Book excerpt: Juvenile chinook salmon (Oncorhynchus tshawytscha) were experimentally infected with Renibacterium salmoninarum (Rs), the causative agent of bacterial kidney disease (BKD), to investigate the effects of BKD on three aspects of juvenile salmonid performance: (1) predator avoidance ability; (2) smoltification; and (3) physiological responses to stress. For these experiments, fish with different Rs-infection profiles (created by using an immersion challenge method) were sampled to assess physiological change and subjected to various performance tests during disease progression. When equal numbers of Rs-challenged and unchallenged fish were subjected to predation by northern pikeminnow (Ptychocheilus oregonensis) or smallmouth bass (Micropterus dolomieui), Rs-challenged fish were eaten in significantly greater numbers than controls by nearly two to one. A progressively worsening infection with Rs did not alter the normal changes in gill ATPase and condition factor associated with smoltification in juvenile chinook salmon. A dramatic proliferation of BKD was associated with maximal responses of indicators of smoltification, suggesting that the process of smoltification itself can trigger outbreaks of disease. When Rs-infected fish were subjected to three 60-s bouts of severe handling that were separated by 48-72 h, this experience did not lead to higher infection levels or increased mortality when compared to diseased fish that did not receive the stressors. Furthermore, the kinetics of plasma cortisol, glucose, and lactate over 24-h following each stressor were similar between fish with moderate to high BKD and those that had low or no detectable infection. Fish with moderate to high Rs infections had higher titers of cortisol and lactate prior to each application of the stressor and were also unable to consistently elicit a significant hyperglycemia in response to the stressors when compared to fish with low infection levels. During all experiments, fish consistently developed decreased hematocrits and blood glucose levels and increased levels of cortisol and lactate as the disease worsened, indicating that BKD is stressful, particularly during the later stages. Collectively, these results illustrate the impact of BKD on juvenile salmonids and have also ascribed some ecological significance to this disease beyond that of direct pathogen-related mortality.

The disease has a major impact on spring chinook salmon populations in the Columbia River system. There is strong evidence that RS can be transmitted from parent to progeny, and segregation of progeny based on levels of antigen detected in adult fish may obviate this mode of transmission. Results are presented from the third year of a four year study to investigate segregation of broodstock as a tool for controlling BKD. Segregation of adult fish infected with RS has been achieved using enzyme-linked immunosorbent assays (ELlSAs) optimized in the first and second year of this project. Gametes from both 1990 and 1991 broodstock, either injected with erythromycin or receiving no antibiotic injection were successfully segregated into groups having either high or low levels of the RS soluble antigen. Offspring have been monitored every three months from the 1990 broodstock and are being monitored from the 1991 broodstock. Antigen levels in the offspring from the 1990 segregation experiment at Marion Forks Hatchery were low and clinical BKD was not observed in any of the juvenile fish. At Carson National Fish Hatchery, antigen levels were also low in fish which were sampled December 1990 through July 1991. Total mortality was low throughout these sampling periods. An increase in mortality was observed in November-December 1991, and preliminary evidence suggests that motality may have been due BKD. The epizootic appears to have equally effected both offspring from high and low RS antigen level parents. Antigen levels in moribund fish are being examined to confirm the prevalence of RS infection.

