Available in PDF, EPUB and Kindle. Book excerpt: This book overviews cancer immunity from broad scientific fields, based on the concept that cancer is a sort of by-product of infection, inflammation, and host immune response. The innate and acquired arms of the immune system mainly participate in tumor immune surveillance, and their activation is critically modulated by the situation of the tumor microenvironment. Many types of immune cells join the formation of the microenvironment. In particular, macrophages and dendritic cells enter the tumor mass to be main players in the inflammatory milieu of tumors. After introducing these topics, the book discusses immunotherapy for cancer patients as an outgrowth of this concept of infection and inflammation. With the contributions of leading scientists actively involved in the field of antitumor immunity study, this book encourages readers to understand the mechanism of general cancers based on inflammation and will facilitate prevention and the development of therapeutics for cancer.

Book excerpt: A link between inflammation and cancer has been established many years ago, yet it is only recently that the potential significance of this connection has become apparent. Although several examples of chronic inflammatory conditions, often induced by persistent irritation and/or infection, developing into cancer have been known for some time, there has been a notable resistance to contemplate the possibility that this association may apply in a causative way to other cancers. Examples for such progression from chronic inflammation to cancer are colon carcinoma developing with increased frequency in patients with ulcerative colitis, and the increased incidence of bladder cancer in patients suffering from chronic Schistosoma infection. Inflammation and cancer have been recognized to be linked in another context for many years, i.e., with regards to pathologies resembling chronic lacerations or 'wounds that do not heal.' More recently, the immunology of wound healing has given us clues as to the mechanistic link between inflammation and cancer, in as much as wounds and chronic inflammation turn off local cell-mediated immune responses and switch on growth factor release as well the growth of new blood vessels - angiogenesis. Both of these are features of most types of tumours, which suggest that tumours may require an immunologically shielded milieu and a growth factor-rich environment.

Book excerpt: A new perspective on the link between inflammation and cancer Inflammation is the human body’s normal biological response to threats in the modern world, as well as a defense against the harmful influence of pathogens, the environment, and poor nutrition. But what happens when the inflammatory response is triggered repeatedly and sustained for long periods of time? Cancer and Inflammation Mechanisms: Chemical, Biological, and Clinical Aspects discusses the mechanisms by which chronic inflammation can lead to cancer, the various causative agents, and possible prevention methods. A compilation of the latest information coming out of the various fields of cancer research, this book provides a detailed look at inflammation-related carcinogenesis from the perspective of researchers at the forefront of the field. It takes an interdisciplinary approach to the topic, and provides comprehensive information about the major factors at work in inflammation, cancer, and the intersection of the two conditions. Topics include: A general overview of inflammation-related cancer The biochemistry of inflammation and its effects on DNA Molecular biology and the role of microRNA in carcinogenesis Specific causative agents including oncogenic viruses, asbestos, and nanomaterial Anti-inflammatories, nutraceuticals, and other preventative measures A deeper understanding of the mechanisms behind inflammation-related carcinogenesis can lead to better patient outcomes by improving diagnostics and prevention, as well as altering the approach to treatment. Cancer and Inflammation Mechanisms: Chemical, Biological, and Clinical Aspects provides the knowledge base researchers need to push the field forward.

Available in PDF, EPUB and Kindle. Book excerpt: This book is focused on the analysis of the role played by immune cell components in the angiogenic process associated with inflammation and tumor growth. Both innate and adaptive immune cells are involved in the mechanisms of endothelial cell proliferation, migration and activation, through the production and release of a large spectrum of pro-angiogenic mediators. These may create the specific microenvironment that favors an increased rate of tissue vascularization. The link between chronic inflammation and tumorigenesis was first proposed by Rudolf Virchow in 1863 after the observation that infiltrating leukocytes are a hallmark of tumors and first established a causative connection between the lymph reticular infiltrate at sites of chronic inflammation and the development of cancer. Tumors were described as wounds that never heal and surgeons have long described the tendency of tumors to recur in healing resection margin and it has been reported that wound healing environment provides an opportunistic matrix for tumor growth. As angiogenesis is the result of a net balance between the activities exerted by positive and negative regulators, this book will also provide information on some anti-angiogenic properties of immune cells that may be utilized for a potential pharmacological use as anti-angiogenic agents in inflammation as well as in cancer. The work is written for researchers in the field and also for graduate students which approach this matter.

