Book excerpt: Adoption studies have been used in humans to investigate the relative genetic and environmental influences on development. In this study, we compare rhesus macaque infants raised by adoptive mothers to those raised by their biological mothers in order to evaluate genetic and environmental effects on plasma cortisol and ACTH levels during the first 6 months of life. Blood samples were obtained from both infants and mothers once each month and assayed for cortisol and ACTH. Regression analyses showed interindividual stability across the first 6 months of life for both hormones (average r>0.30). To assess genetic and environmental effects, ACTH and cortisol levels in adopted infants (n=38) were correlated with ACTH and cortisol levels from both their unfamiliar biological mothers and their adopted mothers. Using multiple regression to control for sex differences, results showed that both cortisol (average r=0.27, p=0.05) and ACTH (average r=0.42, p=0.009) levels were correlated with levels observed in adopted mothers. Book excerpt: Nursery-reared infants have a variety of behavioral and physiological differences from their mother-reared counterparts, but nursery rearing remains a necessity in some circumstances. The goal of this research was to investigate three early rearing environments of captive rhesus macaques, and to seek ways to improve the welfare of nursery-reared infants. We gave a complex mechanical responsive surrogate to one group of continuously peer-paired infants and compared them to a group with an identical but non-responsive surrogate. We found no significant differences between the two groups, so we combined them into one group (CS) and compared them with a non-surrogate control group (NS). CS infants showed less partner clinging and affiliation, and more foraging and tactile/oral exploration of their home environment than NS infants. In the second study, we used BioBehavioral Assessment to investigate whether CS infants differed from NS infants in personality and coping styles in the absence of their pair-mates and surrogates under a variety of mildly stressful situations. CS infants were rated with a significantly higher level of confidence than NS infants. They also showed a trend towards more exploratory behavior, and significantly higher rates of cooing than NS infants. These studies provide evidence that the exposure to a mechanical surrogate for peer-paired nursery-reared infants can help to create animals that are more confident in their home cages as well as better able to cope with environmental challenges. In the third study, we investigated the behavioral development of indoor mother-reared infants (MRI) and outdoor mother-reared infants (MRO), and found significant differences between the two groups. MRI infants had more frequent tactile/oral exploration, self-play, foraging, scratching, affiliation with mother, rejection by mother, proximity to and non-ventral-ventral contact with mother than MRO infants. MRO infants had more frequent sleep, nursing on mother, retrieval and restraint by mother, away position from and ventral-ventral contact with mother than in MRI infants. This study provided evidence that differences between outdoor mother-reared infants and indoor nursery-reared infants found in other studies may be due not only to a lack of a mother, but also to differences in the indoor versus outdoor environments.

Book excerpt: Adverse experiences that occur during early critical periods of development modify activity of neuroendocrine systems, such as the hypothalamic-pituitary-adrenal (HPA) axis and oxytocin system. This dissertation examines the effects of nursery rearing, an established model of adverse early experiences, on activity of the HPA axis and oxytocin system in infant and adult rhesus macaques (Macaca mulatta). In addition, influence of oxytocin system activity on the HPA axis and behavioral reactivity was examined. In infant monkeys, nursery-rearing was associated with lower HPA axis, yet higher oxytocin system activity, following the acute stress of developmental assessment. Nursery rearing may result in dysregulation of the HPA axis and a lower overall set point of activity. Mother-reared infants demonstrated higher reactivity during the assessment, which likely reflects a protest response to maternal separation and the strong attachment bond formed in mother-reared infants. Higher oxytocin system activity, lower HPA axis activity, and lower behavioral reactivity during separation and testing in nursery reared monkeys support interactions between HPA axis and oxytocin system activity and corresponds with the pattern of rodent neuroendocrine activity in response to an acute stressor. At 8 months of age, infants were weaned and relocated to mixed-rearing social housing. Infants responded with increased HPA axis and oxytocin system activity that did not vary by rearing condition. Although there was no effect of rearing on overall oxytocin system activity, nursery-reared infants that had been reared with a surrogate-mother, versus rearing with peers, had lower hair cortisol concentrations than mother-reared counterparts at the time of weaning, further supporting the view that nursery-rearing confers a lower set point of HPA axis activity. In contrast, neither HPA axis activity, oxytocin system concentration, nor behavioral reactivity as measured by the Human Intruder Test varied as a function of rearing history in adult rhesus monkeys. This dissertation demonstrates that, although nursery rearing is associated with changes in HPA axis and oxytocin system activity in infant rhesus monkeys, early rearing experiences may not exert prolonged effects. Finally, these data provide some support for an interaction between the HPA axis and oxytocin system, although only during early development.

Book excerpt: Diarrhea has been recognized as a health concern for non-human primates in research settings since the early days of primate research. Here we report a systematic examination of multiple risk factors involved in the development of spontaneously developed diarrhea in rhesus macaques through a retrospective, longitudinal cohort study involving two unique datasets -- BioBehavioral Assessment as infants and Social Network Analysis. Significant variation is found in behavioral and health outcomes for non-human primates reared indoors. This population is most at risk for the development of abnormal behaviors including stereotypic and self-injurious behaviors, and for chronic idiopathic diarrhea. In the captive setting, the frequency of diarrhea requiring clinical treatment is high and is often not ascribable to any identifiable pathogen . We found that immune status, rearing history, and sex influenced an animal's risk of diarrhea-related morbidity and mortality, in concordance with previous literature. While previous reports have suggested that rearing by the mother in an indoor environment is preferable to nursery rearing, we found that whether gestation occurred indoors in single or pair housing or outdoors in a large social group better explained the variability in diarrhea rate seen in our study population of indoor-reared monkeys. Individuals with a higher closeness centrality in the aggression network and those with lower frequency of reconciliation following receipt of aggression were more likely to be treated for clinically significant diarrhea during the study. These results suggest that social stress influences diarrhea but is one cause among many. These results confirm the importance of considering variables beyond standard physiological measures when assessing the overall well-being of an animal. Colony management decisions such as gestation and rearing location and social group conformation exert importance influences on animal health.
