Book excerpt: The Plasma Proteins: Structure, Function, and Genetic Control, Second Edition, Volume I is a systematic account of the structure, function, and genetic control of plasma proteins. Clinical relevance is introduced in terms of principles, with emphasis on human proteins. Animal proteins are also used as examples in some cases. Comprised of nine chapters, this volume begins with a historical background on plasma proteins, along with their nomenclature, characterization, and genetic markers. The primary structure and three-dimensional conformation of plasma proteins are also considered. The discussion then turns to the chemical, physical, and biological properties of various plasma proteins such as serum albumin, lipoproteins, and immunoglobulins. Subsequent chapters deal with protease inhibitors in plasma; purification, physical properties, chemical composition, and molecular structure of transferrin; biosynthesis and metabolism of serum lipoproteins; and physical, chemical, and functional properties of the proteins of the complement system. The final chapter is devoted to ?2-microglobulin, with particular reference to its purification and physical properties; chemical composition and structure; physiological function, biosynthesis, and catabolism; and presence and function in cell membranes. Book excerpt: The Plasma Proteins: Structure, Function, and Genetic Control is a systematic account of the structure, function, and genetic control of plasma proteins. Advances in the characterization and posttranslational modification of plasma proteins are discussed, along with the structure of a variety of plasma proteins such as serum albumin, glycoproteins, and serum lipoproteins, and immunoglobulins. Comprised of six chapters, this volume begins with a review of progress in plasma proteins, focusing on their three-dimensional structure, characterization, and microheterogeneity as well as genetic polymorphism and chromosomal mapping. The second chapter gives a comprehensive summary of the structure of nearly 100 proteins, from serum albumin and glycoproteins to serum lipoproteins, thyroxine-binding proteins, and immunoglobulins. The reader is then introduced to human plasma proteins of unknown function, together with high-resolution two-dimensional electrophoretic mapping. The remaining chapters explore developments in the structural study of carbon hydrates; synthesis, structure, and function of the oligosaccharides of plasma glycoproteins; and the evolution of the vertebrate plasma proteins. This monograph will be of interest to molecular biologists and biochemists.

Book excerpt: The Plasma Proteins, Volume II: Biosynthesis, Metabolism, Alternations in Disease is a 10-chapter text that explores the physiological role and metabolic interrelationships of the human plasma proteins in the normal state and in disease. The first two chapters cover the physical properties, chemical composition, function, methods of analysis of human serum lipoproteins and plasma enzymes. The subsequent chapter considers the normal levels of hormones in plasma or serum and their distribution in the plasma protein fractions. These topics are followed by discussions on the blood coagulation system, the serum proteins in the animal kingdom at maturity and during embryonic development, and the biosynthesis of plasma proteins. The remaining chapters examine the qualitative abnormalities in various plasma proteins. These chapters also discuss the modification in plasma protein synthesis induced by genetic variation. Such alterations are described for albumin, ceruloplasmin, haptoglobin, iron-binding globulin, fibrinogen, antihemophilic globulin, and other blood clotting factors, as well as ?-globulin. Biochemists, physiologists, and medical researchers will find this book invaluable.

Book excerpt: Clinical Biochemistry: Contemporary Theories and Techniques, Volume 2 is a collection of papers that deals with coagulation chemistry, inborn errors of metabolism (IEM), and the biochemistry of aging. One paper explains the biochemistry and clinical importance of lipoprotein-X (LP-X) as a marker for obstructive jaundice and also as a pointer in the deficiency of lecithin-cholesterol acyltransferase (LCAT), a rare inborn error of metabolism. Another paper presents guidelines in determining radioimmunoassay that are used, for example, in identifying enzymes produced by various malignant lesions. One paper reviews the basic molecular events and interactions involved in the blood clotting process and its related systems. To determine inborn errors of metabolism, the investigator can use screening techniques, prenatal diagnosis or therapy, and laboratory procedures related to IEM. To correct errors at the gene level, transgenosis and genetic engineering use recombinant DNA research techniques involving the introduction of a foreign DNA into the host cell. Some examples of IEM are phenylketonuria and hyperphenylalaninemia without PKU. This collection can prove useful for the clinical chemists, endocrinologists, internists, medical practitioners, and investigators involved in research on biochemistry.

