The body of work presented testifies to a resurgence of interest in the zona pellucida, prompted largely by a renewed appreciation of its unique properties and its important functions during early mam malian development. It is gratifying to realize that the zona pellucida has at last achieved a well-deserved attention. Whereas extracellular coats of nonmammalian eggs have been studied intensively for many years, the zona pellucida by comparison, has until now taken a back seat. Fortunately, the research presented here signals an end to this situ ation. Many of the difficulties inherent in working with relatively small amounts of material have been overcome, paving the way for application of a wider variety of experimental approaches to the zona pellucida by a larger community of investigators. Hopefully, this monograph will serve as a catalyst in this regard. As revealed in these pages, application of contemporary experi mental methodologies has had significant impact on both pure and applied research on the zona pellucida. For example, modem im munological approaches have been used to assess molecular fea tures of zona pellucida structure and function, as well as to evalu ate the zona pellucida as a potential contraceptive target antigen.

Book excerpt: In 1964, the Fertilization and Gamete Physiology Research Training Program (FERGAP) was established at the Marine Biological Laboratories, Woods Hole, Massachusetts. Over the course of the next 12 years, under the directorship of Dr. Charles B. Metz, FERGAP brought together, trained, and inspired a generation of students in reproductive biology from all over the world. As students of C. B. Metz and as FERGAP trainees, we would like to dedicate this collected work on comparative mammalian fertilization to our teacher and mentor, Dr. Charles B. Metz. Like a number of authors contributing to this volume, we have been struck by the significant impact that C. B. Metz and FERGAP had on the development of students of reproductive biology. Applying both the classical and molecular techniques of cell biology and immunology to problems of gamete biology, Dr. Metz emphasized a comparative and analytical approach that was reflected in his own research on fertilization in Paramecia, sea urchins, frogs, and mammals. It is hoped that this volume will serve to stimulate students to discover the myriad of fascinating research problems in gamete and reproductive biology. Bonnie S. Dunbar Michael G. This book was released on 2012-12-06 with total page 163 pages. Available in PDF, EPUB and Kindle. Book excerpt: Fertilization in mammals normally occUrs within the oviduct, where it is relatively inaccessible to study. However, as a result of painstaking research, most of it carried out over the last five years, this barrier to experimentation has been largely overcome by the development of in vitro fertilization techniques for at least 11 different species, including man. The result has been a rapid increase in our knowledge of the physiological and bio chemical mechanisms involved in the fertilization process. The aim of this book, which is an extension of my recent review of cell surface interactions in fertilization (Gwatkin, 1976), is to present a brief, but well documented, account of the new knowledge that has been attained. Although this book deals with mammalian fertilization mechanisms, I have included some recent experiments on am phibian and invertebrate gametes to supplement the mammalian picture. This information is particularly valuable as the rela tively large number of eggs available from these lower forms has advanced our knowledge of certain fertilization mechanisms beyond what is known in mammals. However, in the interest of brevity, I have omitted details of morphology and minor varia tions between species. For these, and other aspects not covered here, the reader is referred to the books of Austin (1965, 1968), vii viii PREFACE Austin and Short (1972), Metz and Monroy (1969), Monroy (1965), Lord Rothschild (1956a), and Zamboni (l971a).

Book excerpt: This volume, and its companion volume on the spermatozoon, provide an authoritative and wide ranging account of the gametes and their reproductive role and function in humans. Acknowledged authorities from around the world contribute a detailed and timely account of the oocyte. The volume starts with an evolutionary perspective before focusing on the molecular and cellular biology of the oocyte and its structure and function. The development and maturation of the oocyte is fully dealt with to include the endocrine and paracrine regulation of ovarian function. Practical issues such as oocyte storage and ovarian stimulation are also fully covered. The causes of female infertility are also an important theme. The volume concludes with a thought provoking chapter on ethical considerations. The volume will be an essential source of information for all clinicians and scientists with an interest in human reproduction.

