Two animal experiments were conducted to investigate the effect of diets enriched in ALA or LA in German Simmental and Holstein bulls. The influence of LA and ALA enriched diets on the protein expression of lipogenic enzymes in cattle, the tissue specific differences and the relationship between protein expression of lipogenic enzymes, enzyme activities and the products of these enzymes as well as the absorption and deposition of n-3 and n-6 fatty acids in ruminants and beef products was analyzed. Feeding a diet enriched in LA or ALA results in an increased deposition of these fatty acids, induced chain elongation, desaturation and deposition of the long-chain products in tissues of ruminants. Essential fatty acids are preferentially deposited in longissimus muscle (MLD) compared to subcutaneous adipose tissue (SAT) and also preferred in erythrocytes; conclusively they are good long-time marker for the fatty acid intake. Book excerpt: Beef fatty acid (FA) composition has emerged as a trait of economic importance as consumers have become more aware of the relationship between diet and health. As a result, they are paying more attention on the health impact of the food they consume including the type of dietary fat. Clinical studies have shown that the type of dietary fat has a more profound impact on human health than the amount of fat in the diet. In animal studies, phenotypic variations of fatty acid contents in animal tissues are commonly observed. The objectives of this study were: (1) to estimate heritabilities, genetic and phenotypic correlation for FAs in the brisket adipose tissue, and heritabilities for FAs in the subcutaneous adipose tissue (SQ) and longissimus lumborum (LL) muscle in beef cattle, (2): to estimate phenotypic and genetic correlations between FAs in the LL muscle with 6 carcass and 13 meat quality traits of economic importance, (3): to identify single nucleotide polymorphism markers (SNP) associated with FA composition in order to help in designing effective genetic selection tool / genotype based management program for selection of beef with healthier FA profile to meet consumers' demands. Heritability for 15 individual and 10 groups of FAs having a concentration greater than 0.5% was estimated in the brisket adipose of 223 Angus based crossbred steers, and for over 80 individual and groups of FAs in the LL muscle and SQ adipose tissue of 1366 crossbred steers and heifers using a univariate animal model. Across the three tissues, heritability ranged from 0 for 7c-17:1 in the LL and 7c-17:1, 12t-18:1, 6t, 8t-18:2, 9t, 11t-18:2, n-6/n-3 in the SQ to 0.68 ± 0.1 for 9c-16:1 in the LL nuscle tissue. The result suggested the presence of host animal genetic effects on some FAs, including harmful 14:0 (0.61 ±0.13 )and 16:0 (0.54 ±0.1 ), beneficial MUFA (0.44±0.09), and health index (0.54±0.1) in the muscle. The results also showed that non-genetic factors played a major role in determining the concentration of many other FAs in the muscle including 9c, 11t-18:2 conjugated linoleic acid (CLA, 0.16 ±0.07) and vaccenic acid (18:1, trans-11, 0.24 ±0.08), which have been shown to benefit humam health. Phenotypic correlation between FAs in the brisket adipose tissue did not indicate significant antagonistic relationships between harmful and healthy FAs. The relationship revealed that reducing the concentration of harmful 16:0 would yield a correlated response of increased concentration of beneficial 9c-18:1, 11t-18:1, 9c,11t-18:2, and 18:3n3. However, in the LL muscle, genetic correlations revealed antagonistic relationships between monounsaturated FA (MUFA) and hot carcass weight (HCW) (-0.4±0.13) and between 18:3n3, 22:3n6, and total n-3 with marbling (MARB) (-0.82±0.11,-0.60±0.17, -0.84±0.11). For FAs and meat quality, unfavourable genetic relationships existed between 11t-18:1 with WBSF_29d, shear force on 26-day aged steaks and flavor (0.49±0.22, -0.37±0.32). There was also a moderate to strong antagonistic genetic relationship between beneficial polyunsaturated FAs, 9c, 11t-18:2, 18:3n3, 20:5n3, 22:6n3 and total omega 3 with meat quality traits particularly flavor, tenderness and juiciness. A two step bayesian analysis approach was used to evaluate the association of each of 15 individual and 10 grouped FAs for associations with 947 polymorphic SNP markers in 556 growth- and fat metabolism- related genes. In this context, the various health benefits of consuming omega-3 (n-3) polyunsaturated fatty acids (PUFAs) have been widely reported. Supplementation of beef cattle diets with flaxseed, a rich source of alpha-linolenic acid (ALA), has been shown to increase n-3 FAs content of muscle and fat tissues. Towards the goal of enhancing beef FA composition, 64 crossbred cull cows (~30 months of age) with similar breed composition were randomized by weight/body condition, and fed one of four 50:50 