Book excerpt: The advent of increasingly affordable next-generation sequencing technologies are key to understanding the role dispersal, drift and selection play in producing signals of population divergence in many marine species. In particular, dispersal has been difficult to track in the oceans, obscuring its contributions to population structure. The dispersing larval phase of the flat porcelain crab, Petrolisthes cinctipes, exhibits behaviors that likely restrict its along-shore displacement despite a rather long development time at sea. In this work, I attempted to resolve previously detected chaotic patterns among P. cinctipes populations with a published Bayesian model showing that a substantial proportion of larvae likely recruit back to their natal populations. I used a large SNP (single nucleotide polymorphism) dataset to (1) analyze 159 adult crabs from 15 populations for patterns of genetic divergence and (2) directly assign 71 returning larvae back to their population of origin. Admixture results and discriminant analysis of principal components showed a strong signal of panmixia across northern California, making larval assignment impractical. Markers putatively under selection revealed the only clear break in the sampling range—between the southernmost population (just south of San Francisco Bay) and the remaining populations to the north. Despite this panmixia, P. cinctipes population genetics is also characterized by weak chaotic patterns, where populations at close proximity may be more diverged than those separated by hundreds of kilometers. Furthermore, we have previous evidence that such patterns change year-to-year. Underlying this phenomenon might be finer-structured processes that produce weak and ephemeral signals of divergence. A handful of recent studies have shown that cohesive dispersal of related individuals may provide a mechanism for generating such signals. I test for the possibility that cohesive dispersal of related larvae may contribute to observed chaotic patterns in Petrolisthes cinctipes because larval crabs exhibit clustered association with conspecifics throughout their dispersal, settling onshore gregariously in age-unified cohorts. These aspects of its life history point to the possibility of long-term kin association. Using a bank of 1,268 SNPs, I characterized the pairwise relatedness between individuals as well as within-group average relatedness of five settling cohorts of post-larvae collected in 2014. No pair exhibited relatedness greater than would be expected among non-relatives, and group relatedness measurements did not differ significantly from zero. It may be that P. cinctipes larvae are unable to maintain cohesion due to the extreme turbulence of the nearshore and surfzone environments. These same barriers to cohesive delivery of siblings may also be key to our understanding of variation in recruitment in this shore crab. In the course of my genetic sampling efforts among 14 sites across three years, I uncovered a pattern of little to no recruitment in a 100 km section of the Sonoma-Mendocino County coastline. This “recruitment gap” persisted for the years 2012 and 2013, disappearing in 2014 and leading me to wonder what might have driven the spatiotemporal variation. I measured a number of potential drivers at each site, obtained regional and basin-scale indices thought to relate to variation in recruitment. All predictor variables were used in a regression tree analysis that indicated local effects of surfzone width and the proportion of each site’s exposure to northwest compass direction. The temporal variation among years was attributed to patterns of relatively high temperature in 2014, though this result is confounded by the limitations of only three years of data. Despite the clear implication of site-level variables as drivers of the recruitment patterns, I discuss the results in the context possible influences across spatial and temporal scales, and highlight the ways in which knowledge of porcelain crab biology enriches our ability to infer a recruitment variability narrative in this system.

