Rabbit 775, highly immunized against horse serum, yielded a serum which passively sensitized a guinea pig of 300 grams in amounts of cc. The precipitin tests showed that cc. In 1 cc. Of salt solution gave a distinct precipitate with of horse serum; with larger amounts of horse serum the precipitate was voluminous. One cubic centimeter of Serum 775, added to nine parts of per cent salt solution, was heated for one-half hour at Precipitin tests with 3 cc. Of this heated mixture failed to give any reaction with either of of horse serum, though the tubes were observed several days. That the mere dilution could not be held responsible for this result is abundantly proven by Experiment 6 of this paper. On the day following inactivation, five guinea pigs were given subcutaneous inoculations of the heated mixture, three receiving 1 cc. Each, and the remaining two, each 2 cc. The former group had an amount corresponding to one sensitizing dose, while the latter received two. Two days later all the ani mals were given intravenous injection of cc. Of horse serum. The first group showed symptoms of varying intensity, but none either had convulsions, or died. The two pigs in the second group, however, immediately died. Book excerpt: Excerpt from The Journal of Immunology, Vol. 4 Many attempts have been made to devise a simple and accurate laboratory method for determining the presence of toxic and antitoxic substances in the blood of persons suffering from acute infections of various kinds and in the blood of normal individuals. Several methods adapted for the study of certain toxins have resulted. Many of these in the hands of most workers have been difficult to manipulate. Others have failed to give the qualitative and quantitative data necessary. It has been evident since the beginning of the study of toxins and antitoxins that their presence or absence must be determined by testing their effect upon animal tissues. Other and more accurate methods will probably not be devised until the chemical and physical properties of these substances are better understood. For a long time it has seemed apparent that the tissue culture method might aid very materially in making such determinations. Stimulated by the urgent need of a more careful study of many of the cases of diphtheria and other infections as they have been seen in the hospital and as they are reported from army camps we undertook recently to apply the method in this direction. As is well known the tissue cells of many animals grow readily not only in their own plasma but also in drops of plasma of many other animals. Book excerpt: Excerpt from The Journal of Immunology, Vol. 5 In the year 1901 Neisser and Wechsberg described a peculiar function of immune sera, which they called "Complementablenkung," and which later has been named the "Phenomenon of Neisser and Wechsberg." The phenomenon consisted in the observation that, whereas small or medium doses of bactericidal sera exhibited the bactericidal function with the homologous strain, the larger doses were without effect. Thus an inactivated immune serum against Vibrio Metschnikoff with added complement, showed bactericidal action in doses of 0.05 to 0.0025 cc., but in larger doses it had no such effect. Likewise, Neisser and Wechsberg showed that the dose of complement-bearing serum, which was sufficient to activate a certain dose of an inactivated serum, was not capable of doing so when larger doses of immune serum were added. The explanation of this paradoxical serum function was sought, by the authors, in the great richness of the immune serum in bactericidal amboceptors. These antibodies, which, in the immune serum, must be present in far greater quantities than the complement in the activating serum, were supposed to unite with the complement and form thereby a lysin that dissolves the bacillus after becoming attached to it; but on account of the predominance in the number of the amboceptors, there being insufficient complement to satisfy all of the amboceptors, some of the latter were assumed to remain as unaltered amboceptors, free of complement, and thus devoid of bactericidal function. Book excerpt: Excerpt from The Journal of Infectious Diseases, 1904, Vol. 1 On the other hand, human serum, unlike all other sera, was found to exert a specific action upon the trypanosomes of Nagana in infected mice and rats. The subcutaneous injection of 5 to 1 cc of the human serum into these animals, at the time when the trypanosomes are just beginning to appear in the blood. Causes the parasites to disappear in from twenty-four to thirty-six hours. The blood then remains free from the organisms for four to eight days, exceptionally for twelve, eighteen, or nineteen days, after which they reappear. On subsequent injection of serum they again disappear for a short time. In this way the life of the ani mal may be prolonged, as with injections of arsenite of soda, but eventually death results. In four instances, however, one or two injections of the serum in mice were followed by recovery. More recently (july, 1903) Laveran showed that human serum exerts a similar action upon the trypanosomes of Oaderas and Surra.