Book excerpt: The depressed status of Snake River stocks of chinook and steelhead and the recent listings of many salmon stocks in the Columbia Basin have led to several analytical evaluations and management advice aimed at recovery of these stocks. These different analytical reviews address the effectiveness of different hydrosystem options as well as the potential for recovery through improvements that increase survival at other life stages (e.g., habitat, harvest). Hydrosystem options evaluated included status quo, maximizing transportation, and the option of breaching the lower four dams on the Snake River (also called drawdown and natural river options), the main topic of the Lower Snake River Juvenile Salmon Migration Feasibility Report / Environmental Impact Statement (USACE). The first review was completed by PATH (Plan for Testing and Analyzing Hypotheses), an open forum composed of modelers, fishery biologists and statisticians from all three states (Oregon, Washington, and Idaho), the federal government (Army Corps of Engineers (USACE), US Fish and Wildlife Service (USFWS), National Marine Fishery Service (NMFS), Bonneville Power Administration (BPA), the treaty tribes of the Columbia Basin (represented by the Columbia River Inter-tribal Fish Commission -CRITFC), and the Northwest Power Planning Council (NPPC). The PATH approach was based on a decision analysis that showed which management actions are the most robust to remaining uncertainties (i.e. the least risky) and allows a decision to be made with full consideration of uncertainty and risk. PATH analyses were followed by the NMFS effort called CRI- the Cumulative Risk Initiative. CRI analyses explore the demographic effects of hypothetical reductions in mortality at different life stages based on current conditions. PATH and CRI analyses were followed by an analytical comparison of their approaches and results completed by a sub group of PATH composed of scientists from the states of Oregon, Idaho, Washington, CRITFC, and the USFWS. In addition, specific analyses have considered the potential for improvement at certain life stages (e.g., freshwater spawning and rearing; Petrosky et al., in press) and key uncertainties that affect the likely effectiveness of dam breach (e.g., delayed hydrosystem mortality; Budy et al., in review). This annex synthesizes analyses and results PATH, NMFS CRI, and comparative and follow-up analyses which have been completed since and are summarized here and described in greater detail elsewhere. Although the results vary somewhat among approaches, all available science appears to suggest that dam breach has the greatest biological potential for recovering Snake River salmon and steelhead.

There is strong evidence that R. safmoninarum can be transmitted from parent to progeny, and therefore culling of gametes from infected parents should obviate this mode of transmission. This report presents the results from the first year of our four year study to investigate segregation of broodstock as a tool for controlling BKD. The segregations will use Enzyme-Linked Immunosorbent Assays (ELISAs) as detection systems to identify, in tissues of infected fish, proteins produced by R. salmoninarum. A first step in the development of the described detection systems was the optimization of the production of important antigenic proteins from R. salmoninarum. Different culture media were qualitatively and quantitatively evaluated for their ability to support production of cellular and soluble proteins. The major factor affecting antigen quality was the presence and absence of calf serum. Media components and R. salmoninarum growth products could not be separated during harvest of proteins from the cultures containing serum. This caused problems with the quantitation of actual bacterial proteins in the preparation. Thus media without serum is currently employed. Two independent ELISA techniques for the identification of infected parents were examined. One technique is based on polyclonal antisera produced in rabbits and the second is based on mouse monoclonal antibodies (Mabs). To develop the latter system, several Mabs against a major R. salmoninarum antigenic protein were produced. These Mabs were used for the detection of R. salmoninarum antigens in infected fish and also to characterize proteins produced by the bacterium. Both ELISAs were deemed suitable for the segregation of parents into the high and low BKD groups required for this study. An alternate system for the detection of R. salmoninarum proteins, the Western blot, was also investigated, This technique was 100 to 1000 fold less sensitive than either ELISA system and therefore will be useful only for confirmation of highly positive tissues. Future work will attempt to increase the sensitivity of the Western blotting system. Finally, two hatcheries were identified for use in the described segregation experiments. The Carson National Fish Hatchery (Skamania County, WA) and the Marion Forks Fish Hatchery (Linn County, OR) will be used for the experiments.

Screening of eggs from infected adults has not revealed any detectable antigen present in the egg tissue. Development of a rapid, field ELISA has been accomplished this year. The field ELISA utilizes monoclonal antibodies currently employed in the monoclonal antibody-based ELISA. The sensitivity of the field ELISA approaches that of the monoclonal ELISA, and has been tested on 150 adult chinook salmon. A high correlation exists between samples assayed by the monoclonal ELISA, field ELISA, and direct FAT. An alternative system for detecting RS soluble antigen, the Western blot, has also been improved. Using a chemiluminescent substrate, the sensitivity of detection has been increase 50--100 fold. 16 refs., 15 figs., 2 tabs.
