Book excerpt: The diversity and variability of macromolecular components that mediate initial microbial attachment to surfaces in ambient seawater is being determined employing immunological techniques. Antisera against microbial adhesion enhancing (MAE) macro-molecules have been raised in chickens and the immunoglobin-G (IgG) fraction utilized for isolation of crude MAE macromolecules from samples of cell free coastal seawater and marine microbial culture media by immunoaffinity chromatography. The objective of the present study is to produce monoclonal antibodies (MAbs) against AE macro-molecules, and use these to chromatographically isolate and purify individual MAE macromolecules from mixtures of such components. Variability and diversity of MAE macromolecules produced by biofouling microorganisms, those found in ambient seawater and on biofouled surfaces will be assessed employing a spectrum of AE MAbs. The specificity of the interactions of the various MAE macromolecules with glass and metallic surfaces will be assessed. Alternations in the physical characteristics of the test surfaces and changes in their susceptibility to microbial biofouling will be correlated with these interactions. (kt).

Book excerpt: An ELIZA method using immune chicken IgG coated plates does not appear to be specific for microbial adhesion enhancing macromolecules (MAEM). Hyperimmune mouse IgG made against MAEM(but not normal mouse IgG) reacts significantly with coated plates in the presence or absence of MAEM antigens. Assays yield titers of approximately 100,000 for the hyperimmune mouse sera over that of normal control. Hyperimmune mouse sera consistently showed this property despite modifications in the assay method. The very high titer of hyperimmune mouse serum suggests that immunization has produced an antibody(ies) against MAEM antigens, however, the assay could not discriminate specific from nonspecific reactions. Efforts will continue to focus on the problem, by examining the effects of D-galactose and other simple sugars on this binding. Elevated salt concentrations will be examined as a means of minimizing non-antigen mediated reactions. Keywords: Hybridoma. (AW).

Book excerpt: Biofouling is a costly problem, and it is encountered in a wide spectrum of technical systems, ranging from the shipping industry, power industry, water purification, automobile industry, paint and pharmaceuticals, to the microelectronics and food industries. Micro- and macroorganisms attach to surfaces and accumulate there, forming biofilms that cause interferences – a fundamentally natural process. Usually, a medical paradigm is applied: kill biofilms and the problem is solved. This leads to excessive biocide use. However, the success of this strategy is very limited; furthermore it leads to equipment damage and environmental pollution. Simply trying to kill the fouling organisms is clearly not seen as a successful strategy while cleaning is put forward as much more important. In this book, strategies to prevent adhesion, to mitigate the extent and effects of biofouling, and to detect and remove fouling layers are presented. Holistic approaches to the fouling process are elaborated, taking into account options such as nutrient limitation, repellent and easy-to-clean surfaces for fouling layer limitation, and replacing biocides with more environmentally friendly methods – in other words: learning how to live with fouling biofilms without suffering the damage they can do.

In particular in environmental biofilms with many species, it is difficult to impossible isolate, separate the various EPS molecules they are encased in and to define which species produced which EPS.
