5 investigates the evolution of alloying AuAg nanoparticles through a combination of SAXS/PDDF analysis, 3D reconstruction, and molecular dynamics (MD) simulation. The study presents the 3D electron density distribution of alloying AuAg nanoparticles. The 3D reconstruction with electron density mapping provides a straightforward visualization of the electron density distribution pattern of the alloying AuAg nanoparticles. The success of the SAXS experiment lies in the development of the 3D-SAXS pipeline, which involves the use of SharPy and 3D reconstruction programs, making 3D SAXS a promising alternative to electron microscopy for visualizing the morphology of nanoparticle systems.

Gold nanoparticles are nanomaterials that have less than 100 nm at least in one dimension. The composition and the structure/geometry of the nanoparticles determine the chemical properties and reactivity. Modern research focus on method development in atomically monodisperse nanoparticle synthesis. This dissertation describes the method development for synthesis and isolation of atomically precise gold and gold-silver alloy nanoparticles and their comprehensive characterization and atomic structure investigation. Chapter one offers an introduction to the synthesis and isolation methods. These nanoparticles can be represented in the form of Aux(SR)y, where SR is the thiol ligand. The co-reduction method was used for alloy nanoparticle synthesis using a fixed total metal molar ratio. After considering various possible elements, silver were selected to study the formation of alloys to atomically precise gold nanoparticles. Chapter two includes a discussion of the characterization methods used in the nanoparticles community including scanning transmission electron microscopy, X-ray techniques, UV-visible spectroscopy. Alloying provides a way to tune the properties of materials which is very different from those of their monometallic counterpart. Chapter 3 describes such an effect on Au25(SR)18 gold nanomolecules by alloying with silver. The atomic arrangement of Au and Ag atoms in Au25-xAgx(SR)18 was determined by X-ray crystallography and it was found that Ag atoms were specifically localized in the 12 vertices of the icosahedral core. Among ultra-small nanomolecules, Au38(SR)24 is one of highly attractive nanomolecules due to high stability, availability of single crystal X-ray structure, unique spectroscopic features and intrinsic chirality. Chapter 4 highlights alloying effect on Au38(SR)24 nanomolecules. Chapter 5 describes the modulation of chemical and physical property of Au144(SR)60 by silver doping. UV-visible spectroscopy shows the Ag incorporation affects the electronic structure of the nanomolecules. The maximum number of Ag atoms substitute found to be 60. Chapter 6 describes the first composition determination of super-stable plasmonic nanoparticles in the 2 nm (or 76.3 kDa mass region) and its alloying. This atomically monodisperse plasmonic molecule contains exactly 329 gold atoms and 84 ligands. Apart from the mass spectrometric composition, further characterization was conducted using scanning transmission electron microscopy equipped with high angle annular dark field imaging (HAADF-STEM), high energy X-ray based atomic pair distribution function (PDF) analysis and small angle X-ray scattering (SAXS). Chapter 7 discusses the two largest nanocrystals produced at 2.4 and 2.9 nm with a composition of Au500±10(SR)120±3 and Au940±20(SR)160±4. Most importantly, we were able to successfully extend the mass spectrometric window up to 200 kDa for compositional determination and to study the molecular nature of nanocrystals.Chapter 8 highlight the contributions made from this dissertations toward the advancement of thiol protected nanoparticle research. Book excerpt: The diversity of nanoparticle shapes generated by condensation from gaseous matter reflects the fundamental competition between thermodynamic equilibration and the persistence of metastable configurations during growth. In the kinetically limited regime, intermediate geometries that are favoured only in early formation stages can be imprinted in the finally observed ensemble of differently structured specimens. Here we demonstrate that single-shot wide-angle scattering of femtosecond soft X-ray free-electron laser pulses allows three-dimensional characterization of the resulting metastable nanoparticle structures. For individual free silver particles, which can be considered frozen in space for the duration of photon exposure, both shape and orientation are uncovered from measured scattering images. We identify regular shapes, including species with fivefold symmetry and surprisingly large aspect ratio up to particle radii of the order of 100 nm. Our approach includes scattering effects beyond Born's approximation and is remarkably efficient--opening up new routes in ultrafast nanophysics and free-electron laser science.

