Book excerpt: We are using the feline immunodeficiency virus (FIV)-infected cat to model HIV mother-to-child-transmission (MTCT). Vertical transmission of either virus may result not only in infected offspring, but also failed pregnancy. In HIV infections, maternal hematological and virological parameters predict MTCT. We hypothesized that such parameters would likewise be predictors of FIV vertical transfer. We inoculated ten cats with FIV-B-2542; 10 cats were uninoculated. Cats were allowed to breed naturally. Fetuses were delivered at approximately week 3 (early) gestation by cesarean section. Fetal and placental tissues were collected. Blood samples were collected from the day of inoculation through delivery. We quantified CD4:CD8 T cell ratios, proviral load, and plasma viremia, and monitored seroreactivity to FIV proteins in longitudinal sera from both groups of cats. We documented clinical and reproductive outcome. The infected group produced reduced litter size and more failed pregnancies; CD4:CD8 ratios were depressed by 3.5 months p.i. Proviral DNA was detected in 14 of 14 (100%) placentas tested and 12 of 14 (86%) fetuses. However, the parameters assessed were not predictive of reproductive outcome and suggested a role for placental immunopathology in compromised pregnancy. Regulatory T cells (Treg) are anti-inflammatory and essential in maintaining pregnancy. Th17 cells are pro-inflammatory and associated with pregnancy failure. The activation of these cell populations is regulated by the cytokines TGF-[Beta] and IL-6. We hypothesized that placental immunology may result from altered dynamics of these cell populations. Using immunofluorescence confocal microscopy to measure Treg and Th17 markers FoxP3 and ROR[subscript Gamma], respectively, we quantified these cells in placental specimens from FIV-infected and control cats at early and late (week 8) gestation. Significantly higher levels of ROR[subscript Gamma] were measured in FIV-infected placentas at early pregnancy; these cells co-localized at the maternal-fetal interface. We quantified the expression of Treg immunomodulators by quantitative PCR, noting higher expression of TGF-[Beta] in infected queens. A positive correlation of ROR[subscript Gamma] with IL-6 occurred in control placentas, as predicted, but not in infected placentas. Collectively, the data suggest that an inflammatory placental microenvironment at early pregnancy in infected queens may result, in part, from dysregulation of the Treg/Th17 balance.

Similar to the human, lymphoma is the most common hematopoietic tumor in cat and epitheliotropic intestinal T cell lymphoma has been found to resemble the WHO type II enteropathy-associated T-cell lymphoma in humans. Although, the cat holds potential as an immunological animal model, integrative study of feline T cell was limited, and identification of T cell lineage/subpopulation was restricted due to the lack of specific antibodies. The recent release of a high-quality feline genome assembly coupled with a more approachable high-throughput sequencing technique provide a basis for the studying of antigen receptor gene loci in cats. The research outcome would benefit the use of cats as a model for spontaneous diseases and contribute to feline health. The primary scope of this dissertation research was to broaden the knowledge of feline mucosal immunity by investigating T cell receptor locus structure and expressed repertoire, developing T cell clonalities and analysis of novel lymphocyte populations. The following three objectives were discussed: 1) characterization of the genomic organization and expressed repertoire of the feline T cell receptor alpha/delta (TRA/TRD), beta (TRB) and gamma (TRG) loci; 2) development of clonality assays for feline T cell lymphoma; 3) identification of Fe7.3D12 antibody binding protein expressed on feline intestinal intraepithelial lymphocytes. To study the structure of feline TR genes, we employed a Hidden Markov Model and blastn-based approaches to comprehensively identify TRA/TRD, TRB and TRG germline genes on the Felis_catus_9.0 assembly. High-throughput sequencing was used to access the expressed repertoire in normal lymphoid organs. Gene nomenclature and functionality in this study was approved by the international immunogenetics information system (IMGT). The feline TRG locus contains 7 TRGV, 12 TRGJ and 6 TRGC, the TRB locus contains 32 TRBV, 2 TRBD, 10 TRBJ and 2 TRBC genes and the TRA/TRD locus contains 60 TRAV, 63 TRAJ, 1 TRAC, 11 TRDV, 2 TRDD, 4 TRDJ and 1 TRDC genes. The genomic organization of the feline TR loci was similar to the