Book excerpt: Detailed protocols for analyzing DNA strand breaks, specific RNA/DNA expressions, and DNA binding proteins such as transcription factors at the individual cell level. It describes in situ nick translation and TUNEL in detail, along with radioactive and non-radioactive methods for in situ hybridization. Of special significance is the description of Southwestern histochemistry, which makes it possible to analyze the expression of transcription regulatory factors in individual cells. This volume provides an overview of the current status of molecular histochemistry along with practical how-to-do-it details. The methodology is set out in easy-to-follow language by leading scientists working in the field today. Both a rich source of the latest information and a practical lab manual.

Available in PDF, EPUB and Kindle. Book excerpt: During recent years enzyme histochemical reactions have increasingly been considered as important, the reason being that enzyme histo chemistry is now a well-established link between morphology and bio chemistry. The development of numerous new methods and in particular the improvement of existing techniques contributed to the expansion of enzyme histochemical reactions. Today, the use of these methods allows detailed insight into molecular processes of single cells and their constituents. The selection of a suitable method for enzyme histochemical investigations needs thorough knowledge and critical evaluation of the reactions de scribed for the histochemical demonstration of enzymes and introduced in laboratory practice. Often, it is difficult for scientists primarily concerned with the application of methods and for laboratory assistants to comment on the value of an enzyme histochemical reaction. Our book will serve as a guide in this respect. It contains the most important histochemical methods for the localization of enzymes, all of which were checked by the authors themselves. These methods were often modified and frequently used for numerous different investigations of healthy and diseased organs in basic research and in routine practice.

Book excerpt: Somuchofwhatweknowaboutthepathogenesisofhumandiseasehascomefromthe systematic and careful study of histological material. Indeed, every internal medicine discipline has its landmark papers describing the clinico-pathological correlations. However, increasingly, it is molecular and cellular biology that provides the necessary mechanistic insights. For many years, it was thought that the two skill sets were mutually exclusive, but we hope that this book shows that this is not necessarily so. Implicitinthescienceofhistologyisthepreservationandarchivingoftissue.PartIof the book concentrates on the preparation of tissue, providing an overview of fixation, embedding, and processing (Chapter 1), and in Chapters 2 and 3, the required techniques for the retrieval of RNA from histological sections. Both routine and specialist histological staining techniques are provided in Part II. These include pro- cols for immuno (Chapters 4–7), lectin (Chapter 8), and hybridization (Chapter 9) histochemistry, histologicalstaining (Chapters10and11),aswellasspecificmethods for the in situ identification of hypoxia (Chapter 12) and apoptosis (Chapter 13). Finally, Part III details advances in imaging (Chapters 14–16) and image analysis (Chapter 17). It is hoped that this volume will provide molecular biologists with the basic his- chemical techniques and histologists with the molecular techniques to realise the potential of their resource. We are indebted to the authors for their generosity in sharing these protocols.

This book was released on 2003-05-01 with total page 582 pages. Available in PDF, EPUB and Kindle. Book excerpt: Histotechnology and histomorphometry are the major methodologies in bone and cartila- related research. Handbook of Histology Methods for Bone and Cartilage is an outgrowth of the editors’ own quest for information on bone and cartilage histology and histomorphometry. It is designed to be an experimental guide for personnel who work in the areas of basic and clinical bone and cartilage, orthopedic, or dental research. It is the first inclusive and organized reference book on histological and histomorphometrical techniques on bone and cartilage specimens. The topic has not previously been covered adequately by any existing books in the field. Handbook of Histology Methods for Bone and Cartilage has six major parts and is designed to be concise as well as inclusive, and more practical than theoretical. The text is simple and straightforward. Large numbers of tables, line drawings, and micro- or macro-photographs, are used to help readers better understand the content. Full bibliographies at the end of each chapter guide readers to more detailed information. A book of this length cannot discuss every method for bone and cartilage histology that has been used over the years, but it is hoped that major methods and their applications have been included.

Available in PDF, EPUB and Kindle. Book excerpt: The liver is an exceptionally complex and diverse organ that functions both as an exocrine and an endocrine gland. It secretes bile, which contains many con stituents in addition to bile salts, and it synthesizes and releases many substances in response to the body's demands, including prohormones, albumin, clotting factors, glucose, fatty acids, and various lipoproteins. It has a dual blood supply providing a rich mixture of nutrients and other absorbed substances via the portal vein and oxygen-rich blood via the hepatic artery. This functional heterogeneity is accompanied by cellular heterogeneity. The liver contains many cell types including hepatic parachymal cells, Kiipffer cells, Ito cells, and endothelial cells. The most abundant cell type, the parenchymal cells, are biochemically and structurally heterogeneous. The cells in the oxygen-rich areas of the portal triad appear more dependent on oxidative metabolism, whereas those around the central vein (pericentral, perivenous, or centrolobular areas) are more dependent upon an anaerobic mechanism. Throughout this volume the latter three terms are used synonymously by various authors to indicate the five to eight layers of cells radiating from the central vein. Structural and metabolic heterogeneity of hepatic parenchymal cells has been demonstrated by a variety of approaches, including histochemical, ultra structural, and ultramicrobiochemical studies. This microheterogeneity is linked to the physiological functions of the liver and its response to injurious substances.