Available in PDF, EPUB and Kindle. Book excerpt: Methods in Mammalian Reproduction presents some of the techniques for manipulating, analyzing, observing, testing, and generally experimenting with mammalian mothers and their gametes and embryos. Mammalian reproduction involves an intimate relationship between mother and embryo. The first 18 chapters are arranged in an order that follows a developmental sequence from oocyte to fetal organs and the remaining seven chapters deal with the maternal side of the relationship. With strong focus on laboratory rodents and lagomorphs, the book starts with an introduction to in vitro oocyte maturation and experimental production of mammalian parthenogenetic. It goes on to describe the microtechniques in pre-implantation of embryos, production of chimeras, techniques for early embryonic tissue separation, mammalian embryo preservation by freezing, and in vitro development of whole mouse embryos beyond the implantation stage. Chapters 11-15 discuss the in vitro implantation of mouse blastocysts, advances in rabbit embryo and in large mammal embryo cultures, embryo transfer in large domestic mammals, and manipulation of marsupial embryos and pouch young. The following chapters cover reproduction experiments using marsupials, domestic farm species, and primates including humans. Finally, the concluding chapters tackle the use of amniocentesis in prenatal diagnosis, collection and analysis of female genital tract secretions, analysis of antifertility action of intrauterine devices, and surgical induction of endometriosis. This book will be helpful to students, teachers, researchers, and clinical researchers who demand for more and better procedures for analysis of mammalian reproduction.

Book excerpt: Biology of Fertilization, Volume 2: Biology of the Sperm is the second in a three-volume series that brings together various lines of research about reproduction in general and fertilization in particular. It is devoted to spermatogenesis, sperm physiology, and the initial interactions of sperm with egg components. The book is organized into three parts. Part I on spermatogenesis and sperm physiology includes studies on the evolution of the sperm cell; regulatory mechanisms, sperm antigen differentiation, and maturation of sperm in the reproductive tracts in mammals; physicochemical mechanisms of the acrosome filament extrusion process; and enzymes associated with sperm cell function. Part II deals with chemotaxis in microorganisms: bacteria and slime molds. Part III on gamete recognition and binding includes studies on identifying, isolating, and characterizing specific sperm and egg surface components involved in sperm-egg interaction; fertilization in the alga Fucus; fertilization in insects; and the role of lysins in fertilization.

Available in PDF, EPUB and Kindle. Book excerpt: The Oviduct and Its Functions focuses on the role of the oviduct in reproduction. The book emphasizes that this organ combines factors such as environment, hormonal balance, and biochemical constituents to make reproduction possible. Composed of contributions that are divided into 13 chapters, the book presents the comparative anatomy, histology, and morphology of the mammalian oviduct. The selection particularly notes the structural features of this organ. The text proceeds with the discussions on histochemistry and electron microscopy of the bovine oviduct, and then notes that few studies have been reported on this concern. The discussions shift with the presentation of the neural control of the oviduct. Concerns include the presence of alpha and beta receptors in the organ; adrenergic and cholinergic innervation; and the influence of drugs on ovum transport. The text further underscores fluid flow and composition of oviducal fluids in animals. The book points out that the oviduct is vital in the capacitation process and fertilization and responsible for the transport of gametes and survival of sperm. The text also notes that the female reproductive tract is responsible in sperm storage. The book ends with discussions on the influence of genetics, ovulation, and pregnancy on the functions of the oviduct. The compilation is vital for readers interested in studying the functions of the oviduct.

The opening chapters examine the control of glucose transport by insulin, the participation of nuclear poly(adenosine diphosphate-ribosyl)ation in hormonal mechanisms, and the concept of ovulation control. These topics are followed by discussions on the immunohistochemistry of human chorionic gonadotropin; the adrenocorticotropic hormone (ACTH) action on adrenal cholesterol metabolism and its interaction with adrenal receptors; the structure-activity relationships of neurohypophyseal hormones; and the interactions between different functional sites in the epidermal growth factor receptor. A chapter focuses on the juvenile hormone-binding proteins. The remaining chapters consider the actions of steroid hormones. These chapters specifically describe the androgen receptor, the interaction of the glucocorticoid receptors with specific DNA sequences, and glucocorticoid actions on fibroblasts. A discussion on the potential role of RNA in the glucocorticoid receptor mechanism is also included in these chapters. This book will be of great benefit to endocrinologists, biochemists, biologists, and researchers who are interested in hormonal action and regulation.