Available in PDF, EPUB and Kindle. Book excerpt: This book, now in a thoroughly revised and updated second edition, provides the latest information on cancer metastasis from the perspective of inflammation and presents new ideas on the complicated mechanisms of metastasis and potential therapeutic targets. Key features include discussion of mechanisms recently identified to be involved in the resolution phase of inflammation, presentation of the latest evidence regarding the roles of endogenous TLR4 ligands in metastasis, and thorough explanation of the concept of homeostatic inflammation and current understanding of the significance of its dysregulation for metastasis. Structure-based thinking is another important element of the book, and it is proposed that inflammation forms a functional triangle with angiogenesis and coagulation, at the center of which cancer is located. Examples of the many additional specific topics covered in this edition include the functional involvement of new types of RNA in cancer, the insights offered by recent advances in bioinformatics, and the potential of a casein kinase 1α inhibitor in the treatment of acute myeloid leukemia. The book will be a valuable update and resource for both experienced and younger researchers. Homeostasis, originated from an idea of internal milieu by Claude Bernard, is eventually maintained by endogenous elements. The essential features of inflammation are leukocyte mobilization and increased vascular permeability, which could take place in many homeostatic or physiological conditions at low levels. Homeostatic inflammation is a concept to explain pathological settings such as metastasis in which irrespective of its level those inflammatory features are misused with endogenous molecules (see Chap. 14,15). As inflammation comprises many biological fields, targeting a single molecule for a disease could potentially make a therapeutic contribution to other diseases. For example, one focus is applied here to the roles of calprotectin in lung metastasis, which is implicated in psychiatric disorders and COVID-19 as shown by recent evidence.

Book excerpt: In recent years there have been various discoveries connecting inflammation and lung cancer and clearly there is growing interest in this area of cancer research. The link between unresolved inflammation and cancer has been well established with estimates that 15% of cancer deaths are inflammation‐related. Evidence for this link includes the following: a) some inflammatory diseases are associated with increased risk of cancer development; b) inflammatory mediators are present surrounding and within most tumors; c) overexpression of inflammatory cytokines increases cancer development and progression in murine studies; d) inhibition of inflammatory mediators decreases cancer development and progression; and e) the use of non‐steroidal anti‐inflammatory drugs (NSAIDs) has been found to decrease cancer incidence and delay progression. The volume will present aspects of the inflammatory tumor microenvironment (TME), its many roles in tumor progression and metastasis, including creation of a hypoxic environment, increased angiogenesis and invasion, changes in expression of micro‐RNAs (miRNAs) and an increase in a stem cell phenotype. The book will also cover the mechanisms of inflammatory mediators. Chronic overexpression of inflammatory mediators in the TME, as seen in smokers and patients with non‐small cell lung cancer (NSCLC), can also lead to increased tumor initiation, progression, invasion and metastasis. The volume will provide a comprehensive perspective of the latest findings and summaries of progress made regarding inflammation and its connection to lung cancer.

Book excerpt: It is now becoming very clear that the development and progression of tumor towards the malignant (metastatic) phenotype depends tightly on the interaction between the tumor cells and the tumor microenvironment. Tumor cells respond to stimuli generated within the tumor microenvironment for their growth advantage while the tumor cell themselves reshape and remodel the architecture and function of their extracellular matrices. The term tumor microenvironment is a wide umbrella consisting of stromal cells such as fibroblasts and endothelial cells and infiltration immune cells including T and B cells, macrophages, and other inflammatory cells (PMNs). These different components of the tumor microenvironment could have stimulatory and inhibitory effects on tumor progression by regulating the gene expression repertoire within the tumor cells on one hand and the stroma cells on the other. In this volume we have seven contributors who will discuss several different aspects on the cross talk within the tumor microenvironment components leading to the acquisition of the metastatic phenotype. It is our hope that these state-of-the-art studies will shed further light on our understanding of these complicated processes.

Book excerpt: Chronic inflammation predisposes to some forms of cancer and the host response to malignant disease shows several parallels with inflammation and wound healing. The cells involved in inflammation are detected in a range of common cancers, together with the inflammatory cytokines and members of the chemokine ligand/receptor systems. Neutralization or deletion of the gene for some inflammatory cytokines confers resistance to tumour induction and experimental metastasis. Over-expression of such cytokines in tumour cells may enhance malignant potential. Certain chemokines are likely to subvert antitumour immunity by favouring development of ineffective Type 2 responses. Tumour cells may even utilize chemokine receptors in homing to lymph nodes and other organs. Thus, the cells, cytokines and chemokines found in tumours are more likely to contribute to tumour growth, progression and immunosuppression than they are to mount an effective host antitumour response. This book draws together contributions from an international group of scientists and clinicians from diverse disciplines, ranging from epidemiology to immunology, cell biology, molecular oncology, molecular medicine and pharmacology to debate these and related issues. Topics covered include the epidemiological links between cancer and inflammation, the parallels between inflammation and cancer, the role of inflammation in cancer, inflammatory genes as risk factors for cancer initiation and progression, inflammation and cancer angiogenesis, and preventative and therapeutic strategies.
