Molecular biology approaches have made a large contribution to our current understanding of how mechanical loading can alter gene expression and muscle protein synthesis rates in skeletal muscle. The steady-state level of a given mRNA is a function of both its rate of synthesis and its rate of degradation, implying that the rate of change in response to a cellular stimulus is dependent on the dynamics of mRNA turnover. Changes in muscle mRNA steady-state levels can be taken as an index of changes in gene expression, constituting currently one of the best described and understood molecular events that underlie muscle plasticity. The discovery of new growth factors involved in the regulation of muscle development provides a better understanding of the molecular mechanisms involved in the adaptation of skeletal muscle to exercise training. Since it has been shown that changes in contractile function can be brought about by switching on one subset and repressing another subset of genes, it was hypothesized that the steady state level of multiple mRNAs (growth factors and metabolic genes) involved in regulatory functions in a muscle which is frequently recruited during exercise is different between endurance trained versus untrained rats. Recent scientific data indicate that myostatin constitutes a limiting factor in normal muscle development playing a crucial role in skeletal muscle hypertrophy. Nevertheless, there are only scarce data so far about the role of myostatin in the exercise-induced skeletal muscle adaptation. The transient changes in regulatory and structural gene expression provide the molecular basis of the adaptability of the skeletal muscle to exercise stimulus. To get an insight into the molecular mechanisms of skeletal muscle adaptation, the main objective of this study was to examine the effect of both the short- and long- term effect of exercise (five day of swimming vs. chronic wheel running) on IGF-I and MSTN (positive and negative skeletal muscle regulators respectively) mRNA contents in male Wistar rats. Book excerpt: "The traditional Finnish steam sauna is theorized to aid skeletal muscle hypertrophy by stimulating Heat Shock Protein 70 (HSP70), which acts as a molecular chaperone to the folding of functional skeletal muscle. The aim of this study was to investigate if stimulating HSP70 by using a sauna (45 - 50 ͦC, 80% Humidity) three times per week, for 15 minutes, could aid skeletal muscle hypertrophy during six weeks of resistance training in a young (21.38 ± 1.9 yrs.), recreationally trained male population. Thirteen subjects were randomly distributed into 3 groups [resistance Training + sauna (RT+S, n=5), RT + Relaxation (RT+R, n=5), and complete control (CON, n=3) or no training]. Primary dependent variables, observed in a pre- and post-test format, included: lean body mass (LBM), HSP70 concentration, and a 5 repetition maximum (5RM) back squat. When comparing groups (i.e., RT+S, RT+R, and CON), no significant main effects or interactions were observed (p > 0.05) over the 6-week intervention period for LBM, HSP70, and 5RM. The hypothesis that HSP70 would be upregulated to a greater extent with concurrently larger LBM and 5RM improvements in RT+S vs. the other groups was not supported. Interestingly, although HSP70 and LBM were highest in RT+S after 6-weeks of heavy resistance training, RT+R improved the most on 5RM. Sauna use in combination with resistance training does not appear to augment muscle hypertrophy or strength."--Abstract.

Book excerpt: In addition to suppressing protein synthesis, 5'-AMP-activated protein kinase (AMPK) stimulates muscle protein degradation through forkhead box transcription factor 3A (FOXO3A), known to promote transcription of mRNAs encoding degradation pathway proteins. Phosphorylation of FOXO3A at Ser318/321 by Akt (protein kinase B, known to promote muscle growth), normally prevents FOXO3A action by preventing its translocation to the nucleus. However, AMPK inhibits FOXO3A Ser318/321 phosphorylation by Akt, thus allowing its translocation and consequently transcription and upregulation of degradation pathways. AMPK phosphorylation as well as degradation pathways are more greatly elevated in old vs. young rats and humans in response to muscle loading. Moreover, older individuals exhibit lower muscle glycogen content, a condition known to accentuate AMPK activity at rest and during aerobic exercise. However, the possible relationship between muscle glycogen content, AMPK activation, and FOXO3A phosphorylation in response to resistance exercise in young and old individuals has not been previously examined. We hypothesized that FOXO3A (Ser318/321) phosphorylation would be lower in response to acute resistance exercise in the skeletal muscles of older versus younger individuals. It was further hypothesized that this lower FOXO3A phosphorylation response would be related to lower glycogen content and higher AMPK activation in the skeletal muscles of older versus younger individuals. Seven young (21.7 + 2.1 yrs) and 11 old (67.0 + 8.6 yrs) subjects performed an acute bout of leg extension resistance exercise. Muscle biopsies were obtained pre-exercise (PRE), immediately post-exercise (0P), 1-hour post-exercise (1P), and 2-hours post-exercise (2P). Glycogen content was measured in muscle samples, as were the phosphorylations (via western blot) of AMPK, acetyl-CoA carboxylase (ACC; a marker of AMPK activity), and FOXO3A. AMPK phosphorylation was significantly increased in old, but not young, subjects immediately post-exercise; however, no differences in the responses of AMPK activity (assessed by phospho-ACC) or FOXO3A phosphorylation between age groups were observed. Nevertheless, amongst all subjects regardless of age, higher glycogen content was associated with lower phospho/total ACC ratio and higher FOXO3A phosphorylation at the PRE, 0P, and 1P time points. Additionally, the FOXO3A phosphorylation response to resistance exercise was significantly greater in subjects selected for high as compared to low pre-exercise muscle glycogen content. These findings suggest that increasing muscle glycogen content may enhance the FOXO3A Ser318/321 phosphorylation response, and thus potentially lessen the muscle protein degradation response, to resistance exercise regardless of age. However, a strong relationship was not seen between AMPK activity (as measured by the phospho/total ACC ratio) and FOXO3A Ser318/321 phosphorylation, suggesting that another glycogen-mediated factor may play a role in influencing the protein degradation pathway following resistance training in younger and older subjects.