previously described gnathostomata with the exception of the feline TRG locus which was organized in 5 complete and 1 incomplete V-J-(J)-C cassettes. Functional feline T cell receptor V genes formed monophyletic clades with their orthologs and were subjected to the birth-and-death model of evolution. Expressed repertoire analyses showed a biased V/J usage mainly in the TRG and TRD loci. In addition to the understanding of feline TR gene topology and expressed repertoire benefit to future research on feline medicine and translational studies on immunopathology diseases in human, this data will also facilitate the development and improvement of molecular diagnostic tests for lymphoproliferative disorders, a common diseases in domestic cats. Molecular clonality assays have been used as an adjunctive test for the diagnosis of lymphoma when histopathological assessment is inconclusive. However, the assays occasionally yield false negative results and the target TR gene assessment is restricted only to the TRG locus. To improve the sensitivity of feline T cell clonality assays, we employed knowledge of topology and expressed repertoire of feline T cell receptor loci obtained from previous studies to develop new clonality assays targeting multiple TR loci. Three novel multiplex clonality assays targeting TRG, TRD and TRB loci were successfully optimized and 6 TRG primers, 5 TRD primers and 27 TRB primers were multiplexed in one reaction tube per locus. New multiplex clonality assays and the original singleplex TRG assay were validated on series of neoplastic and non-neoplastic lymphoid tissues. In testing phase, the TRG and TRD primer sets were able to detect rare rearrangement utilized by invariant T lymphocyte population in both normal and diseased livers. The primers which recognized the canonical rearrangement were omitted to avoid false positivity. The invariant T lymphocyte population residing in liver will be further investigated. The multiplex TRG assay yielded the highest sensitivity (95.5%) and the multiplex TRD assay yielded the lowest sensitivity (62.9%). All assays yielded 100% specificity except for the multiplex TRG assay (97.3%). The combination of the multiplex TRG and TRB assays yielded the highest sensitivity (98.9%) and specificity (97.0%). We recommend performing this combination of multiple gene target clonality assays to increase the overall sensitivity and specificity for the diagnosis of feline T-cell lymphomas. Intraepithelial lymphocytes (IELs) are exclusively comprised of T lymphocyte populations that function in both mucosal immunity and inflammation-induced pathology. In the development of antibodies to feline (GlcNAc)6 integrin (CD103), an antibody (Ab) clone designated Fe7.3D12 (3D12) was generated. The IELs expressing the 3D12-binding molecule exhibited potential regulatory function and a high incidence of 3D12 positivity in feline epitheliotropic T cell lymphoma was also observed. This dissertation chapter objective was to identify this molecule for further characterization of IEL subpopulations and potentially elucidate intestinal mucosal immunopathology. In this effort, Protein lysates from feline IELs were immunoprecipitated (IP) using 3D12 conjugated magnetic beads. Mass spectrometry revealed that the best match for the 3D12 binding protein was a previously uncharacterized 30.8 KDa protein (Uniprot: M3X555) containing a predicted transmembrane domain at amino acid residues 56-78 and a predicted C-type lectin domain at amino acid residues 133-274. The M3X555 protein is encoded by an uncharacterized gene (Ensembl: ENSFCAG00000025057) that has 63.0% homology with the killer cell lectin-like receptor family J member 1 (KLRJ, Ly49) gene of Bos Taurus. The M3X555 protein was successfully expressed on HEK293 cells and confirmed by immunofluorescence and WB using 3D12 Ab. The relationship to the Natural Killer (NK) cell gene complex and homology with C-type lectin-like receptors expressed by NK cells and cytotoxic T-cells are indicative of a probable biological significance. Thus, the membrane protein recognized by 3D12 Ab is likely associated with IEL biology and possibly in the pathogenesis of feline intestinal T cell lymphoma. Conclusion of this dissertation research has extended the knowledge of feline mucosal immunology by providing a large-scale dataset of T cell receptor genes and expressed repertoire. This comprehensive data has facilitated the development of clonality assays with high sensitivity that can be used for both the diagnosis of feline T cell lymphomas and the investigation of feline T cell population and neoplasms.