Book excerpt: Clinical Biochemistry: Contemporary Theories and Techniques, Volume 1 is a collection of papers that deals with the biochemistry of aging, managerial techniques, the evaluation of kits, and autoimmune diseases. One paper discusses laboratory management for clinical chemists—the administration of people, the application of budgets, the planning process, and the adoption of decision-making strategies. The government has also issued federal legislations such as the "Clinical Laboratory Improvement Act, 1967" and the "National Heath Planning and Resources Development Act of 1974" which are changing the way laboratories are doing business. Another paper describes areas of safety concerns specific to the environment of the laboratory that require technologies not readily available to the laboratory technician. These safety problems concern radioactivity and infectious etiologic agents. Another paper discusses criteria recommendations for kit selection in clinical laboratories, for example, the list issued by the Center for Disease Control and the standards issued by the National Committee for Clinical Laboratory Standards. Another paper explains the uses of mathematics in clinical chemistry, including the application of the Allen Correction, the Henderson-Hasselbalch Equations, empirical curve fitting, standard deviation, standard error. Other papers present guidelines in dealing with autoimmune diseases and in determining specific proteins in plasma, cerebrospinal fluid, and other biological fluids. This collection is suitable for clinic and laboratory administrators and managers, for chemical chemists, and investigators or technicians involved in laboratory work.

Available in PDF, EPUB and Kindle. Book excerpt: Engström-Finean Biological Ultrastructure, Second Edition, looks upon biological phenomena essentially as patterns of physical and chemical events organized in space and time. It attempts to describe the spatial aspects of this organization, emphasizing in particular the fundamental roles of individual types of molecules. It also introduces a minimum background of fundamental structural information and of techniques so as to aid in the appreciation of the significance of experimental data. The plan of the book in the present edition remains unchanged but the tremendous advances of recent years have necessitated changes in emphasis in the treatment of methods and fundamentals of structure and a complete reappraisal of biological ultrastructure. Key topics discussed include methods in ultrastructural research, the principles of molecular structure; the role of proteins, nucleic acids, lipids, carbohydrates, mineral salts; and the role of ultrastructure in biology and medicine.

Available in PDF, EPUB and Kindle. Book excerpt: Plasma Protein Metabolism: Regulation of Synthesis, Distribution, and Degradation covers the concepts concerning the physiological and pathophysiological factors regulating the distribution, degradation, and synthesis of plasma proteins. This book is organized into nine parts encompassing 32 chapters. The first parts present the assumptions and methodology involved in the various in vivo and in vitro techniques that provide insights to protein metabolism. The next parts describe the techniques of protein isolation, characterization, labeling, and mathematical analysis of the data, as well as the methods for directly quantitating protein synthetic rates in nonsteady state conditions. Other parts are devoted to the factors involved in regulating the serum levels of albumin, acute phase reactants, immunoglobulins, clotting factors, complement and hormone-binding proteins. The controlling factors include such general and specific physiological regulators of protein synthesis and catabolism as levels of specific serum proteins, hormonal regulators, variations of temperature and oncotic pressure, antigenic stimulation, and nutritional factors. The remaining parts analyze the pathophysiological factors including disorders of protein synthesis, distribution, exogenous catabolism, and external loss and stress that produce abnormal levels of serum proteins. This book is intended primarily intended to protein chemists and researchers.

Book excerpt: Enzymes in Blood Plasma gives a comprehensive account of the current state of research and practical application of biochemistry and biology, as well as the pathology and clinical aspects of plasma enzymes. Enzymes in blood plasma, for reasons of principle and methods, are not limited to plasma-specific enzymes, which are only briefly mentioned. Emphasis will rather be placed on plasma-nonspecific enzymes, i.e., enzymes of identical properties in plasma and serum. Their action is, for technical reasons, generally determined in serum. The enzymes in plasma will be stressed in this presentation, but the situation in other extracellular spaces and even extracorporeal areas such as urine, excreta, and feces will be included. There exists no basic difference of enzyme function in these biological areas. Many of the findings reported in this volume were obtained with the old units and methods, rather than the International Unit. For this reason, the section dealing with methods includes a table for conversion, which can be used for comparison of data, insofar as these data were obtained under optimalconditions of measurement.
