Book excerpt: Although many pursue understanding of the relationship between protein structure and function for the thrill of pure science, the pay-off in a much broader sense is the ability to manipulate the Earth's chemistry and biology to improve the quality of life for mankind. Immediately goals of this area of research include identification of the life-supporting functions of proteins, and the fundamental forces that facilitate these functions. Upon reaching these goals, we shall have the understanding to direct and the tools required to implement changes that will dramatically improve the quality of life. For example, under standing the chemical mechanism of diseases will facilitate development of new therapeutic drugs. Likewise, understanding of chemical mechanisms of plant growth will be used with biotechnology to improve food production under adverse climatic conditions. The challenge to understand details of protein structure/function relationships is enormous and requires an international effort for success. To direct the chemistry and biology of our environment in a positive sense will require efforts from bright, imaginative scientists located throughout the world. Although the emergence of FAX, e-mail, and the World Wide Web has revolutionized international communication, there remains a need for scientists located in distant parts of the world to occasionally meet face to face.

Book excerpt: Serum albumin is a large protein present in cow serum protein (BSA, Bovine Serum Albumin) rich in essential amino acids and compounds with disulfide bridges and thiol groups. This protein passes into breast milk from the bloodstream by diffusion, has antioxidant capacity and protects fats from oxidation. Albumin has the ability to inhibit tumor factors and bind to fat, mobilize them in the body to perform other functions such as obtaining energy or forming membranes. It is also able to block the conversion of angiotensin I into angiotensin II (vasoconstrictor) which gives it antihypertensive action. In addition, it has the ability to reduce cholesterol absorption, complex heavy metals and is an opioid agonist (acts on opioid receptors and promotes analgesia). BSA is the most studied and produced protein worldwide in the last 80 years. This protein brings large dividends to the world economy with a very prominent and favorable annual growth at a cost of 100 USD per gram. Proteins are different from "traditional" low-molecular-weight drugs. As a group, they exhibit a number of biopharmaceutical and formulation problems. These problems have drawn considerable interest from both industrial and aca demic environments, forcing pharmaceutical scientists to explore a domain previ ously examined only by peptide and protein chemists. Biopharmaceutical aspects of proteins, e.g., low oral bioavailability, have been extensively investigated. Although all possible conventional routes of ad ministration have been examined for proteins, no real, generally applicable alter native to parenteral administration in order to achieve systemic effects has yet been discovered. Several of these biopharmaceutical options have been discussed in Volume 4 of this series, Biological Barriers to Protein Delivery. Proteins are composed of many amino acids, several of which are notorious for their chemical instability. Rational design of formulations that optimize the native structure and/or bioactivity of a protein is therefore of great importance when long shelf life is required, as it is for pharmaceutical products. This issue has also been examined in two prior volumes of this series: Volume 2: Stability of Protein Pharmaceuticals (Part A) and Volume 5: Stability and Characterization of Protein and Peptide Drugs.