We report a novel two-step method for the synthesis of uniform quasi-spherical and monodisperse gold-silver alloy nanoparticles with diameters around 80 nm. Our method makes use of gold acetate (rather than chloroauric acid) as a gold precursor to avoid the formation of silver chloride precipitates during synthesis. Gold and silver are distributed uniformly within the cores of all Au-Ag alloy nanoparticles, while the surface is enriched in silver. This silver enriched surface layer is thicker and more prominent for 25% Au alloy nanoparticles than for 50% and 75% Au alloy nanoparticles. Inspired by previous work studying the mechanism of photo-mediated triangular silver nanoprism growth, where the oxidative dissolution of small silver particles was facilitated by Bis(p-sulfonatophenyl)phenylphosphine dihydrate dipotassium (BSPP), we investigate silver etching of our alloy nanoparticles. Our measurements show that a novel nanostructure is formed after etching where smaller nanoparticles are deposited on the surface of gold-silver alloy nanoparticles.Finally, we present some preliminary results of Rayleigh scattering measurements on our synthesized gold-silver alloy nanoparticles with dark-field hyperspectral microscope at the individual nanoparticle level. In allen Fällen wurden die Silberionen durch im System enthaltene Komponenten reduziert; es wurden keine zusätzlichen Reduktionsmittel verwendet. Insgesamt wurden vier unterschiedliche Systeme als mögliche Template für die Nanopartikelherstellung untersucht. Zwei unterschiedliche synthetische Strategien wurden verfolgt. Zum einen wurden die Nanopartikel mittels chemischer Reduktion der Silberionen durch die nichtionischen oder zwitterionischen Tensidmoleküle, die die Flüssigkristallphase bilden, synthetisiert. Zum anderen wurden die Nanopartikel durch UV-Bestrahlung von DNA-haltigen Flüssigkristallen hergestellt, wobei DNA als Templat und als Photosensibilisator dient. Dabei wird im ersten Stadium ein Ag-DNA-Komplex gebildet, indem Silberionen in die Doppelhelix eingebettet werden. Bestrahlung dieses Komplexes mit UV-Licht, das von den Nukleobasen absorbiert wird, induzierte die Bildung von Nanopartikeln innerhalb der DNA-Doppelhelix. Der Einfluss der Bildung der Silbernanopartikel auf die lyotrop-flüssigkristalline Phase wurde mit 2H- und 31P-NMR-Spektroskopie, Kleinwinkelröntgenstreuung und Polarisations-mikroskopie untersucht. Der Einfluss des Mesophasentemplats auf die Bildungsgeschwindigkeit sowie die Nanopartikelgröße und -morphologie wurde durch UV-Spektroskopie, Transmissionselektronenmikroskopie und Atomkraftmikroskopie untersucht. - This thesis is focused on the synthesis of silver nanoparticles in various lyotropic liquid crystalline phases formed by biologically friendly surfactants with or without dodecyltrimethylammonium-DNA complex in aqueous solution with different additives. In all cases, silver ions were reduced by components contained in the system, no additional reductants were employed. In total, four different systems have been investigated as potential templates for the nanoparticle preparation. Two different synthetic strategies have been used. Firstly, nanoparticles were synthesized by the chemical reduction of silver ions by the nonionic or zwitterionic surfactant molecules which were used to form the liquid crystalline phase. Secondly, nanoparticles have been synthesized by UV irradiation of DNA-containing liquid crystals where DNA acts as templating and photosensitizing agent. At the first stage, an Ag-DNA complex is formed by embedding silver ions into the double helix. Illumination of this complex with UV light that is absorbed by the nucleobases induced the formation of nanoparticles within the DNA double helix. The effect of the formation of silver nanoparticles on the lyotropic liquid crystalline phase was investigated by 2H and 31P NMR spectroscopy, small angle X-ray scattering, and polarizing optical microscopy. The influence of the template mesophase on the formation rate and nanoparticle size and morphology was studied using UV-spectroscopy, transmission electron and atomic force microscopy techniques.

The self-assembly of gold nanorods to form 3-dimensional nanostructures was examined. While the self-assembly was not successful, the aggregated gold nanorods exhibited alarge SERS enhancement. This suggests that the incorporation of gold nanorods intoSERS substrates is a viable avenue for future research.

The self-assembly of gold nanorods to form 3-dimensional nanostructures was examined. While the self-assembly was not successful, the aggregated gold nanorods exhibited alarge SERS enhancement. This suggests that the incorporation of gold nanorods intoSERS substrates is a viable avenue for future research.

Book excerpt: Following a 6-h inhalation exposure to aerosolized 20 and 110 nm diameter silver nanoparticles, lung tissues from rats were investigated with X-ray absorption spectroscopy, which can identify the chemical state of silver species. Lung tissues were processed immediately after sacrifice of the animals at 0, 1, 3, and 7 days post exposure and the samples were stored in an inert and low-temperature environment until measured. We found that it is critical to follow a proper processing, storage and measurement protocol; otherwise only silver oxides are detected after inhalation even for the larger nanoparticles. The results of X-ray absorption spectroscopy measurements taken in air at 85 K suggest that the dominating silver species in all the postexposure lung tissues were metallic silver, not silver oxide, or solvated silver cations. The results further indicate that the silver nanoparticles in the tissues were transformed from the original nanoparticles to other forms of metallic silver nanomaterials and the rate of this transformation depended on the size of the original nanoparticles. Furthermore, we found that 20 nm diameter silver nanoparticles were significantly modified after aerosolization and 6-h inhalation/deposition, whereas larger, 110 nm diameter nanoparticles were largely unchanged. Over the seven-day postexposure period the smaller 20 nm silver nanoparticles underwent less change in the lung tissue than the larger 110 nm silver nanoparticles. In contrast, silica-coated gold nanoparticles did not undergo any modification processes and remained as the initial nanoparticles throughout the 7-day study period.
