Book excerpt: Sex determination refers to the process by which cues from genes (genotypic sex determination) or environmental conditions (environmental sex determination) trigger bipotential gonads to develop into either ovaries or testes. Temperature-dependent sex determination (TSD) is a well-studied form of environmental sex determination in reptiles. The red-eared slider turtle (Trachemys scripta elegans) exhibits a form of TSD where cool incubation temperatures produce males and warm incubation temperatures produce females. In species with TSD, gonadal differentiation is regulated by network of genes and hormones that induce male or female development. TSD is most frequently studied in the laboratory using constant male and female incubation temperatures, which fail to capture the variability that organisms experience in the wild. We use ecologically relevant laboratory incubations to better understand the underlying mechanisms organizing the sex-determining process in T. scripta. Fluctuating incubations and exogenous hormones are used to produce ecologically relevant expression profiles for sex-determining genes and to decouple the effects of temperature and hormone environment on gonadal differentiation. This research represents the first time that fluctuating temperatures and hormones have been used in concert to characterize the molecular mechanisms underlying TSD and highlights the value of using laboratory conditions that more closely mimic environmental conditions.

Book excerpt: In turtles and numerous other reptilian species, sex is determined by the environmental temperatures during embryonic development, which is referred to temperature-dependent sex determination (TSD). In the type of TSD common in turtles, cool temperatures (below 29 ℗ʻC) during embryonic development produce mainly males while warm temperatures (above 29 ℗ʻC) produce mainly females. Gonadal differentiation in TSD species involves numerous genes that are also necessary to produce a testis or ovary in species with genotypic sex determination. However, most of what we know about TSD comes from studies performed in the laboratory using constant temperature incubation conditions, which are not reflective of natural temperatures that fluctuate daily and/or seasonally. As climate change models predict that heat waves will increase in length, frequency, and intensity as climate change progresses, it is of utmost importance to understand how more ecologically relevant incubation conditions affect thermally sensitive species, such as species with TSD. My research with the red-eared slider turtle (Trachemys scripta) focuses on how more variable incubation conditions, such as heat waves varying in timing or continuity, affect sex determination and the expression of underlying genes. Results show that when heat waves, coupled with fluctuating temperature regimes, were applied during the middle third of development it resulted in female-biased sex ratios, while heat waves applied before or after this period resulted in male-biased sex ratios. Results also show that continuous heat waves produce more females compared to the same amount of heat exposure broken up in a discontinuous fashion. More continuous heat waves also induce expression of Cyp19A1, a gene involved in ovarian development while discontinuous heat waves do not. My research also shows that expression of Kdm6b, a gene involved in testicular development, is significantly lower under fluctuating, cool temperatures compared to constant, cool temperatures. Lastly, results show that exposing embryos to conflicting thermal cues delays commitment to gonadal fate, in that early heat exposure causes embryos to stay sensitive to heat exposure longer into developmental stages that are later than those traditionally considered the thermosensitive period. These results highlight the importance of studying how temperature affects biological processes using ecologically relevant temperatures; using constant temperatures ignores natural variability in temperature. Understanding of how biological processes operate under natural conditions is critically needed, particularly in the face of a rapidly changing climate.

Book excerpt: Red-eared sliders (Trachemys scripta elegans) are considered one of the most abundant and successful invasive turtle species in the world. Currently, red-eared sliders are known to have reproducing populations on every continent with the exception of Antarctica. In areas where they have become established, red-eared sliders often outcompete and in some areas extirpate native turtle fauna. A key adaptation recognized in red-eared sliders is their ability to survive a wide range of environmental temperatures. Across their native range, the turtles are noted to survive long-term winter freezing temperatures, as well as prolonged exposure to summer temperature extremes. Although temperature plays an important role in shaping red-eared slider ecology and distribution, relatively little is known about their thermal requirements. This study is the first to use Critical and Chronic Thermal Methodology to define the red-eared slider's thermal niche. The chronic maximum temperature was 43°C ± 0.3, while the chronic minimum was 18°C ± 0.3. Critical Thermal Maximum increased in correlation to acclimation temperature, 41.8°C ± 0.3 at 15°C, 43.2°C ± 0.3 at 22°C, 44.1°C ± 0.5 at 28°C, and 45.7°C ± 0.2 when acclimated to 35°C. Critical Thermal Minimum also increased in relation to the acclimation temperature, -0.04°C ± 0.3 at 15°C, 2.7°C ± 0.1 at 22°C, 3.3°C ± 0.3 at 28°C, and 5.1°C ± 0.2 when acclimated at 35°C. Using the results of this study, the thermal polygon of red-eared sliders was able to be quantified and paves the way for future thermal studies in this species.

Despite dramatic differences in the mechanisms by which different taxa determine the initial sex of offspring, each group has found its own way of adjusting offspring sex ratios in response to social and environmental cues. For example, stress is a well-known modulator of offspring sex in members of all groups studied to date. Food availability, and limitation in particular, is another common cue that stimulates biases in offspring sex ratios in a wide variety of species. Offspring sex can be adjusted at the primary level, which occurs prior to conception, or at the secondary level, during embryonic development. While the mechanistic pathways that ultimately result in sex ratio biases and the developmental time-points sensitive to those mechanisms likely differ among taxa, the key involvement of steroid hormones in the process of sex ratio adjustment appears to be pervasive throughout. This book reviews the systems of sex determination at play in different vertebrate groups, summarizes the evidence that members of all vertebrate taxa can facultatively adjust offspring sex, and discusses when and how these adjustments can take place.