forage:concentrate diets on a DM basis (16 cows/treatment), containing ground barley grain with either hay or silage, supplemented with 0 or 15% ground flaxseed (DM basis). Cows were slaughtered after spending 140 days on the treatment diets. For the initial transcriptional analysis, five cows from each of the four diets (20 in total), were selected using an index equation (Index=0.40(18:3n-3) + 0.60(Long Chain:3n)) which ranked cows based on the amount of alpha-linolenic acid (18:3n-3) and long chain omega-3 (3n) FAs in kidney fat collected at slaughter. Cattle scoring the highest indexes were selected from diets which included flax and those scoring the lowest indexes were selected from diets which did not contain flax. RNA was isolated from Longissimus thoracis (LT) muscle, subcutaneous fat (SC), and kidney fat (KF) tissues of each of the 20 cows, and hybridized in duplicate to BOMC 24K 70-mer microarrays. Book excerpt: In this study subcutaneous, inter- and intramuscular fats from 42 Hereford steers were analyzed for their fatty acid composition using gas-liquid chromatography. The same tissues were analyzed from one pair each of identical twins of Angus and Shorthorn breeding. Muscle and liver lipids from these latter four animals were fractionated on silicic acid columns into glycerides and phospholipids prior to the fatty acid analysis. Feedlot performance and carcass characteristics data were also available on these animals. The data were analyzed statistically using analysis of variance techniques and simple correlations were calculated between all the variables studied with the exception of linolenic acid. Of the ten fatty acids quantitated, only the 17-carbon fatty acid and an 18-carbon branched chain acid were found to differ significantly between rations. Since both these acids are believed to be synthesized by certain rumen bacteria, these differences may reflect conditions in the rumen. No direct evidence is available. However, it is probable that this involves the availability of suitable precursors or rumen conditions favorable to these species of organisms. A highly significant relationship (r = .47) was found between average daily gain and linoleic acid. This relationship is probably also influenced by conditions in the rumen. A more rapid rate of passage or other factors which result in less extensive hydrogenation of polyunsaturated acids might also be conducive to more rapid growth. Essential fatty acids, such as linoleic, might be expected to reduce growth rates when they are deficient, however, no evidence of such a deficiency existed. Backfat thickness gave highly significant negative correlations with both myristic (r = .44) and palmitic acid (4 = .46) and was positively related to the 17- and 18-carbon fatty acids with the exception of stearic. Rib-eye area/CWT and estimated cutability, conversely, were positively and highly significantly correlated with the 14- and 16-carbon fatty acids and showed a negative relationship with the longer chained fatty acids. These relationships may indicate metabolic differences between the animals which deposit more lean tissue with less fat and those depositing greater amounts of fat. If, as these data indicate, meatier animals accumulate more of the 14- and 16-carbon fatty acids in the subcutaneous fat, a useful selection tool will be provided to the animal breeder. These 14- and 16-carbon fatty acids, in general showed a positive relationship between one another. When correlated with the 17- and 18-carbon fatty acids a negative relationship was observed. These negative relationships were particularly pronounced in the case of oleic and linoleic acids. It was suggested that the substitution of shorter chain saturated fatty acids for the longer chain unsaturates is the result of an attempt by the animal to maintain physical homeostasis of its depot fats. This assumption is based on the observation that the removal of carbon atoms has the same effect on some of the physical properties as the addition of a double bond. The data obtained in this study indicated that myristic and palmitic acid were readily substituted for oleic acid. The separation of liver and muscle lipid into glyceride and phospholipid resulted in the detection of additional components. Both muscle and liver phospholipids contained large amounts of arachidonic and eicosatrienoic acids. These acids were detected only in liver glycerides. An unknown fraction was observed in both liver glycerides and phospholipids and in muscle phospholipid. This fraction may represent a fatty aldehyde since they have been previously reported in the liver as free aldehydes and as constituents of phospholipid. The only fraction showing similar trends within twin pairs was the muscle glycerides. The muscle phospholipids and all the liver fractions showed considerable variability both within and between twin pairs.