Resistance training interventions are not completely effective in stimulating muscle protein synthesis in aged muscle and thus do not completely combat age-related atrophy. Phosphorylation (and thus theoretically activation) of the energy-sensing molecule 5'-AMP-activated protein kinase (AMPK, known to inhibit the muscle protein synthesis pathway) has been shown to be elevated for up to three hours in response to a resistance training bout in the muscles of older, but not younger individuals. Data in rats indicate that, in response to muscle contractions, elevated AMPK activity can accentuate the inhibitory phosphorylation (and thus deactivation) of its downstream intermediate, eukaryotic elongation factor 2 (eEF2, which normally stimulates protein translation and synthesis). AMPK activity is inhibited by high muscle glycogen levels. Interestingly, older individuals exhibit a lower muscle glycogen content compared to younger individuals, which may account for the greater AMPK phosphorylation response to resistance exercise in older individuals. The relationship between muscle glycogen content, AMPK activity, and eEF2 phosphorylation in response to an acute bout of resistance exercise has not yet been examined in young or old individuals. We hypothesized that inhibitory eEF2 phosphorylation would be higher in response to an acute resistance exercise bout in the skeletal muscles of older versus younger individuals. We further hypothesized that this higher eEF2 phosphorylation response would be related to a higher AMPK activation, and that higher AMPK activation would be related to lower glycogen content, in the skeletal muscles of older versus younger individuals. Seven young (21.7 " 0.8 yrs) and 10 old (67.0 " 2.6 yrs) untrained but physically active men and women performed 3 sets of leg extensions at a 10-repetition maximum resistance until failure after an overnight fast. Muscle biopsies were obtained from the vastus lateralis pre-exercise (PRE), immediately post exercise (0P), 1-hour post exercise (1P), and 2-hours post exercise (2P). Glycogen content was measured in muscle samples, as were the phosphorylations, by western blot, of AMPK (Thr172), acetyl-CoA carboxylase (ACC, a marker of in vivo AMPK activity) (Ser79), and eEF2 (Thr56). Muscle glycogen content was significantly lower in the old vs. young subjects at the PRE time point and decreased in response to exercise in both age groups; however, glycogen content decreased to a greater degree in young subjects such that it was equal between young and old at all post-exercise timepoints. As expected, AMPK phosphorylation was significantly increased in the old subjects immediately post exercise, but no such response was noted in the young. However, no age-related differences were observed in AMPK activity as measured by ACC phosphorylation, which was significantly elevated at 0P and 1P in both age groups. Similarly, the eEF2 phosphorylation response (elevated vs. PRE at 0P and decreased vs. PRE at 1P and 2P in both age groups) was also not affected by age. Regardless of age, higher muscle glycogen content was associated with lower AMPK activity (as assessed by phospho-ACC content) at 0P and 1P, and this lower AMPK activity was associated with lower inhibitory phosphorylation of eEF2 at those same timepoints. These findings indicate the possibility that higher muscle glycogen content may result in lower AMPK activation and consequently lower inhibitory eEF2 phosphorylation in response to a resistance training session in the muscles of both younger and older individuals, thereby potentially enabling greater translation elongation, protein synthesis, and muscle growth regardless of age.
