Book excerpt: High-Performance Liquid Chromatography of Proteins and Peptides contains the proceedings of the first International Symposium on High-Performance Liquid Chromatography of Proteins and Peptides, held in Washington, D.C., on November 16-17, 1981. The symposium focused on the use of high-performance liquid chromatography (HPLC) in the analysis, characterization, and isolation of peptides and proteins and encompassed six sessions covering size exclusion, ion exchange, and reversed phase chromatography, as well as the use of high-performance liquid chromatography (HPLC) in protein structural studies and peptide isolation. This book is comprised of 28 chapters and begins with a discussion on the status of high-performance ion-exchange chromatography of proteins, followed by an analysis of peptic fragmentation of human immunoglobulin G using HPLC. The physicochemical basis of peptide retention with chemically bonded hydrocarbonaceous silicas and the isolation of biologically active peptides from tissue extracts are also examined. Subsequent chapters explore some additional applications of HPLC, such as cord blood screening for hemoglobin disorders; purification of commercial trypsin and chymotrypsin; characterization of human alcohol dehydrogenase isoenzymes; and structural studies of neurophysins, photolabeled derivatives, and biosynthetic precursors. This monograph should be of value to students and researchers interested in the use of HPLC to study proteins and peptides.

Available in PDF, EPUB and Kindle. Book excerpt: This volume represents the proceedings of the second international meeting on chiral separations held at the University of Surrey between the 12th and 15th of September 1989. Like the preceding meeting, it was jointly organised by the Chromatographic Society and the Robens Institute of the University of Surrey in response to the continued interest in this area of separation science. Of particular interest to the organisers was the very clear change in the nature of the delegates attending this second symposium as compared with the first. At the previous meeting the majority of the delegates were composed of chromatographers with problems in the area of chiral separations who were keen to learn as much as possible about these techniques from the handful of recognised experts in this area. In this second symposium the divide between expert and novice was much less apparent, with the latter providing many interesting and useful contributions to the scientific programme in termt; of both oral and poster presentations.

Available in PDF, EPUB and Kindle. Book excerpt: Explains why modern supercritical fluid chromatography (SFC) is the leading "green" analytical and purification separations technology. Modern supercritical fluid chromatography (SFC) is the leading method used to analyze and purify chiral and achiral chemical compounds, many of which are pharmaceuticals, pharmaceutical candidates, and natural products including cannabis-related compounds. This book covers current SFC instrumentation as it relates to greater robustness, better reproducibility, and increased analytical sensitivity. Modern Supercritical Fluid Chromatography: Carbon Dioxide Containing Mobile Phases covers the history, instrumentation, method development and applications of SFC. The authors provided readers with an overview of analytical and preparative SFC equipment, stationary phases, and mobile phase choices. Topics covered include: Milestones of Supercritical Fluid Chromatography; Physical Properties of Supercritical Fluids; Instrumentation for SFC; Detection in SFC; Achiral SFC Method Development; Chiral SFC Method Development; and Preparative Scale SFC. The book also includes highlights of modern applications of SFC in the final chapters—namely pharmaceuticals, consumer products, foods, polymers, petroleum-related mixtures, and cannabis—and discusses the future of SFC. Provides a clear explanation of the physical and chemical properties of supercritical fluids, which gives the reader a better understanding of the basis for improved performance in SFC compared to HPLC and GC Describes the advantages of SFC as a green alternative to HPLC and GC for the analysis of both polar, water-soluble, and non-polar analytes Details both achiral and chiral SFC method development, including modifiers, additives, the impact of temperature and pressure, and stationary phase choices Details why SFC is the premier modern preparative chromatographic technique used to purify components of mixtures for subsequent uses, both from performance and economic perspectives Covers numerous detectors, with an emphasis on SFC-MS, SFC-UV, and SFC-ELSD (evaporative light scattering detection) Describes the application of SFC to numerous high-value application areas Modern Supercritical Fluid Chromatography: Carbon Dioxide Containing Mobile Phases will be of great interest to professionals, students, and professors involved in analytical, bioanalytical, separations science, medicinal, petroleum, and environmental chemistries. It will also appeal to pharmaceutical scientists, natural-product scientists, food and consumer-products scientists, chemical engineers, and managers in these areas.

Available in PDF, EPUB and Kindle. Book excerpt: Proteins are the servants of life. They occur in all component parts of living organisms and are staggering in their functional var- ty, despite their chemical similarity. Even the simplest single-cell organism contains a thousand different proteins, fulfilling a wide range of life-supporting roles. Their production is controlled by the cell’s genetic machinery, and a malfunction of even one protein in the cell will give rise to pathological symptoms. Additions to the total number of known proteins are constantly being made on an increasing scale through the discovery of mutant strains or their production by genetic manipulation; this latter technology has become known as protein engineering. The in vivo functioning of proteins depends critically on the chemical structure of individual peptide chains, but also on the detailed folding of the chains themselves and on their assembly into larger supramolecular structures. The molecules and their fu- tional assemblies possess a limited in vitro stability. Special methods are required for their intact isolation from the source material and for their analysis, both qualitatively and quantitatively. Proteins are also increasingly used as “industrial components,” e.g., in biosensors and immobilized enzymes, because of their specificity, selectivity, and sensitivity. This requires novel and refined proce- ing methods by which the protein isolate can be converted into a form in which it can be utilized.