Book excerpt: Organismal tolerance to abiotic environmental stresses contributes significantly to setting the distribution limits of organisms, as demonstrated by vertical zonation patterns in the marine intertidal zone. In this thesis, the ultimate (evolutionary) and proximate (mechanistic) causes of tolerance to temperature and emersion stresses associated with the intertidal zone were examined using porcelain crabs, genus Petrolisthes. Species of Petrolisthes from intertidal and subtidal microhabitats of four biogeographic regions of the Eastern Pacific were used in phylogenetically-based comparative analyses of morphological, physiological, and biochemical adaptation to environmental stress. A phylogenetic tree based on the sequence of the 16sRNA gene was developed to facilitate these analyses. Organismal thermal tolerance limits are adapted to match maximal microhabitat temperatures. Acclimation of thermal tolerance limits suggests that temperate intertidal zone species are living close to their thermal maximum in nature. Respiratory responses to emersion vary among species from different vertical zones. Experimental examination of oxygen consumption rates and lactate accumulation during emersion suggests that intertidal species are able to respire in air using thin membranous regions on the ventral meral segments of their legs (leg membranes). Leg membrane size is positively correlated with body size across species, but not within a single species. Evolutionary analyses indicate that leg membranes may not have evolved for purposes of aerial respiration, but their presence may have allowed intertidal and subtidal species to achieve larger body sizes and higher metabolic rates. The thermal stabilities of an enzyme, lactate dehydrogenase (LDH), from 22 species of Petrolisthes varied widely, but were not correlated with maximal habitat temperatures. Comparative analyses did not indicate any evolutionary relationship between LDH thermal stability and microhabitat conditions. Experimental evidence suggests that interspecific differences in LDH stability are genetically based, and are due both to intrinsic properties of the LDH molecules and extrinsic protein stabilizers. Elucidation of the mechanism(s) of LDH stabilization in Petrolisthes may provide novel insight to the field of protein stabilization. These results studies suggest that individual traits may be subjected to differing levels of selection, and thus the analysis of environmental adaptation requires careful consideration of the biological significance of the traits being examined.

Book excerpt: The green porcelain crab, Petrolisthes armatus, is a highly abundant invader of intertidal oyster reef habitats in Atlantic waters of the southeastern US. Cold water temperatures likely limit the species' poleward range, which is predicted to shift as water temperatures rise. I established five study locations along the species' poleward expansion front from Savannah, GA (Skidaway Island) to Wilmington, NC. Trends in P. armatus populations and resident crab assemblages were assessed seasonally at each location, and live crabs were collected for cold tolerance incubation trials. The taxonomic composition of crab assemblages differed between locations within each season, with obvious dissimilarities between northern and southern locations in the summer and fall, primarily driven by differences in P. armatus abundance. Abundances were highest in the summer and decreased from southern to northern locations. Sex ratios varied seasonally; becoming male biased in the fall. Adult-sized crabs were collected in the spring, indicating that overwintering of adults does occur. Chronic cold temperature trials were conducted seasonally to expose crabs from each location to decreasing water temperatures and assess sublethal and lethal effects. Crabs from northern locations were more tolerant of cold water temperatures in the fall, possibly due to increased acclimatization plasticity present in individuals from northern populations in response to a comparatively colder environment. Females were more tolerant than males in the summer and spring, possibly due to differences in male and female reproductive stressors. Smaller crabs were more tolerant than larger crabs regardless of season indicating molt cycle may play a role in cold tolerance. Given the trend of warming ocean waters, which may move thermal barriers to P. armatus range expansion to higher latitudes, and apparent higher cold tolerance in poleward populations, there is potential that this species will continue its poleward range expansion.

Book excerpt: The study of thermoregulation in endotherms has contributed much to the emergence of the concept of control theory in biology. By the same token, the study of tempera ture adjustment in ectotherms is likely to have a far-reaching influence on ideas on the regulation of metabolism in general. The reason for this is that ectotherms, in adapting to the vagaries of a thermally unstable environment, deploy a range of subtle molecular and organismic strategies. Thus the experimenter, using temperature changes as a tool, is well equipped to analyze some of these strategies. This approach has enabled some important mechanisms of temperature-induced adaptation to be elucidated; the most striking of these are the effects on metabolism of changes in the conformation of enzymes and the transfer properties of membranes. Furthermore, there is a vague but persistent feeling among those working in this field that changes in the nervous system will ultimately prove to be the agency by which many of the molecular mechanisms of temperature adaptation are controlled. Should this indeed be the case, a new phase would soon begin in our understanding of the interactions between the systemic and the cellular levels of organization. However, it is not only questions about the causes of temperature adaptation that can provide answers of potential importance to the general biologist; of equal significance are questions as to the meaning of temperature adaptation in a particular organism.
